Mossy fibers are the sole excitatory projection from dentate gyrus granule cells to the hippocampus, where they release glutamate, dynorphin, and zinc. In addition, mossy fiber terminals show intense immunoreactivity for the inhibitory neurotransmitter GABA. Fast inhibitory transmission at mossy fiber synapses, however, has not previously been reported. Here, we show that electrical or chemical stimuli that recruit dentate granule cells elicit monosynaptic GABA(A) receptor-mediated synaptic signals in CA3 pyramidal neurons. These inhibitory signals satisfy the criteria that distinguish mossy fiber-CA3 synapses: high sensitivity to metabotropic glutamate receptor agonists, facilitation during repetitive stimulation, and NMDA receptor-independent long-term potentiation. GABAergic transmission from the dentate gyrus to CA3 has major implications not only for information flow into the hippocampus but also for developmental and pathological processes involving the hippocampus.  相似文献

Voltage-gated sodium channels underlie the upstroke of action potentials and are fundamental to neuronal excitability. Small changes in the behavior of these channels are sufficient to change neuronal firing and trigger seizures. These channels are subject to highly conserved alternative splicing, affecting the short linker between the third transmembrane segment (S3) and the voltage sensor (S4) in their first domain. The biophysical consequences of this alternative splicing are incompletely understood. Here we focus on type 1 sodium channels (Nav1.1) that are implicated in human epilepsy. We show that the functional consequences of alternative splicing are highly sensitive to recording conditions, including the identity of the major intracellular anion and the recording temperature. In particular, the inactivation kinetics of channels containing the alternate exon 5N are more sensitive to intracellular fluoride ions and to changing temperature than channels containing exon 5A. Moreover, Nav1.1 channels containing exon 5N recover from inactivation more rapidly at physiological temperatures. Three amino acids differ between exons 5A and 5N. However, the changes in sensitivity and stability of inactivation were reproduced by a single conserved change from aspartate to asparagine in channels containing exon 5A, which was sufficient to make them behave like channels containing the complete exon 5N sequence. These data suggest that splicing at this site can modify the inactivation of sodium channels and reveal a possible interaction between splicing and anti-epileptic drugs that stabilize sodium channel inactivation.  相似文献

p27Kip1 controls cell proliferation by binding to and regulating the activity of cyclin-dependent kinases (Cdks). Here we show that Cdk inhibition and p27 stability are regulated through direct phosphorylation by tyrosine kinases. A conserved tyrosine residue (Y88) in the Cdk-binding domain of p27 can be phosphorylated by the Src-family kinase Lyn and the oncogene product BCR-ABL. Y88 phosphorylation does not prevent p27 binding to cyclin A/Cdk2. Instead, it causes phosphorylated Y88 and the entire inhibitory 3(10)-helix of p27 to be ejected from the Cdk2 active site, thus restoring partial Cdk activity. Importantly, this allows Y88-phosphorylated p27 to be efficiently phosphorylated on threonine 187 by Cdk2 which in turn promotes its SCF-Skp2-dependent degradation. This direct link between transforming tyrosine kinases and p27 may provide an explanation for Cdk kinase activities observed in p27 complexes and for premature p27 elimination in cells that have been transformed by activated tyrosine kinases.  相似文献

Chromaphyosemion killifishes are a very promising taxon for the examination of general principles and mechanisms underlying speciation. The polygamous mating system and high degree of sexual dimorphism in this group suggest that sexual selection and female mating preferences play a crucial role in the current radiation process. Because the emergence of mating preferences for individuals of the own population during allopatry is a necessary precondition for speciation by sexual selection, either via diverging mate recognition traits or reinforcement, we examined whether females of different nominal species (Chromaphyosemion bivittatum vs. Chromaphyosemion volcanum) or populations (C. bivittatum “Toko” vs. C. bivittatum “Ilor”) prefer their own males as mating partners. For this purpose, two different methods were used. First, we conducted simultaneous choice tests where females could choose between two different males, and second, we counted the eggs produced by females in consecutive matings with different males. Both approaches produced the same result: females of the populations under examination preferred to mate with their own males. Preference for own males was symmetrical in all tests. Males, on the other hand, do not discriminate between females, not even on the level of nominal species. Thus, our results are in accordance with the hypothesis that female choice is an important factor for speciation in Chromaphyosemion. Additionally, we present some preliminary data on post-zygotic isolation between the populations.  相似文献

