The action of different prefixing agents including water and a dilute CaCl2 solution on plants with high chromosome number showed that all the solutions tried exhibited marked effect on the physical state of the plasma. Also the plasmatic constitution of the pretreated tissue differed from the appearance of one fixed directly in dye-acid mixture for a few seconds without any pre-treatment. For the clarification of the karyotypes, the best results have been obtained with paradichlorobenzene. It was used as a prefixing agent with a short schedule for temporary smears of difficult materials, particularly those with high chromosome number. The method comprised treatment of the root tips in a saturated aqueous solution of the chemical for 3 hours at 12-16°C., followed by 4-5 seconds heating just to boiling in orcein-hydrochloric acid mixture, then smearing in 1% aceto-orcein solution. 相似文献

The well-known role of antibiotics in killing sensitive organisms has been challenged by the effects they exert at subinhibitory concentrations. Unfortunately, there are very few published reports on the advantages these molecules may confer to their producers. This study describes the construction of a genetically verified deletion mutant of Streptomyces flaviscleroticus unable to synthesize chromomycin. This mutant was characterized by a rapid loss of viability in stationary phase that was correlated with high oxidative stress and altered antioxidant defences. Altered levels of key metabolites in the mutant signalled a redistribution of the glycolytic flux toward the PPP to generate NADPH to fight oxidative stress as well as reduction of ATP-phosphofructokinase and Krebs cycle enzymes activities. These changes were correlated with a shift in the preference for carbon utilization from glucose to amino acids. Remarkably, chromomycin at subinhibitory concentration increased longevity of the non-producer and restored most of the phenotypic features’ characteristic of the wild type strain. Altogether these observations suggest that chromomycin may have antioxidant properties that would explain, at least in part, some of the phenotypes of the mutant. Our observations warrant reconsideration of the secondary metabolite definition and raise the possibility of crucial roles for their producers. 相似文献

thioredoxin-fold superfamily. Glutaredoxins are classified into two types: dithiol and monothiol. Monothiol glutaredoxins which
carry the signature “CGFS“ as a redox active motif is known for its role in oxidative stress, inside the cell. In the present analysis,
the 138 amino acid long monothiol glutaredoxin, AgGRX1 from Ashbya gossypii was identified and has been used for the analysis.
The multiple sequence alignment of the AgGRX1 protein sequence revealed the characteristic motif of typical monothiol
glutaredoxin as observed in various other organisms. The proposed structure of the AgGRX1 protein was used to analyze signature
folds related to the thioredoxin superfamily. Further, the study highlighted the structural features pertaining to the complex

α-Amylase from mung beans (Vigna radiata) was immobilized on two different matrices, Amberlite MB 150 and chitosan beads. Maximum immobilization obtained was 72% and 69% in case of Amberlite and chitosan beads, respectively. The pH optima of soluble α-amylase were 5.6, whereas that for immobilized amylase on chitosan and Amberlite was 7.0. Soluble amylase and Amberlite immobilized amylase showed maximum activity at 65 °C, whereas chitosan immobilized amylase showed maximum activity at 75 °C. α-Amylase immobilized on Amberlite showed apparent Km of 2.77 mg/ml, whereas α-amylase immobilized on chitosan showed an apparent Km of 5 mg/ml. The Amberlite-amylase and chitosan-amylase showed a residual activity of 43% and 27%, respectively, after 10 uses. The loss of activity for free amylase after 100 days of storage at 4 °C was 70%, whereas that for Amberlite- and chitosan-amylases, under the same experimental conditions, the losses were 45% and 55%, respectively. The easy availability of mung bean α-amylase, the ease of its immobilization on low-cost matrices and good stability upon immobilization in the present study makes it a suitable product for further use in industrial applications. 相似文献

