通过田间小区试验,设N0(0 kg·hm-2)、N1(45 kg·hm-2)、N2(90 kg·hm-2)、N3(135 kg·hm-2)4个施氮水平,研究不同施氮水平下小麦与蚕豆间作对蚕豆赤斑病发生和冠层微气候的影响,探讨间作系统氮肥调控下冠层微气候变化及其与蚕豆赤斑病发生的关系.结果表明: 施氮提高了蚕豆单、间作种植模式下蚕豆赤斑病发病盛期的病情指数,增幅27.2%~58.0%,增加了病情进展曲线下面积(AUDPC),增幅15.0%~101.8%,N3水平下赤斑病病情指数和AUDPC最高.施氮使蚕豆冠层温度降低0.2~1.1 ℃,冠层透光率降低1.7%~29.7%,冠层相对湿度增加0.5%~28.7%.与单作相比,间作蚕豆赤斑病病情指数显著降低36.3%~48.1%,AUDPC显著降低44.0%~53.6%,冠层温度和透光率分别提高2.1%~8.7%和12.0%~53.8%,相对湿度降低11.6%~31.6%.相关分析表明,冠层温度和透光率与赤斑病病情指数呈显著负相关,而湿度与病情指数呈显著正相关.表明高氮恶化了冠层微气候环境,加重了蚕豆赤斑病的发生和危害,而间作对蚕豆冠层微气候的改善是控制蚕豆赤斑病发展的重要原因.  相似文献

Citrus essential oils (CEOs) are important flavors in the food and confectionary industries. A lipase process was proposed for enhancing the flavor profiles and increasing the proportions of esters in CEOs. The effects of the enzymatic process were explored by detecting the constituents of the CEOs of American sweet orange oil (ASO) and Brazil mandarin oil (BMO) through GC/MS and sensory evaluation by a trained panel, and positive effects were confirmed by both methods. A further eleven kinds of CEOs were treated via the lipase process and increments of 10 – 1170% were achieved in the proportions of esters, which were mostly ethyl esters. Enhancement in fruity odor, especially the top note, was demonstrated by all CEOs after enzymatic processing. All CEOs were tested for antimicrobial activities, and only ASO displayed fairly ideal antimicrobial activities. Meanwhile, modified ASO showed a certain increase in antimicrobial activities. This methodology might be considered a sustainable route for acquiring ‘natural’ essential oils with enhanced flavor profiles and simultaneously enhancing the comprehensive utilization of citrus fruits.  相似文献

在完成了对各种微生物基因组的测序以后,功能基因学的研究变得尤为重要。研究基因功能最直接的方法便是将待研究的基因失活。最初构建基因突变体是采用大肠杆菌的RecA系统,但是RecA重组系统操作复杂,重组效率低。最近建立了Red重组系统,该系统由3个蛋白组成:α蛋白(即λ核酸外切酶),β蛋白,Gam蛋白。应用Red系统进行基因敲除,可以直接利用线性打靶DNA,两侧同源臂长度在35~60 bp即可发生同源重组,且重组效率高。 Abstract:Since many DNA-sequencing projects of varied microorganisms have been completed,studies on their functional genomics become more important.Inactivation of an interesting gene is a direct method to characterize its function.Though the Esherichia coli RecA recombination system can be used to produce gene mutants,it needs a complex manipulation process.Furthermore,its efficiency is very low.Recently a Red recombination system was developed.This recombination system consists of three proteins:α protein(λ exonuclease),β protein and Gam protein.In this system,the linear targeting DNA which contains a selectable marker flanked with a homologous region as short as only 35~60 bp can be directly targeted for gene knock-out with a higher efficiency.  相似文献

Adipose tissue hypoxia is an early phenotype in obesity, associated with macrophage infiltration and local inflammation. Here we test the hypothesis that adipocytes in culture respond to a hypoxic environment with the release of pro-inflammatory factors that stimulate macrophage migration and cause muscle insulin resistance. 3T3-L1 adipocytes cultured in a 1% O2 atmosphere responded with a classic hypoxia response by elevating protein expression of HIF-1α. This was associated with elevated mRNA expression and peptide release of cytokines TNFα, IL-6 and the chemokine monocyte chemoattractant protein-1 (MCP-1). The mRNA and protein expression of the anti-inflammatory adipokine adiponectin was reduced. Conditioned medium from hypoxia-treated adipocytes (CM-H), inhibited insulin-stimulated and raised basal cell surface levels of GLUT4myc stably expressed in C2C12 myotubes. Insulin stimulation of Akt and AS160 phosphorylation, key regulators of GLUT4myc exocytosis, was markedly impaired. CM-H also caused activation of JNK and S6K, and elevated serine phosphorylation of IRS1 in the C2C12 myotubes. These effects were implicated in reducing propagation of insulin signaling to Akt and AS160. Heat inactivation of CM-H reversed its dual effects on GLUT4myc traffic in muscle cells. Interestingly, antibody-mediated neutralization of IL-6 in CM-H lowered its effect on both the basal and insulin-stimulated cell surface GLUT4myc compared to unmodified CM-H. IL-6 may have regulated GLUT4myc traffic through its action on AMPK. Additionally, antibody-mediated neutralization of MCP-1 partly reversed the inhibition of insulin-stimulated GLUT4myc exocytosis caused by unmodified CM-H. In Transwell co-culture, hypoxia-challenged adipocytes attracted RAW 264.7 macrophages, consistent with elevated release of MCP-1 from adipocytes during hypoxia. Neutralization of MCP-1 in adipocyte CM-H prevented macrophage migration towards it and partly reversed the effect of CM-H on insulin response in muscle cells. We conclude that adipose tissue hypoxia may be an important trigger of its inflammatory response observed in obesity, and the elevated chemokine MCP-1 may contribute to increased macrophage migration towards adipose tissue and subsequent decreased insulin responsiveness of glucose uptake in muscle.  相似文献

