Seed Rain in High-Altitude Restoration Plots in Switzerland   总被引:2,自引:0,他引:2  
Seed rain was studied in restoration plots installed in 1985 and 1987, respectively, on an alpine downhill ski run at circa 2,500 m above sea level. The study was initiated in late autumn 1996 and completed in autumn 1998; it included temporal and spatial variation in density per m2, as well as alpha diversity (species richness), and species composition of the seed rain versus that of the resident vegetation. This is the first report on post-restoration monitoring of seed rain above the timberline. Seed rain density and alpha diversity varied seasonally, with the first peak occurring immediately after spring snowmelt and the second in early autumn. The density of seed rain varied between plots and years (1,528–1,778 seeds per m2 in one plot [RPF] versus 1,096–3,557 seeds per m2 in another plot [RPG]). Total species number per plot was nearly twice as high in RPF as in RPG in both study years. Seed rain totaled 18 species; all but one represented either transplants introduced in restoration or colonizers established in the plots soon after restoration. Distribution of species in seed rain was largely asymmetric and only a few species provided substantial contributions. Composition of species and their respective contribution to seed rain differed between plots and was clearly influenced by performance of some species used in restoration as transplants; together they provided as much as 51% of the total seed rain. The results of the study demonstrate that restoration enhanced increase of species richness as well as seed rain in situ.  相似文献

Short sediment cores from two small oxbow lakes located in the Vistula valley near Warsaw (Poland) were analysed for chemical composition and zooplankton (Cladocera) remains. The results of these analyses were evaluated using ordination techniques and the results were used to reconstruct the history of human impact of the lakes. The cores were dated using the 210Pb method. These analyses show that the collected sediments (60 cm- and 67 cm-long-cores) were deposited during the last 150–200 years. Cladocera assemblages were different in the two lakes; however, Bosmina longirostris was dominant among subfossil Cladocera species in both lakes. The presence of this species indicates the high trophic state of the lakes. Changes in Cladocera species composition suggest that a significant increase in the lakes’ trophic levels has occurred since the 1920s. The change could be related to the construction of flood dikes along the river Vistula, the isolation of the studied lakes from the river, changes in land use, or some combination of these factors. The chemical composition of sediments, particularly the increase in phosphorus and sulphur concentrations, confirms that the construction of dikes against flood was the major factor inducing eutrophication of the oxbow lakes. This study shows that the Warsaw small water bodies are especially sensitive to local disturbances caused by human activity, mainly water management, land use changes and agriculture. The acid-base properties and Cu(II), Ni(II), Ag(I) and Hg(II) binding abilities of PAMAM dendrimer, L, and of the simple model compounds, the tetraamides of EDTA and PDTA, L1, were studied in solution by pH-metric methods and by 1H NMR and UV-Vis spectroscopy. PAMAM is hexabasic and six pKa values have been determined and assigned. PAMAM forms five identifiable complexes with copper(II), [CuLH4]6+, [CuLH2]4+, [CuLH]3+, [CuL]2+ and [CuLH-1]+ in the pH range 2-11 and three with nickel(II), [NiLH]3+, [NiL]2+ and [NiLH-1]+ in the pH range 7-11. The complex [CuLH4]6+, which contains two tertiary nitrogen and three amide oxygen atoms coordinated to the metal ion, is less stable than the analogous EDTA and PDTA tetraamide complexes [CuL1]2+, which contain two tertiary nitrogen and four amide oxygen atoms, due to ring size and charge effects. With increasing pH, [CuLH4]6+ undergoes deprotonation of two coordinated amide groups to give [CuLH2]4+ with a concomitant change from O-amide to N-amidate coordination. Surprisingly and in contrast to the tetraamide complexes [CuL1]2+, these two deprotonation steps could not be separated. As expected the nickel(II) complexes are less stable than their copper(II) analogues. The tetra-N-methylamides of EDTA, L1(b), and PDTA form mononuclear and binuclear complexes with Hg(II). In the case of L1(b) these have stoichiometries HgL1(b)Cl2, [HgL1(b)H−2Cl2]2−, [Hg2L1(b)Cl2]2+, Hg2L1(b)H−2Cl2 and [Hg2L1(b)H−5Cl2]3−. Based on 1H NMR and pH-metric data the proposed structure for HgL1(b)Cl2, the main tetraamide ligand containing species in the pH range <3-6.5, contains L1(b) coordinated to the metal ion through the two tertiary nitrogens and two amide oxygens while the structure of [HgL1(b)H−2Cl2]2−, the main tetraamide ligand species at pH 7.5-9.0, contains the ligand similarly coordinated but through two amidate nitrogen atoms instead of amide oxygens. The proposed structure of [Hg2L1(b)Cl2]2+, a minor species at pH 3-6.5, also based on 1H NMR and pH-metric data, contains each Hg(II) coordinated to a tertiary amino nitrogen, two amide oxygens and a chloride ligand while that of [Hg2L1(b)H−5Cl2]3−, contains each Hg(II) coordinated to a tertiary amino nitrogen, two amidate nitrogens, a chloride and a hydroxo ligand in the case of one of the Hg(II) ions. The parent EDTA and PDTA amides only form mononuclear complexes. PAMAM also forms dinuclear as well as mononuclear complexes with mercury(II) and silver(I). In the pH range 3-11 six complexes with Hg(II) i.e. [HgLH4Cl2]4+, [HgLH3Cl2]3+, [Hg2LCl2]2+, [Hg2LH−1Cl2]+, [HgLH−1Cl2] and [HgLH−2Cl2]2− were identified and only two with Ag(I), [AgLH3]4+ and [Ag2L]2+. Based on stoichiometries, stability constant comparisons and 1H NMR data, structures are proposed for these species. Hence [HgLH4Cl2]4+ is proposed to have a similar structure to [CuLH4]6+ while [Hg2LCl2]2+has a similar structure to [Hg2L1(b)H−5Cl2]3−.  相似文献

