The aim of this study was to analyze the differential effects of three anti-CD4 monoclonal antibodies (mAbs) (with distinct epitope specifities) in the treatment of rat adjuvant arthritis (AA) and on T-cell function and signal transduction. Rat AA was preventively treated by intraperitoneal injection of the anti-CD4 mAbs W3/25, OX35, and RIB5/2 (on days -1, 0, 3, and 6, i.e. 1 day before AA induction, on the day of induction [day 0], and thereafter). The effects on T-cell reactivity in vivo (delayed-type hypersensitivity), ex vivo (ConA-induced proliferation), and in vitro (mixed lymphocyte culture) were assessed. The in vitro effects of anti-CD4 preincubation on T-cell receptor (TCR)/CD3-induced cytokine production and signal transduction were also analyzed. While preventive treatment with OX35 and W3/25 significantly ameliorated AA from the onset, treatment with RIB5/2 even accelerated the onset of AA by approximately 2 days (day 10), and ameliorated the arthritis only in the late phase (day 27). Differential clinical effects at the onset of AA were paralleled by a differential influence of the mAbs on T-cell functions, i.e. in comparison with OX35 and W3/25, the 'accelerating' mAb RIB5/2 failed to increase the delayed-type hypersentivity (DTH) to Mycobacterium tuberculosis, increased the in vitro tumor necrosis factor (TNF)-alpha secretion, and more strongly induced NF-kappaB binding activity after anti-CD4 preincubation and subsequent TCR/CD3-stimulation. Depending on their epitope specificity, different anti-CD4 mAbs differentially influence individual proinflammatory functions of T cells. This fine regulation may explain the differential efficacy in the treatment of AA and may contribute to the understanding of such treatments in other immunopathologies.  相似文献

An inverse correlation has been described between the levels of DNA methylation in specific segments of adenovirus DNA integrated into the genomes of transformed and tumor cells and the extent to which these segments are expressed as messenger RNA. In the adenovirus type 2 (Ad2)-transformed hamster cell lines HE2 and HE3, the virus-specific DNA binding protein (DBP) is not expressed, and the DNA in the DBP gene is completely methylated in all 5'-CCGG-3' sites. At least part of the late promoter/leader sequence of the DBP gene is present in cell lines HE2 and HE3. In line HE1, on the other hand, the DBP is expressed, and the DNA in the DBP gene is unmethylated at the 5'-CCGG-3' (HpaII) sites. The late promotor/leader sequence of the DBP gene is expressed in cytoplasmic RNA isolated from line HE1. The effect of DNA methylation has also been tested in vitro in a microinjection system using Xenopus laevis oocytes. Unmethylated DNA fragments of Ad2 (E2a region) have been found to serve as active templates. When the same fragments are methylated at the 5'-CCGG-3' sites by the HpaII DNA-methyltransferase, viral RNA synthesis is inhibited upon microinjection into oocyte nuclei. These results provide direct evidence for the notion that DNA methylated at highly specific sites is somehow involved in the regulation of gene expression.  相似文献

Marine animals related to Recent onychophorans form a significant component in Cambrian faunas. Twelve characters are analysed for homologies in the seven best known Cambrian onychophorans. New morphological evidence and homology analyses for several characters indicate an anteroposterior reversal of Hallucigenia and Microdictyon . Proposed expansion of the trunk in Microdietyon during compaction is rejected. A jaw is tentatively identified in Onychodictyon . The shape of the annulations and the disposition of the tenth leg pair in Aysheaia are reinterpreted, and the suggestion of two somites to the first appendage pair is rejected. A suggested morphocline may mirror the phylogeny of the group. The taxonomic confusion surrounding the supposed radiolarian family Eoconchariidae is cleared  相似文献