Levels of inbreeding are highly variable in natural populations. Inbreeding can be due to random factors (like population size), limited dispersal, or active mate choice for relatives. Because of inbreeding depression, mating with kin is often avoided, although sometimes intermediately related individuals are preferred (optimal outbreeding). However, theory predicts that the advantages of mating with close kin can override the effects of inbreeding depression, but in the animal kingdom, empirical evidence for this is scarce. Here we show that both sexes of Pelvicachromis taeniatus, an African cichlid with biparental brood care, prefer mating with unfamiliar close kin over nonkin, suggesting inclusive fitness advantages for inbreeding individuals. Biparental care requires synchronous behavior among parents. Since parental care is costly, there is a conflict between parents over care, which can reduce offspring fitness. Relatedness is expected to enhance cooperation among individuals. The comparison of the parental behavior of in- and outbreeding pairs showed that related parents were more cooperative and invested more than unrelated parents. Since we found no evidence for inbreeding depression, our results suggest that in P. taeniatus, inbreeding is an advantageous strategy.  相似文献

Distichodus antonii is an endemic fish species of the Congo River basin in which the stocks of wild populations are threatened by overfishing pressure. Knowledge of its reproductive biology would be useful in consideration of conservation and management options for the species. Therefore, this study investigated changes in ovarian activity and levels of steroid profiles in wild populations in relation to variation in temperature and rainfall. Adult females (n = 101, body weight of 3 183 ± 14.75 g, SE) were captured monthly over one year (2013–2014). Apart from evaluation of oocyte diameters and gonad developmental stages, gonado-, hepato-, lipososomatic indices (GSI, HSI, LSI) and plasma levels of sex steroids (testosterone-T, estradiol-17β-E2) were determined. The results suggested a synchronous development of oocytes with two annual reproductive seasons over the one-year study. Plasma T and E2 levels peaked during spawning periods likely reflecting active oogenesis. The highest values of morphosomatic indices were observed during the longest rainfall period in September, and were associated with high steroidogenic activity evidenced by increased E2 production. In addition, more vitellogenic oocytes (September and October) were observed during the latter season than during the short rainy season (in May).  相似文献

Computer-manipulated visual stimuli are a well-established tool to experimentally study animal behaviour. They provide the opportunity to manipulate single or combined visual stimuli selectively, while other potentially confounding variables remain constant. A wide array of different presentation methods of artificial stimuli has been used in recent research. Furthermore, a wide range of basic hardware and software has been used to conduct experiments. The outcomes of experimental trials using computer-manipulated visual stimuli differed among studies. Failing or contradictory results were mostly discussed in a behavioural and ecological context. However, the results sometimes may be basically flawed due to methodological traps in the experimental design. Based on characteristics and restrictions of technical standards, we discuss which kinds of computer stimuli and visual display units are available today and their suitability for experimental trials when studying animal behaviour. A computer-manipulated stimulus displayed by a certain visual display unit may be accurate to investigate behaviour in a specific species, if various preconditions are met. However, simply due to technical characteristics of the visual display unit, the set-up may be unsuitable for other test species. Thus, future research should critically apply the included technique, with respect to both the intended kind of stimulus and the species under investigation. If these preconditions are met, computer-manipulated stimuli provide a high degree of standardization and the potential to display visual signals without losing a crucial amount of information from the native data.  相似文献

Background  

Large multigene sequence alignments have over recent years been increasingly employed for phylogenomic reconstruction of the eukaryote tree of life. Such supermatrices of sequence data are preferred over single gene alignments as they contain vastly more information about ancient sequence characteristics, and are thus more suitable for resolving deeply diverging relationships. However, as alignments are expanded, increasingly numbers of sites with misleading phylogenetic information are also added. Therefore, a major goal in phylogenomic analyses is to maximize the ratio of information to noise; this can be achieved by the reduction of fast evolving sites.  相似文献

Panorpa similis Esben-Petersen and Panorpa connexa MacLachlan are the most prominent Mecoptera in the Caucasian region. As our knowledge of mating systems and life histories of the different Panorpa species is unsatisfactory, it is the purpose of this contribution to add some interesting information on the life histories and mating behaviour of these two species. Further, we present results on successful breeding of these species, both in the field and the laboratory. Our data suggest that both species are monovoltine under the natural conditions in the Caucasian region, although our breeding experiments revealed a cryptic photoperiodic responsibility in a certain percentage of individuals of Panorpa connexa, which is completely absent in Panorpa similis. First, results on temporally and spatially differing foraging manners of the two species are presented. Male mating tactics of both species comprise copulations with one salivary mass as nuptial gift or copulations without any nuptial gift, but always without the use of force. This paper also gives information on egg deposition, egg hatching and larval development. Finally, we compare aspects of the mating systems for both species with those that have already been investigated for other Panorpa species.  相似文献
