It is unclear whether serum adiponectin concentrations diminish linearly with increasing adiposity and, if not, which factors codetermine this association. These issues were investigated cross-sectionally in 1188 men and women, representative of middle-aged and elderly Turkish adults. Serum total adiponectin was assayed by ELISA. Serum adiponectin values in men, though declining significantly in transition from the bottom to the mid tertile of body mass index (BMI) and waist circumference (WC), were similar in the two respective upper tertiles. In women, serum adiponectin concentrations were not significantly different in any tertile of these indices, were significantly correlated with BMI or WC within the low tertiles and not within the two higher tertiles. In a linear regression analysis for WC (or BMI) in a subset of the sample in which serum sex hormone-binding globulin (SHBG) was available and which additionally comprised adiponectin, fasting insulin and other confounders, only insulin and, in women SHBG, were significantly associated, but not adiponectin. In linear regression analyses for covariates of adiponectin in two models comprising 12 variables, insulin and SHBG concentrations were significantly associated in both genders though not BMI. Whereas in men HDL-cholesterol and CRP were covariates of adiponectin (both p<0.01), SHBG and apolipoprotein B positively associated in women (p<0.001), independent of BMI and fasting insulin levels. CONCLUSIONS: Relationship between excess adiposity and adiponectin levels is inconsistent in Turkish adults. Independently from obesity and hyperinsulinemia, serum adiponectin discloses significant relationship with inflammatory markers and HDL only in men, not in women in whom it is influenced by SHBG, with consequent attenuation of its anti-inflammatory activities.  相似文献

Prolactin (PRL) is a lactogenic hormone that plays a significant role in milk production; its depletion in sheep provokes a severe reduction of milk secretion. Thus, PRL also could be used as a positional marker gene associated with milk production and composition traits. Therefore, the purpose of the study was to identify genotype frequencies of single nucleotide polymorphisms the intron 2 in ovine PRL gene and its possible association genotypes with milk traits in dairy sheep breeds. The genetic structures of ovine PRL gene were examined by PCR-RFLP and DNA sequencing methods in three sheep populations. Four hundred and fifty blood and milk samples were used in the study, which included 150 samples from each of Sakiz, Akkaraman and Awassi ewes respectively. As a result, PRL genotype AA showed a strong association with milk yields content, whereas the animals carrying BB genotype had a higher fat percentage value in the three sheep breeds. Haplotype analysis of the obtained sequences showed the presence of 12 haplotypes in the PRL intron 2 region. In the present study, we have reported for the first time 48 SNPs of the PRL gene for intron 2 in dairy sheep breeds. These preliminary results indicate that the identified SNPs lend themselves readily for further research regarding physiological impacts such as milk production and reproductive traits in other dairy sheep populations.  相似文献

