In mammalian cultured cells the initiation of cytokinesis is regulated – both temporally and spatially – by the overlapping, anti-parallel microtubules of the spindle midzone. This region recruits several key central spindle components: PRC-1, polo-like kinase 1 (Plk-1), the centralspindlin complex, and the chromosome passenger complex (CPC), which together serve to stabilize the microtubule overlap, and also to coordinate the assembly of the cortical actin/myosin cytoskeleton necessary to physically cleave the cell in two. The localization of these crucial elements to the spindle midzone requires members of the kinesin superfamily of microtubule-based motor proteins. Here we focus on reviewing the role played by a variety of kinesins in both building and operating the spindle midzone machinery during cytokinesis. 相似文献

The aim of the study was to investigate how zinc deficiency and supplementation effect some hematologic parameters of rats performing swimming exercise. Forty adult male Spraque-Dawley rats were divided into 4 groups, zinc deficient swimming group (Group 1, n=10, zinc supplemented swimming group (Group 2, n=10), swimming control group (Group 3, n=10), and control group (Group 4, n=10). Blood samples were taken by decapitation and analyzed for the determination of erythrocyte, hemoglobin level, hematocrit, leukocyte, lymphocyte, platelet count and plasma zinc level at the end of the 4 week experiment. Erythrocyte count of group 1 was the lowest whereas erythrocyte count in group 3 was significantly lower than that in group 2 and 4 (p<0.05). Hemoglobin level of group 1 was significantly lower than that of groups 2 and 4 (p<0.05). Hematocrit was significantly lower in both group 1 and group 3 than both groups 2 and 4 (p<0.05). Lymphocyte count in group 2 was significantly higher than in all other groups (p<0.05). Platelet counts in group 2 was significantly lower than in all other groups (p<0.05). Our findings suggest that zinc deficiency effects the hematologic parameters mentioned negatively whereas zinc supplementation has a positive influence. 相似文献

The results are given of the two-year study of the new technology of soil remediation from oil contamination-CLEANSOIL combining physicochemical and biological methods of eliminating oil pollution. We and others have published on the rapid manufacture of micropellet tissues, typically formed from 100–500 cells each. The micropellet geometry enhances cellular biological properties, and in many cases the micropellets can subsequently be utilized as building blocks to assemble complex macrotissues. Generally, micropellets are formed from cells alone, however when replicating matrix-rich tissues such as cartilage it would be ideal if matrix or biomaterials supplements could be incorporated directly into the micropellet during the manufacturing process. Herein we describe a method to efficiently incorporate donor cartilage matrix into tissue engineered cartilage micropellets. We lyophilized bovine cartilage matrix, and then shattered it into microscopic pieces having average dimensions < 10 μm diameter; we termed this microscopic donor matrix “cartilage dust (CD)”. Using a microwell platform, we show that ~0.83 μg CD can be rapidly and efficiently incorporated into single multicellular aggregates formed from 180 bone marrow mesenchymal stem/stromal cells (MSC) each. The microwell platform enabled the rapid manufacture of thousands of replica composite micropellets, with each micropellet having a material/CD core and a cellular surface. This micropellet organization enabled the rapid bulking up of the micropellet core matrix content, and left an adhesive cellular outer surface. This morphological organization enabled the ready assembly of the composite micropellets into macroscopic tissues. Generically, this is a versatile method that enables the rapid and uniform integration of biomaterials into multicellular micropellets that can then be used as tissue building blocks. In this study, the addition of CD resulted in an approximate 8-fold volume increase in the micropellets, with the donor matrix functioning to contribute to an increase in total cartilage matrix content. Composite micropellets were readily assembled into macroscopic cartilage tissues; the incorporation of CD enhanced tissue size and matrix content, but did not enhance chondrogenic gene expression. 相似文献

Estrogen receptor a (ER??) gene has previously been found related with sexual development and reproduction. In this study, on the basis of the sequences of human, cattle and caprine estrogen receptor ?? (ER??) genes, available in the GenBank database, sets of PCR primers were designed and used to amplify the ovine ER?? gene exon 4 region. We identified six single nucleotide polymorphisms (SNP) in the ER?? exon 4. Some variations determined for exon 4 g.43A > G, p.T43A; g.49C > T, p.L49F; g.178A > T, p.T178S led to changes in the amino acids, but no amino acid changes were determined in g.18G > C, g.27C > T, g.96G > A. These fragments were deposited in the GenBank database under accession numbers: JF262030-JF262035. It was noted in particular that White Karaman and Awassi breeds were similar to each other, whereas the Chios breed had a different variation. 相似文献

Chum salmon Oncorhynchus keta from the Tugur River of Khabarovsk krai was studied. It was found that, in the river basin, chum salmon of two ecological
groups reproduces: the first spawns at river sections with a pronounced underflow and the second spawns at river sections
with a groundwater outlet. In the first anadromous migration, usually three maxima of the number of approaches of migrating
fish are observed. The first in time maximum is represented by early (or summer) chum salmon, the second includes migrants
of early and late (or autumn) form, and the third is represented by chum salmon of only late form. Forms of chum salmon from
the Tugur River basin different in dates of the run and the spawning sites differ not only in the ecology of reproduction
but also in biological indices. The later chum salmon is larger; its gonads at entry into the river for spawning are less
mature as compared with early chum salmon. The autumn form of chum salmon reproducing at key spawning grounds is distributed
more widely than the summer form. It is found along the Asian coast from Chukotka in the north to Japan and Korea in the south.
Summer chum salmon occupies only part of the range of the autumn form. 相似文献