It has been shown before by (31)P NMR that Ras bound to the nonhydrolyzable GTP analogue guanosine 5'-O-(beta, gamma-imidotriphosphate) (GppNHp) exists in two conformations which are rapidly interconverting with a rate constant of 3200 s-1 at 30 degrees C [Geyer, M., et al. (1996) Biochemistry 35, 10308-10320]. Here we show that Ran complexed with GTP also exists in two conformational states, 1 and 2, which can be directly inferred from the occurrence of two (31)P NMR resonance lines for the gamma-phosphate group of bound GTP. The exchange between the two states is slow on the NMR time scale with a value of <200 s-1 at 5 degrees C for the corresponding first-order rate constants. In wild-type Ran, the equilibrium constant K' between the two states is 0.7 at 278 K, is different for various mutants, and is strongly dependent on the temperature. The standard enthalpy DeltaH degrees and the standard entropy DeltaS degrees for the conformational transitions determined from the NMR spectra are as follows: DeltaH degrees = 37 kJ mol-1 and DeltaS degrees = 130 J mol-1 K-1 for wild-type Ran.GTP. In complex with the Ran-binding protein RanBP1, one of the Ran.GTP conformations (state 2) is stabilized. The interaction of Ran with the guanine nucleotide exchange factor protein RCC1 was also studied by (31)P NMR spectroscopy. In the presence of nucleotide, the ternary complex of Ran.nucleotide.RCC1, an intermediate in the guanine nucleotide exchange reaction, could be observed. A model for the conformational transition of Ran.GTP is proposed where the two states observed are caused by the structural flexibility of the effector loop of Ran; in solution, state 2 resembles the GTP-bound form found in the crystal structure of the Ran-RanBP complex.  相似文献

The parasitoids of arthropod pests of apple and pear in northern and central Europe and their use as biological control agents are reviewed. The review demonstrates that apple and pear pests are host to a large and varied parasitoid fauna. All important pests are known to be host of parasitoids, but many parasitoids play only a minor part in regulating populations of their host. However, many parasitoid species are important natural enemies and some effectively regulate pest populations in unsprayed and/or commercial (insecticide sprayed) apple or pear orchards either individually or as part of parasitoid guilds. Exploitation/fostering of existing populations of parasitoids has been demonstrated to be an effective or partially effective approach for natural control of several important pest species. Important examples include natural regulation of the apple sawfly by Lathrolestes ensator and Aptesis nigrocincta, of the summer fruit tortrix moth by Colpoclypeus florus and Teleutaea striata, of leaf midges by Platygaster demades, of woolly aphid by Aphelinus mali and of leaf mining moths by guilds of parasitoid species. Introduction of parasitoids is an alternative approach to the exploitation of parasitoids already present in the orchard. This approach has been little explored and its success rate has been low, mainly confined to the control of non-indigenous pests by introducing parasitoids from their native region. Mass production methods for parasitoids are difficult and costly and are likely to be economic only where long-term populations can be established. Even where low cost mass culture techniques are developed, the degree of control may not be high enough to prevent economic pest damage as demonstrated by negative results with mass release of Trichogramma egg parasites for control of tortricids in orchards. Suitability of the orchard habitat is recognized as crucial to the success of individual parasitoids. Key requirements are adequate populations of the pest(s) and/or alternative hosts, suitable shelter, overwintering sites or food sources and avoidance of harmful effects of pesticides. Many species are highly sensitive to broad-spectrum insecticides, especially in the adult life-stage. Avoiding the harmful affects of insecticides is crucial to successful exploitation. The use of insecticides needs to be avoided, either altogether or at crucial times in the parasitoids' life cycle, or less harmful alternatives need to be used. Numerous parasitoids could potentially be exploited as biological control agents but hitherto have received little attention because little is known about them and/or because they are sensitive to broad-spectrum pesticides and are thus virtually absent from commercial orchards. The aim of future studies should be to develop effective strategies for establishing equilibria between important pests and their parasitoids, with pest damage rarely exceeding the economic threshold.  相似文献

