Findings from animal experiments are sometimes contradictory to the idea that the tendon structure is a simple elastic spring in series with muscle fibers, and suggest influence of muscle contraction on the tendon mechanical properties. The purpose of the present study was to investigate the influence of muscle contraction levels on the force-length relationship of the human Achilles tendon during lengthening of the triceps surae muscle-tendon unit. For seven subjects, ankle dorsiflexion was performed without (passive condition) and with contraction of plantar flexor muscles (eccentric conditions, at 3 contraction levels) on an isokinetic dynamometer. Deformation of the Achilles tendon during each trial was measured using ultrasonography. The Achilles tendon force corresponding to the tendon elongation of 10mm in the passive condition was significantly smaller than those in the eccentric conditions (p<0.05 or p<0.01). Within the eccentric conditions, the Achilles tendon force corresponding to the tendon elongation of 10mm was significantly greater in the maximal contraction level than those in submaximal eccentric conditions (p<0.05 or p<0.01). In addition, the tendon stiffness was greater in higher contraction levels (p<0.05 or p<0.01). Present results suggest that the human tendon structure is not a simple elastic spring in series with muscle fibers.  相似文献

We investigated the localization, structure and function of the biliproteins of the oxygenic photosynthetic prokaryote Acaryochloris marina, the sole organism known to date that contains chlorophyll d as the predominant photosynthetic pigment. The biliproteins were isolated by means of sucrose gradient centrifugation, ion exchange and gel filtration chromatography. Up to six biliprotein subunits in a molecular mass range of 15.5-18.4 kDa were found that cross-reacted with antibodies raised against phycocyanin or allophycocyanin from a red alga. N-Terminal sequences of the alpha- and beta-subunits of phycocyanin showed high homogeneity to those of cyanobacteria and red algae, but not to those of cryptomonads. As shown by electron microscopy, the native biliprotein aggregates are organized as rod-shaped structures and located on the cytoplasmic side of the thylakoid membranes predominantly in unstacked thylakoid regions. Biochemical and spectroscopic analysis revealed that they consist of four hexameric units, some of which are composed of phycocyanin alone, others of phycocyanin together with allophycocyanin. Spectroscopic analysis of isolated photosynthetic reaction center complexes demonstrated that the biliproteins are physically attached to the photosystem II complexes, transferring light energy to the photosystem II reaction center chlorophyll d with high efficiency.  相似文献

Chinese hamster ovary (CHO) cells were cultivated in a compact loop bioreactor using MEM-alpha medium supplemented with 10% fetal calf serum. Effects of physical and chemical environments, i.e., pH in the medium, stirring speed of impellers, temperature and partial pressure of oxygen (pO2) upon growth of suspended cells in the bioreactor were determined in batch cultures. Growth behavior was characterized by specific rates of growth (mu), glucose consumption (qG) and lactate production (qL), and the yield coefficients (cell yield from glucose, YX/G, and lactate yield from glucose, YL/G). An effect of medium osmolality was also evaluated with T-flask monolayer cultivation. The best growth was observed at pH 7.6, 37 degrees C, 400 rpm, 50-100% saturation with oxygen and 320 mOsmol kg-1. Corresponding to the previous work with a human melanoma cell line, the sophisticated cultivation and process control systems have been improved for CHO cells.  相似文献

A new pest of Welsh onion and carrot, the dark-winged fungus gnat, Bradysia sp., was found in the northern region of Saitama Prefecture, Japan. DNA sequences of mitochondrial cytochrome c oxidase subunit I revealed that Bradysia sp. is identical to the Chinese chive gnat, Bradysia odoriphaga Yang and Zhang. This suggests that Bradysia sp. may be an invasive species that originates from continental East Asia. Direct sequencing analysis and PCR using a species-specific primer were found to be useful tools for discriminating Bradysia sp. from other native Japanese fungus gnats, such as Bradysia impatiens (Johannsen), Pnyxia scabiei (Hopkins), and Lycoriella ingenua (Dufour).  相似文献

The concentrations of four vitamins, ascorbic acid, nicotinamide, choline and thiamine were evaluated in the culture supernatant of Chinese hamster ovary (CHO) cells. The media used were -modified Eagle's minimum essential medium (MEM-) supplemented with 10% fetal calf serum, and a 1:1 mixture of Ham's F12 and Dulbecco's modified Eagle's medium (DME/F12), containing neither serum nor protein. The reference experiment without cells revealed instability of ascorbic acid and thiamine. Moreover, a significant amount of each vitamin decreased in the culture supernatant. The possibility of growth limitation by vitamin depletion is strongly suggested.  相似文献

This study aimed to investigate whether fatigue-induced changes in synergistic muscle forces match their tendon elongation. The medial gastrocnemius muscle (MG) was fatigued by repeated electrical stimulation (1 min×5 times: interval 30 s, intensity: 20–30% of maximal voluntary plantar flexion torque) applied at the muscle belly under a partial occlusion of blood vessels. Before and after the MG fatigue task, ramp isometric contractions were performed voluntarily, during which tendon elongations were determined by ultrasonography, along with recordings of the surface EMG activities of MG, the soleus (SOL) and the lateral gastrocnemius (LG) muscles. The tendon elongation of MG and SOL in post-fatigue ramp was similar, although evoked MG forces dropped nearly to zero. In addition, for a given torque output, the tendon elongation of SOL significantly decreased while that of LG did not, although the activation levels of both muscles had increased. Results suggest that the fatigue-induced changes in force of the triceps surae muscles do not match their tendon elongation. These results imply that the tendons of the triceps surae muscles are mechanically coupled even after selective fatigue of a single muscle.  相似文献

The purpose of this study was to test the hypothesis that the musculotendon moment arm length is affected by the muscle anatomical cross-sectional area. The moment arm length of the triceps brachii (TB) muscle at 30°, 50°, 70°, 90°, 110° elbow flexion positions was measured in sagittal magnetic resonance images (MRI) of 18 subjects as the perpendicular distance between the center of the pulley of the humerus to the line through the center of the TB tendon. The moment arm increased as the elbow flexion angle decreased, from 1.74±0.13 cm at 110° to 2.39±0.14 cm at 30°. The maximal anatomical cross-sectional area of the TB muscle was significantly correlated with the moment arms at all joint positions (r=0.545–0.803, p<0.05). Furthermore, the circumference of the upper arm was also significantly correlated with the moment arms at all joint positions, except for 70° (r=0.504–0.702, p<0.05). These results indicate that the moment arm length of the TB muscle is affected by the muscle anatomical cross-sectional area.  相似文献
