A specific PCR for the identification of K-12 strains, based on the genetic structure of the O-antigen gene cluster (rfb) of Escherichia coli K-12, is described. The assay clearly differentiates E. coli K-12-derived strains from other E. coli strains used in the laboratory or isolated from human and animal clinical specimens, from food, or from environmental samples. Moreover, lineages of K-12 strains can be distinguished with a second PCR based on the same gene cluster. The method presents a useful tool in identifying K-12 for monitoring strains which are used as biologically safe vehicles in biotechnological research, development, and production processes. 相似文献

Thearubigins are the most abundant group of phenolic pigments found in black tea accounting for an estimated 60% of the solids in a typical black tea infusion. Fifty years ago the term thearubigins was first introduced and up to now the chemical nature of the thearubigins remains largely unresolved if not mysterious despite many efforts clarifying their structure. This paper summarizes some of our attempts to clarify and elucidate the chemical nature of the thearubigins, presenting for 15 commercially representative teas data obtained using combustion analysis, IR spectroscopy, NMR spectroscopy, Diffusion NMR spectroscopy, UV-vis spectroscopy, Circular Dichroism spectroscopy and atomic force microscopy, MALDI-TOF-MS and ESI-FT-ICR-MS. The thearubigin fractions from these 15 teas are remarkably similar with respect to their spectroscopic fingerprint. The data obtained are consistent with the thearubigins being structures of not more than 2000 Da with more than 5000 individual chemical entities detected that are susceptible to concentration-driven aggregation in aqueous solution, and that retain the chiral properties of the flavanols and theaflavins. By applying petrolomics-style data interpretation strategies and by developing novel data interpretation strategies a structural model for the thearubigins was developed. 相似文献

Background
The Mannheimia species encompass a wide variety of bacterial lifestyles, including opportunistic pathogens and commensals of the ruminant respiratory tract, commensals of the ovine rumen, and pathogens of the ruminant integument. Here we present a scenario for the evolution of the leukotoxin promoter among representatives of the five species within genus Mannheimia. DNA microarrays used as 'genomic sensors' have great potential in clinical diagnostics. Biases inherent in random PCR-amplification, cross-hybridization effects, and inadequate microarray analysis, however, limit detection sensitivity and specificity. Here, we have studied the relationships between viral amplification efficiency, hybridization signal, and target-probe annealing specificity using a customized microarray platform. Novel features of this platform include the development of a robust algorithm that accurately predicts PCR bias during DNA amplification and can be used to improve PCR primer design, as well as a powerful statistical concept for inferring pathogen identity from probe recognition signatures. Compared to real-time PCR, the microarray platform identified pathogens with 94% accuracy (76% sensitivity and 100% specificity) in a panel of 36 patient specimens. Our findings show that microarrays can be used for the robust and accurate diagnosis of pathogens, and further substantiate the use of microarray technology in clinical diagnostics. 相似文献

The hydroxycinnamates of the leaves of 12 plants of the Astreraceae family, Achillea millefolium, Arnica montana, Artemesia dracunculus, Cichorium intybus, Cnicus benedictus, Cynara scolymus, Echinops humilis, Inula helenium, Lactuca sativa, Petasites hybridus, Solidago virgaurea, and Tanacetum parthenium were investigated qualitatively by LC-MSn. Thirty-nine chlorogenic acids were detected and all characterized to regioisomeric level on the basis of their fragmentation pattern in the tandem MS spectra, most of them for the first time from these sources with two of them previously not reported in nature. Both chlorogenic acids based on trans and cis-cinnamic acid substituents were identified. Assignment to the level of individual regioisomers was possible for seven caffeoylquinic acids (1-7), 11 dicaffeoylquinic acids (17-27), six feruloylquinic acids (9-14), two p-coumaroylquinic acids (15-16), two caffeoyl-feruloylquinic acids (28 and 29), four caffeoyl-p-coumaroylquinic acids (30-33), three dicaffeoyl-succinoylquinic acids (34-36), two dicaffeoyl-methoxyoxaloylquinic acids (37 and 38), and one tricaffeoylquinic acid (39). Furthermore, one caffeoylshikimic acid (40), one caffeoyltartaric acid (41), three dicaffeoyltartaric acids (42-44), and three caffeoyl-feruloyltartaric acids (45-47) were detected and shown to possess characteristic tandem MS spectra and were tentatively assigned on the basis of their retention time and previously developed hierarchical keys. 相似文献

Apple replant disease (ARD) is a frequently occurring plant disease, which causes retarded growth and mortality of young apple trees in replanted orchards. The aetiology is not well understood, but soil‐borne micro‐organisms are often discussed as primary causal agents of the replant problem. A greenhouse study was conducted in Laimburg, Italy, with orchard soils from the region, with the aim of obtaining information about the influence of soil biotic and abiotic factors on the aetiology of the disease. Apple rootstocks (M9) were planted into soils cultivated with apple trees that were either fumigated with chloropicrin or not fumigated, as well as mixtures of fumigated and non‐fumigated soils. In addition, uncultivated soils (from the inter‐row, from a fallow plot and from a meadow) were taken as controls. Various parameters were measured after 62 days in a controlled pot assay. Soils fumigated with chloropicrin resulted in higher apple shoot growth and lower microbial biomass carbon than non‐fumigated soils. Uncultivated soils had generally the highest microbial biomass carbon and the highest ergosterol contents. No considerable differences between basal respiration, ergosterol content, pH, electrical conductivity, and most nutrient and metal contents were observed between fumigated and non‐fumigated soils. Denaturing gradient gel electrophoresis gels of DNA extracted from the soils revealed differences in the fungal, bacterial and actinobacterial communities of the different soils, indicating significant shifts in microbial community composition after chloropicrin treatment.