PIWIL2, called HILI in humans, is a member of the PIWI subfamily. This subfamily has highly conserved PAZ and Piwi domains and is implicated in several critical functions, including embryonic development, stem-cell self-renewal, RNA silencing, and translational control. However, the underlying molecular mechanism remains largely unknown. Transforming growth factor-β (TGF-β) is a secreted multifunctional protein that controls several developmental processes and the pathogenesis of many diseases. TGF-β signaling is activated by phosphorylation of transmembrane serine/threonine kinase receptors, TGF-β type II (TβRII), and type I (TβRI), which are stabilized by Hsp90 via specific interactions with this molecular chaperone. Here, we present evidence that HILI suppresses TGF-β signaling by physically associating with Hsp90 in human embryonic kidney cells (HEK-293). Our research shows that HILI mediates the loss of TGF-β-induced Smad2/3 phosphorylation. We also demonstrate that HILI interacts with Hsp90 to prevent formation of Hsp90-TβR heteromeric complexes, and improves ubiquitination and degradation of TβRs dependent on the ubiquitin E3 ligase Smurf2. This work reveals a critical negative regulation level of TGF-β signaling mediated by HILI (human PIWIL2) by its ability to interact with Hsp90 and promote TβR degradation.  相似文献

本实验用胆固醇粉喂家兔,造成实验性高脂血症,活体观察球结膜微循环的形态、流态及血管周围状态等15项指标,并用球结膜微循环综合定量评价方法计算了综合积分值。处死后取球结膜进行组织学检查。结果表明:高脂血症家兔球结膜微循环有明显改变,主要为微血管形态异常、血流减慢、红细胞聚集、白微栓、静脉壁上有白色斑块等改变。球结膜微循环综合积分值明显增加,高于实验前。球结膜组织学检查发现上皮层下有泡沫细胞、血管壁增厚、有空泡、静脉内有血栓。虹膜睫状体内除有多数泡沫细胞外,还有胆固醇的菱形结晶。上述改变表明,高脂血症兔球结膜微循环有明显改变。血液有高凝、高聚倾向。黑茶中的茶色素有抗凝、抑制血小板聚集、促进纤溶等作用,口服黑茶可改善高脂血症兔球结膜微循环障碍。  相似文献

Microbial fuel cells (MFCs) represent a new biological method for generating electricity directly from biodegradable compounds. Efficiency of MFCs using manure as substrate is generally low. This study proposed a new design by incorporating biocathodes into a three-chamber MFC, which yielded maximum power densities much higher than those reported in literature. The new design placed cylindrical anode chamber for easy stirring and two symmetrical cathodic chambers with reduced anode-cathode distance. The biocathodes were applied to reduce charge transfer resistance. Additionally, biocathode microbial community was cultured to enrich favorable microorganisms. With external loading of 100 Ω, the power densities for new biocathode MFC using 2, 4, 6, 8 and 10% total solids diary manure reached 7.85±1.0 W m(-3), 7.84±1.20 W m(-3), 8.15±0.20 W m(-3), 7.60±0.97 W m(-3) and 5.63±0.97 W m(-3), respectively. The pH drop as a result of manure hydrolysis limited the power output. To provide detailed information of the microbial community in the biocathode MFC, the 454-pyrosequencing technique was adopted. The Firmicutes, γ-, β-, α- and δ-Proteobacteria, Bacteroidetes and Actinobacteria were the major groups on the anode, while γ-, β-, and α-Proteobacteria, Bacteroidetes and Actinobacteria were the predominant groups on the cathode.  相似文献

Rab7是一个GTP结合蛋白,隶属于Rab家族,该家族在调控膜泡的运输、结合以及细胞内膜结构的组织中行使重要作用.本实验通过RT-PCR技术从盐敏感水稻品种'日本晴'('Nipponbare')获得了OsRab7基因全长序列,成功构建了原核表达载体pET-32a-OsRab7.在IPTG诱导下,该蛋白高效表达,并明显缓解了高盐环境(4.5%~8.5% NaCl)对大肠杆菌的生长胁迫.为进一步研究该基因的功能,将OsRab7在大肠杆菌中的表达蛋白纯化, 并利用p1301B(pCAMBIA1301改造载体)构建了植物表达载体,成功转化来源于水稻株系'中花11'(盐敏感)的愈伤组织,为探讨其在水稻中的表达模式和功能分析及应用于水稻耐盐品种的改良奠定了一定的基础.  相似文献