Naringin has antioxidant properties that could improve redox-sensitive myocardial ischemia reperfusion (IR) injury. This study was designed to investigate whether naringin restores the myocardial damage and dysfunction in vivo after IR and the mechanisms underlying its cardioprotective effects. Naringin (20–80 mg/kg/day, p.o.) or saline were administered to rats for 14 days and the myocardial IR injury was induced on 15th day by occluding the left anterior descending coronary artery for 45 min and subsequent reperfusion for 60 min. Post-IR rats exhibited pronounced cardiac dysfunction as evidenced by significantly decreased mean arterial pressure, heart rate, +LVdP/dt
max (inotropic state), -LVdP/dt
max (lusitropic state) and increased left ventricular end diastolic pressure as compared to sham group, which was improved by naringin. Further, on histopathological and ultrastructural assessments myocardium and myocytes appeared more normal in structure and the infarct size was reduced significantly in naringin 40 and 80 mg/kg/day group. This amelioration of post-IR-associated cardiac injury by naringin was accompanied by increased nitric oxide (NO) bioavailability, decreased NO inactivation to nitrotyrosine, amplified protein expressions of Hsp27, Hsp70, β-catenin and increased p-eNOS/eNOS, p-Akt/Akt, and p-ERK/ERK ratio. In addition, IR-induced TNF-α/IKK-β/NF-κB upregulation and JNK phosphorylation were significantly attenuated by naringin. Moreover, western blotting and immunohistochemistry analysis of apoptotic signaling pathway further established naringin cardioprotective potential as it upregulated Bcl-2 expression and downregulated Bax and Caspase-3 expression with reduced TUNEL positivity. Naringin also normalized the cardiac injury markers (lactate dehydrogenase and creatine kinase-MB), endogenous antioxidant activities (superoxide dismutase, reduced glutathione and glutathione peroxidase) and lipid peroxidation levels. Thus, naringin restored IR injury by preserving myocardial structural integrity and regulating Hsp27, Hsp70, p-eNOS/p-Akt/p-ERK signaling and inflammatory response. 相似文献

Three strains of Streptomyces (CAI-21, CAI-26 and MMA-32) were earlier reported by us as having potential for biocontrol of charcoal rot of sorghum, caused by Macrophomina phaseolina (Tassi) Goid., and plant growth promotion (PGP) of the plant. In the present investigation, the three Streptomyces were characterised for their physiological traits (tolerance of salinity, temperature, pH and resistance to antibiotics) and further evaluated in the field for their PGP of rice, grown by a system of rice intensification methods. All three Streptomyces were able to grow in NaCl concentrations of up to 12% (except MMA-32), at pH values between 5 and 13 and temperatures between 20 and 40°C. They were highly resistant to ampicillin and trimethoprim (>800 ppm), sensitive to chloramphenicol, kanamycin and nalidixic acid (50–100 ppm) and highly sensitive to streptomycin and tetracycline (5–25 ppm). When evaluated for their PGP activity on seedlings of rice, % germination and shoot and root lengths were significantly enhanced over the control. In the field, the Streptomyces strains significantly enhanced the panicle length, filled grain numbers and weight, panicle weight, 1000 seed weight, tiller numbers, total dry matter, root length (39–65%), root volume (13–30%), root dry weight (16–24%), grain yield (9–11%) and stover yield (11–22%) over the control. In the rhizosphere soil (0–15 cm from root) at harvest, the population of actinomycetes was significantly enhanced as was microbial biomass carbon (27–83%) and nitrogen (24–43%), dehydrogenase activity (34–152%), available P (13–34%) and N (30–53%) and % organic carbon (26–28%). This study investigated the distribution of neuropeptide Y (NPY) in the brain of the night-migratory redheaded bunting (Emberiza bruniceps). We first cloned the 275-bp NPY gene in buntings, with ≥95 % homology with known sequences from other birds. The deduced peptide sequence contained all conserved 36 amino acids chain of the mature NPY peptide, but lacked 6 amino acids that form the NPY signal peptide. Using digosigenin-labeled riboprobe prepared from the cloned sequence, the brain cells that synthesize NPY were identified by in-situ hybridization. The NPY peptide containing cell bodies and terminals (fibers) were localized by immunocytochemistry. NPY mRNA and peptide were widespread throughout the bunting brain. This included predominant pallial and sub-pallial areas (cortex piriformis, cortex prepiriformis, hyperpallium apicale, hippocampus, globus pallidus) and thalamic and hypothalamic nuclei (organum vasculosum laminae terminalis, nucleus (n.) dorsolateralis anterior thalami, n. rotundus, n. infundibularis) including the median eminence and hind brain (n. pretectalis, n. opticus basalis, n. reticularis pontis caudalis pars gigantocellularis). The important structures with only NPY-immunoreactive fibers included the olfactory bulb, medial and lateral septal areas, medial preoptic nucleus, medial suprachiasmatic nucleus, paraventricular nucleus, ventromedial hypothalamic nucleus, optic tectum, and ventro-lateral geniculate nucleus. These results demonstrate that NPY is possibly involved in the regulation of several physiological functions (e.g. daily timing feeding, and reproduction) in the migratory bunting. 相似文献
