Downscaling is an important problem because consistent large-area assessments of forest habitat structure, while feasible, are only feasible when using relatively coarse data and indicators. Techniques are needed to enable more detailed and local interpretations of the national statistics. Using the results of national assessments from land-cover maps, this paper demonstrates downscaling in the spatial domain, and in the domain of the habitat model. A moving window device was used to measure structure (habitat amount and connectivity), and those indicators were then analyzed and combined with other information in various ways to illustrate downscaling.  相似文献

Anions were found to have a number of different effects on the reconstituted ADP/ATP carrier from mitochondria. (1) Binding of adenine nucleotides to the active site of the translocator is competitively inhibited by various anions. These anions can be arranged in a sequence of increasing competitive effect due to their order in a lyotropic series, and also due to increasing charge. (2) Apart from this competition effect, the presence of a sufficiently high concentration of anions turned out to be absolutely essential for functional ADP/ATP exchange in the reconstituted system. The activating anions too can be arranged in sequence, similar to that of the competition effect. The adenine nucleotide transport shows sigmoidal dependence on the stimulating anions with a Hill coefficient of n = 2. Addition of anions does not change the basic amount of functionally active translocator molecules. (3) The different effects of anions, i.e., inhibition and activation, were shown to take place at different sites and to be due to different mechanisms. Anions compete with substrates both at the outer (cytosolic) and at the inner (matrix) active site, whereas anion activation is observed solely by interaction with the cytosolic side of the translocator protein. (4) Activation of the reconstituted ADP/ATP exchange by anions could be discriminated from an activating influence of anionic phospholipids in the surroundings of the carrier protein.  相似文献

We used a strategy based on long PCR (polymerase chain reaction) for detection and characterization of mitochondrial DNA (mtDNA) rearrangements in two patients with clinical signs suggesting Pearson syndrome and Kearns-Sayre syndrome (KSS), respectively, and one patient with myopathic symptoms of unidentified origin. Mitochondrial DNA rearrangements were detected by amplification of the complete mitochondrial genome (16.6 kb) using long PCR with primers located in essential regions of the mitochondrial genome and quantified by three-primer PCR. Long PCR with deletion-specific primers was used for identification and quantitative estimation of the different forms of rearranged molecules, such as deletions and duplications. We detected significant amounts of a common 7.4-kb deletion flanked by a 12-bp direct repeat in all tissues tested from the patient with Pearson syndrome. In skeletal muscle from the patient with clinical signs of KSS we found significant amounts of a novel 3.7-kb rearrangement flanked by a 4-bp inverted repeat that was present in the form of deletions as well as duplications. In the patient suffering from myopathic symptoms of unidentified origin we did not detect rearranged mtDNA in blood but found low levels of two rearranged mtDNA populations in skeletal muscle, a previously described 7-kb deletion flanked by a 7-bp direct repeat and a novel 6.6-kb deletion with no repeat. These two populations, however, were unlikely to be the cause of the myopathic symptoms as they were present at low levels (10–40 ppm). Using a strategy based on screening with long PCR we were able to detect and characterize high as well as low levels of mtDNA rearrangements in three patients. We examine three critical aspects of Popper’s formulation of the ‘Logic of Scientific Discovery’—evidence, content and degree of corroboration—and place these concepts in the context of the Tree of Life (ToL) problem with particular reference to molecular systematics. Content, in the sense discussed by Popper, refers to the breadth and scope of existence that a hypothesis purports to explain. Content, in conjunction with the amount of available and relevant evidence, determines the testability, or potential degree of corroboration, of a statement; content distinguishes scientific hypotheses from metaphysical assertions. Degree of corroboration refers to the relative and tentative confidence assigned to one hypothesis over another, based upon the performance of each under critical tests. Here we suggest that systematists attempt to maximize content and evidence to increase the potential degree of corroboration in all phylogenetic endeavors. Discussion of this “total evidence” approach leads to several interesting conclusions about generating ToL hypotheses.  相似文献