Vitrification is becoming a preferred method for pre‐implantation embryo cryopreservation. The objective of this study was to determine the differentially expressed genes of in vivo‐ and in vitro‐produced bovine embryos after vitrification. In vitro‐ (IVF) and in vivo‐derived (IVV) bovine blastocysts were identified as follows: in vitro‐produced fresh (IVF‐F), in vitro‐produced vitrified (IVF‐V), in vivo‐derived fresh (IVV‐F), in vivo‐derived vitrified (IVV‐V). The microarray results showed that 53 genes were differentially regulated between IVF and IVV, and 121 genes were differentially regulated between fresh and vitrified blastocysts (P < 0.05). There were 6, 268, 962, and 17 differentially regulated genes between IVF‐F × IVV‐F, IVF‐V × IVV‐V, IVF‐F × IVF‐V, and IVV‐F × IVV‐V, respectively (P < 0.05). While gene expression was significantly different between fresh and vitrified IVF blastocysts (P < 0.05), it was similar between fresh and vitrified IVV blastocysts. Significantly up‐regulated KEGG pathways included ribosome, oxidative phosphorylation, spliceosome, and oocyte meiosis in the fresh IVF blastocyst samples, while sphingolipid and purine metabolisms were up‐regulated in the vitrified IVF blastocyst. The results showed that in vitro bovine blastocyst production protocols used in this study caused no major gene expression differences compared to those of in vivoproduced blastocysts. After vitrification, however, in vitro‐produced blastocysts showed major gene expression differences compared to in vivo blastocysts. This study suggests that in vitro‐produced embryos are of comparable quality to their in vivo counterparts. Vitrification of in vitro blastocysts, on the other hand, causes significant up‐regulation of genes that are involved in stress responses. Mol. Reprod. We determined serum adiponectin's role as a biomarker of metabolic syndrome (MetS), type 2 diabetes (DM) and hypertension among Turkish adults who have a high prevalence of MetS. Individuals with measured serum adiponectin concentrations, constituting a random sample of Turkish adults, were studied cross‐sectionally. MetS was identified by criteria of the Adult Treatment Panel‐III modified for male abdominal obesity. Median age of 547 men and 652 women was 54 years. MetS was identified in 46%. Linear regression analysis among nine variables revealed homeostatic model assessment (HOMA) index in both sexes and C‐reactive protein (CRP) only in men as inversely associated covariates of adiponectin, and sex hormone‐binding globulin (SHBG) as positive covariate in women. Age‐adjusted sex‐specifically dichotomized high vs. low adiponectin levels were significantly associated with DM (odds ratio (OR) 0.55, P = 0.01) and hypertension (OR 0.64, P = 0.012) in women, but not in men. Further adjustment for smoking status and presence of high/low BMI did not alter this sex‐based relationship. As regards association with MetS, low adiponectin and high BMI interacted significantly in each sex. Yet adiponectin was associated only in men additively to the simultaneously adjusted five MetS components. We conclude that adiponectin concentrations, clearly linked to metabolic disorders, may diverge among sexes regarding protection against cardiometabolic risk through anti‐inflammatory or antioxidative function, Turkish men alone revealing significant dysfunction independent of obesity. This dysfunction may underlie also the association of adiponectin levels with MetS in men to be independent of the MetS components.  相似文献

Three cytoplasmic-nuclear male-sterile (CMS) lines, one each derived from Cajanus sericeus (A(1) cytoplasm), Cajanus scarabaeoides (A(2) cytoplasm), and Cajanus cajanifolius (A(4) cytoplasm), were crossed to 7 pigeonpea (Cajanus cajan (L.) Millsp.) cultivars in a line x tester mating scheme to study the fertility restoration of the CMS lines. Twenty-one F(1) hybrid combinations were planted in unreplicated 3-row plots in 3 environments. There was no effect of environments on the expression of fertility restoration. Pigeonpea cultivar ICPL 129-3 restored fertility in A(1) cytoplasm and maintained male sterility in the other 2 (A(2) and A(4)) cytoplasms. Among crosses involving CMS line (of A(4) cytoplasm) ICPA 2039 one hybrid combination was male-sterile and another male fertile. The remaining 5 combinations segregated for male-fertility (66-84% fertility restoration). Such testers can easily be purified for use in hybrid breeding programs by selfing and single-plant selection for 2-3 generations.  相似文献

Efforts were made to study microsporogenesis and genetics of fertility restoration of A(4) cytoplasmic-nuclear male-sterility (CMS) system in pigeonpea. The process of microsporogenesis in the male-sterile (ICPA 2039) and its maintainer (ICPB 2039) plants was normal up to the tetrad formation stage. The tapetal cells in the male-sterile anthers degenerated soon after tetrad formation, resulting in shriveled and degenerated microspores. In the maintainer plants, the tapetal cells were normal and microspores were functional. The breakdown of the tapetum before the completion of microsporogenesis was the major cause for the expression of male sterility in A(4) CMS system. The studies on the inheritance of fertility restoration showed that in 3 crosses, a single dominant gene; in 1 cross, 2 duplicate genes; and in another cross, 2 complimentary genes governed the fertility restoration.  相似文献
