Objectives: The objective of this study was to study the association of polymorphisms MTHFR C677T (rs1801133) and MTR A2756G (rs1805087) with the risk of varicose veins in ethnical Russians.Methods: We genotyped 475 patients with varicose veins, 168 individual without chronic venous disease, and the population-based group of 896 subjects. Association was studied using logistic regression analysis adopting co-dominant, additive, recessive, and dominant models of inheritance.Results: None of the polymorphisms showed a statistically significant association with the risk of varicose veins.Conclusions: Our results provide evidence that the studied polymorphisms do not contribute to genetic susceptibility to varicose veins in ethnical Russians. 相似文献
Exact mechanisms of autoimmune disease development are still yet unknown. However, it is known that the development of autoimmune diseases is associated with defects in the immune system, namely, the violation of the bone marrow hematopoietic stem cells (HSCs) differentiation profiles. Different characteristics of autoimmune reaction development in experimental autoimmune encephalomyelitis (EAE) prone Th mice characterizing T-lymphocytes response were analyzed using standard approaches. Profiles of several HSCs differentiation of bone marrow (BFU-E, CFU-E, CFU-GM, CFU-GEMM, T- and B-lymphocytes) of Th male and female mice during spontaneous development of EAE were noticeably different. Patterns of total lymphocytes, B- and T-cells proliferation in several different organs (bone marrow, blood, spleen, thymus, and lymph nodes) were also remarkably different. In addition, there were in time noticeable differences in their changes for some organs of male and female mice. Characters of changes in the profiles of CD4 and CD8 cells proliferation in some organs not always coincide with those for total T lymphocytes. The changes in the differentiation profiles of HSCs and the level of lymphocytes proliferation in the bone marrow and other organs were associated with the increase in the concentration of antibodies against DNA, myelin basic protein, and myelin oligodendrocyte glycoprotein, and catalytic antibodies hydrolyzing these substrates. Despite some differences in changes in the analyzed parameters, in general, the spontaneous development of EAE in male and female mice occurs to some extent in a comparable way.
The ability of the Alcaligenes faecalis 2 strain to utilize acrylamide and acrylic acid upon cultivation with these compounds as the only sources of carbon and energy has been investigated. Complete utilization of the acrylic acid present in the medium at concentrations below 0.113 g/L was observed by cultivation day 5, at a concentration of 0.225 g/L by day 7, and at a concentration of 0.45 g/L by day 17. Complete utilization of the acrylamide present in the medium at concentrations below 0.4 g/L was observed by day 5, at a concentration of 0.9 g/L by day 7, and at a concentration of 1.8 g/L by day 20. Importantly, bacterial growth did not start before complete transformation of acrylamide into acrylic acid. The rate of acrylamide transformation by growing bacteria and a cell suspension in the stationary growth phase amounted to 12.5 mg/L h at a cell concentration of 610 mg/L and 300 mg/L h, at a concentration of 1500 mg/L. A. faecalis 2 cells immobilized on BVV-22 basalt fibers and Carbopon-B-aktiv at concentrations of 3000 and 800 mg dry cells/L, respectively, transformed acrylamide at a rate of 1200 mg/L h. 相似文献
Hydrogen peroxide is an important second messenger controlling intracellular signaling cascades by selective oxidation of redox active thiolates in proteins. Changes in intracellular [H2O2] can be tracked in real time using HyPer, a ratiometric genetically encoded fluorescent probe. Although HyPer is sensitive and selective for H2O2 due to the properties of its sensing domain derived from the Escherichia coli OxyR protein, many applications may benefit from an improvement of the indicator’s dynamic range. We here report HyPer-2, a probe that fills this demand. Upon saturating [H2O2] exposure, HyPer-2 undergoes an up to sixfold increase of the ratio F500/F420 versus a threefold change in HyPer. HyPer-2 was generated by a single point mutation A406V from HyPer corresponding to A233V in wtOxyR. This mutation was previously shown to destabilize interface between monomers in OxyR dimers. However, in HyPer-2, the A233V mutation stabilizes the dimer and expands the dynamic range of the probe. 相似文献
Silver staining of the nucleoli in pig embryo kidney cells (PK) was studied during the cell cycle and also upon mature nucleoli modifications induced by UV microirradiation. During anaphase only four silver-stained granules were revealed in each daughter set of chromosomes in the four nucleolus-organizing regions (NORs). In the following 1-2 hours, the number of granules in the NORs rapidly increased up to 25-30 per nucleus. During the next 20-25 hours of the cell cycle, the number of silver-stained granules was slowly doubling as the nucleoli grew in size. UV microirradiation of one nucleolus in the nucleus with two nucleoli induced a profound degradation of the injured nucleolus and a compensatory hypertrophy of the intact one. Such nucleolar modifications were accompanied by redistribution of the silver-stained granules between the injured and non-injured nucleoli and by alterations in the levels of nucleolar RNA synthesis in the NORs. These data support a hypothesis that silver-stained proteins may be involved in the regulation of the nucleolar activity. 相似文献
Strains of basidiomycetous yeasts isolated from different sources were studied in order to determine the content of carotenoid pigments and ubiquinone Q10 for subsequent selection work to obtain producers of these substances. The high specific productivity of carotenoids (600–700 mg/g) was revealed in the representatives of the following species: Cystofilobasidium capitatum, Rhodosporidium diobovatum, R. sphaerocarpum, Rhodotorula glutinis, Rh. minuta, and Sporobolomyces roseus. The ratio of the major pigments (torulene, torularhodine, and β-carotene) in the representatives of different species was studied. Certain specific features of pigment formation in relation to the taxonomic position of the yeasts were determined. Eurybiont species with substantial ecological lability are the most active producers of carotenoids and ubiquinone Q10 among the epiphytes. It is the first time a comparative analysis of the coenzyme Q10 content in different taxa has been performed using several strains of the same species. The maximal coenzyme Q10 production (1.84 mg/g of dry biomass) was found in the yeast species R. sphaerocarpum. 相似文献
A new variety of Actinoplanes, named Actinoplanes ianthinogenes subsp. octamycini Gause et Svechnikova is described. It produces an antibiotic complex consisting of purpuromycin and a new antibiotic belonging to the group of polyens, i. e. octamycin.
