Mud (with or without Chironomus) taken from the bottom of a shallow (6 m) eutrophic reservoir, was maintained (about 3 weeks) in trays supported 30 cm and 70 cm above the bottom. At the end of the experiment it contained 7-9 times more Chironomus plumosus individuals than did the mud at the bottom. Numbers of young (≤12 mm) larvae were up to 18 times higher, and youngest (≤6 mm) larvae (absent in the bottom at the end of the experiment) were abundant. Such patterns were more evident at the higher (70 cm) than the lower (30 cm) level. Numbers of the oldest (≥18 mm) larvae at the higher level were only 6 times higher (whilst more than 8 times at the lower level) than at the bottom, probably due to quicker development and emergence in higher trays. So the production of the population increased with the increasing distance from the bottom. In the tray at the bottom (which could protrude 1–2 cm above it) numbers were twice those outside it, and the youngest larvae occurred. There was a slight current at the research site. Nevertheless, short time oxygen deficits occurred periodically, and were probably the main cause of considerable reductions in Chironomus numbers at the bottom during the experiment.  相似文献

It was found that both normal human myometrium and uterine leiomyoma contain several glycosaminoglycans. In contrast to many normal and tumour tissues the amount of hyaluronic acid is very low and the proportional amount of sulphated glycosaminoglycans is distinctly higher. It is of interest that heparan sulphate is the major glycosaminoglycan component both in normal myometrium, and in leiomyoma. The amount of hyaluronic acid in myometrium and in the leiomyoma is very low. No significant change in hyaluronate content was observed during the tumour growth. In contrast to that the amount of some sulphated glycosaminoglycans (heparan sulphate, keratan sulphate, chondroitin sulphates and heparin) distinctly increased. It is suggested that some of the GAGs participate in the creation of a storage depot for biologically active molecules (growth factors, enzymes) which are thereby stabilized and protected. Hydrolytic degradation of some GAGs may result in the release of some cytokines which may promote the tumour growth and stimulate collagen biosynthesis by tumour cells.  相似文献

A cross-platform public domain PC image-analysis program for the comet assay   总被引:47,自引:0,他引:47  
The single-cell gel electrophoresis, also known as the comet assay, has gained wide-spread popularity as a simple and reliable method to measure genotoxic and cytotoxic effects of physical and chemical agents as well as kinetics of DNA repair. Cells are generally stained with fluorescent dyes. The analysis of comets--damaged cells which form a typical comet-shaped pattern--is greatly facilitated by the use of a computer image-analysis program. Although several image-analysis programs are available commercially, they are expensive and their source codes are not provided. For Macintosh computers a cost-free public domain macro is available on the Internet. No ready for use, cost-free program exists for the PC platform. We have, therefore, developed such a public domain program under the GNU license for PC computers. The program is called CASP and can be run on a variety of hardware and software platforms. Its practical merit was tested on human lymphocytes exposed to gamma-rays and found to yield reproducible results. Insulin resistance in skeletal muscle is a hallmark feature of type 2 diabetes. An increasing number of enzymes and metabolic pathways have been implicated in the development of insulin resistance. However, the primary cellular cause of insulin resistance remains uncertain. Proteome analysis can quantitate a large number of proteins and their post-translational modifications simultaneously and is a powerful tool to study polygenic diseases like type 2 diabetes. Using this approach on human skeletal muscle biopsies, we have identified eight potential protein markers for type 2 diabetes in the fasting state. The observed changes in protein expression indicate increased cellular stress, e.g. up-regulation of two heat shock proteins, and perturbations in ATP (re)synthesis and mitochondrial metabolism, e.g. down-regulation of ATP synthase beta-subunit and creatine kinase B, in skeletal muscle of patients with type 2 diabetes. Phosphorylation appears to play a key, potentially coordinating role for most of the proteins identified in this study. In particular, we demonstrated that the catalytic beta-subunit of ATP synthase is phosphorylated in vivo and that the levels of a down-regulated ATP synthase beta-subunit phosphoisoform in diabetic muscle correlated inversely with fasting plasma glucose levels. These data suggest a role for phosphorylation of ATP synthase beta-subunit in the regulation of ATP synthesis and that alterations in the regulation of ATP synthesis and cellular stress proteins may contribute to the pathogenesis of type 2 diabetes.  相似文献

Rhodopsins are currently known to belong to two distinct protein families. The visual rhodopsins, found in eyes throughout the animal kingdom, are photosensory pigments. Archaeal rhodopsins, found in extreme halophiles, function as light-driven proton pumps (bacteriorhodopsins), chloride ion pumps (halorhodopsins), or photosensory receptors (sensory rhodopsins). Light absorption by rhodopsins triggers their characteristic photoconversion extending into the (milli)second time range. There are three main paradigms of rhodopsins photoconversion. (1) Initiation of the trans-cis isomerization is the very primary consequence of light absorption. (2) Rhodopsins store light energy via the charge-separation mechanism (the charge of Schiff base is separated from its counterion). (3) Full trans-cis isomerization of the chromophore is a prerequisite for the full biological activity of rhodopsins. These paradigms will be questioned.  相似文献

Abasic sites and non-conventional 3'-ends, e.g. 3'-oxidized fragments (including 3'-phosphate groups) and 3'-mismatched nucleotides, arise at significant frequency in the genome due to spontaneous decay, oxidation or replication errors. To avert the potentially mutagenic or cytotoxic effects of these chromosome modifications/intermediates, organisms are equipped with apurinic/apyrimidinic (AP) endonucleases and 3'-nucleases that initiate repair. Ape1, which shares homology with Escherichia coli exonuclease III (ExoIII), is the major abasic endonuclease in mammals and an important, yet selective, contributor to 3'-end processing. Mammals also possess a second protein (Ape2) with sequence homology to ExoIII, but this protein exhibits comparatively weak AP site-specific and 3'-nuclease activities. Prompted by homology modeling studies, we found that substitutions in the hydrophobic pocket of Ape1 (comprised of F266, W280 and L282) reduce abasic incision potency about fourfold to 450,000-fold, while introduction of an ExoIII-like pocket into Ape2 enhances its AP endonuclease function. We demonstrate that mutations at F266 and W280 of Ape1 increase 3' to 5' DNA exonuclease activity. These results, coupled with prior comparative sequence analysis, indicate that this active-site hydrophobic pocket influences the substrate specificity of a diverse set of sequence-related proteins possessing the conserved four-layered alpha/beta-fold. Lastly, we report that wild-type Ape1 excises 3'-mismatched nucleotides at a rate up to 374-fold higher than correctly base-paired nucleotides, depending greatly on the structure and sequence of the DNA substrate, suggesting a novel, selective role for the human protein in 3'-mismatch repair.  相似文献

Summary Trypsin inhibitor SFTI-1 isolated from sunflower seeds (comprising 14 amino acid residues and two cycles: head-to-tail cyclisation and disulfide bridge) is the smallest naturally occurring plant serine proteinase inhibitor. In our recent paper we have shown that the elimination head-to-tail cyclisation did not change trypsin inhibitory activity as judged by measured by association equilibrium constants K a . The removal of disulfide bridge produced 2.4-fold lower activity. In the present paper we described chymotrypsin inhibitory activity. SFTI-1 inhibits significantly lower bovine α-chymortypsin (K a =(5.20±1.56)×106 M−1). The activity of the analogue with disulfide bridge only was practically the same, whereas the K a value determined for homodetic peptide was almost 3-fold lower. Considering the results obtained and the recent literature data we postulate the lower inhibitory activity against both enzymes of the analogue with head-to-tail cyclisation only reflect its lower proteolytic stability.  相似文献