The impact category ‘land use’ describes in the Life Cycle Assessment (LCA) methodology the environmental impacts of occupying, reshaping and managing land for human purposes. Land use can either be the long-term use of land (e.g. for arable farming) or changing the type of land use (e.g. from natural to urban area). The impact category ‘land use’ comprises those environmental consequences, which impact the environment due to the land use itself, for instance through the reduction of landscape elements, the planting of monocultures or artificial vegetation, or the sealing of surfaces. Important environmental consequences of land use are the decreasing availability of habitats and the decreasing diversity of wildlife species. The assessment of the environmental impacts of land use within LCA studies is the objective of this paper. Land use leads to a degradation of the naturalness of the area utilised. In this respect the naturalness of any area can be defined as the sum of land actually not influenced by humans and the remaining naturalness of land under use. To determine the remaining naturalness of land under use, this study suggests applying the Hemeroby concept. “Hemeroby is a measure for the human influence on ecosystems” (Kowarik 1999). The Hemeroby level of an area describes the intensity of land use and can therefore be used to characterise different types of land use. Characterization factors are proposed, which allow calculating the degradation of the naturalness of an area due to a specific type of land use. Since the resource ‘nature/naturalness’ is on a larger geographical scale by far not homogeneous, the assessment of land use needs to be regionalised. Therefore, the impact category ‘land use’ has been subdivided into the impact sub-categories ‘land use in European biogeographic regions’. Following the general LCA framework, normalization values for the impact sub-categories are calculated in order to facilitate the evaluation of the characterization results with regard to their share in a reference value. Weighting factors, which enable an aggregation of the results of the different land use sub-categories and make them comparable to other impact categories (e.g. climate change or acidification) are suggested based on the assumption that the current land use pattern in the European biogeographic regions is acceptable.  相似文献

1. The relationships between parasitoid egg load, size, and age (3–72 h) for Trichogramma minutum, T. platneri, and T. pretiosum, reared from two factitious hosts, Ephestia kuehniella and Sitotroga cerealella, were evaluated to test the hypothesis that 24‐h egg load can be used to estimate the fecundity of Trichogramma parasitoids. 2. Egg load increased in relation to female age over the first 3 days of adult life for all three Trichogramma species to a mean egg storage capacity of 46.7 eggs for T. minutum, 41.1 for T. pretiosum, and 35.7 for T. platneri. At 24 h of age, T. minutum had matured enough eggs to fill 67% of its storage capacity, in comparison with 74% for T. pretiosum and 91% for T. platneri. There was a positive relationship between egg load and parasitoid size for all ages of the three Trichogramma species reared from both hosts (with the exception of T. platneri at 3 h post emergence), accounting for 14–69% of the variance in egg load. 3. The potential fecundity, realised (3 day cumulative) fecundity, and oviposition rate (potential fecundity/longevity) of T. platneri were all related linearly to size‐dependent variation in 24‐h egg load, but only the realised fecundity of T. pretiosum, and none of the reproductive characteristics of T. minutum. It is suggested that 24‐h egg load may not be an accurate measure of egg storage capacity in parasitoids and should be used cautiously to represent fecundity. 4. The potential fecundity of seven Trichogramma species reared from E. kuehniella varied from 55 to 150, but neither potential fecundity nor oviposition rate was related significantly to egg load (represented by eggs laid during first 24 h). Selection to avoid egg depletion in the attack of gregarious hosts appears most likely to account for the variation in potential fecundity among Trichogramma species.  相似文献

We report abnormal maternal laboratory parameters in twin-to-twin transfusion syndrome (TTTS) at mid-pregnancy. A retrospective chart review was undertaken of 109 patients with TTTS evaluated for placental laser surgery. Complete blood count (CBC), blood type and Rh factor, urine analysis and serum chemistry panel were obtained preoperatively, with the CBC and serum albumin repeated on the first postoperative day. The mean gestational age was 21.2+/-1.7 weeks. Initial abnormal values included hematocrit (32.1+/-3.0%), hemoglobin (11.0+/-1.03 g/dl), serum magnesium (1.71+/-0.17 mg/dl), total protein (6.08+/-0.55 g/dl) and albumin (3.06+/-0.34 g/dl). Despite minimal blood loss and conservative fluid replacement mean hematocrit, hemoglobin, and albumin were 27.3+/-2.74%, 9.3+/-0.94 g/dl and 2.56+/-0.23 g/dl, respectively on postoperative day one. Weight gain (8.0+/-5.5 lb.) and low urinary output were characteristic peri-operative events. Maternal hypoproteinemia and anemia occur in TTTS at mid-pregnancy. This may contribute independently to amniotic fluid production rates in the fetuses, and explain in part the maternal sensitivity to intravenous fluids in multiple pregnancy.  相似文献