Zusammenfassung Es wurden mit histochemischen Methoden Milchdrüsen von Meerschweinchen in physiologischer und durch Kastration während der Laktationsperiode hervorgerufener Involution untersucht. Nachgewiesen wurden alkalische und saure Phosphatase (APh und SPh), unspezifische Esterase, Sukzinodehydrase, SH-Gruppen, Nukleinsäuren und Lipide. — Die Aktivität der SPh nimmt während der Involution zu. Die Bedeutung dieses Enzyms für den Rückbildungsprozeß des Drüsengewebes wird diskutiert. — Das Vorkommen der APh-Aktivität in Drüsenendstücken im Involutionsstadium kann auf die Beteiligung dieses Enzyms an der Rückresorption der mit der Milch ausgeschiedenen Substanzen hinweisen. — Zwischen der Aktivität der unspezifischen Esterase und dem Gehalt sowie der Lokalisation von Lipiden besteht eine Abhängigkeit. — Es konnten keine Unterschiede in der Sukzinodehydraseaktivität und dem DNS-Gehalt aufgezeigt werden. — Die Verminderung von SH-Gruppen und RNS hängt mit dem Aufhören der Sekretionsproduktion durch die Zellen der Drüsenendstücke der Milchdrüsen zusammen.
Histochemical studies on the involution mechanism of the mammary gland
Summary Histochemical methods were used to study mammary glands of guinea pigs in the course of physiologic involution and that induced by castration during lactation. Alkaline and acid phosphatases, unspecific esterase, succinic dehydrogenase, SH groups, nucleic acids and lipids were determined. Acid phosphatase activity was found to be increased in mammary glands, subject to involution. The participation of the enzyme in the involutionary process of the gland tissue is discussed. The distribution of alkaline phosphatase in the secretory sections of the gland during involution would suggest the participation of the enzyme in the reabsorption of the substance secreted with milk. A correlation existing between the activity of unspecific esterase, the level and distribution of lipids in the mammary gland could be established. No differences were detected in the activity of succinic dehydrogenase and DNA level. A decrease in SH groups and RNA content is related to cessation of milk secretion.

Obtaining oat DH lines is only effective via wide crossing with maize. Seven hundred haploid embryos from 21 single F1 progeny obtained from wide crosses with maize were isolated, divided into four groups according to their size (<0.5 mm, 0.5–0.9 mm, 1.0–1.4 mm, and ≥1.5 mm), and transferred into 190–2 regeneration medium with different growth regulators: 0.5 mg L?1 kinetin (KIN) and 0.5 mg L?1 1-naphthaleneacetic acid (NAA); 1 mg L?1 zeatin (ZEA) and 0.5 mg L?1 NAA; or 1 mg L?1 dicamba (DIC), 1 mg L?1 picloram (PIC), and 0.5 mg L?1 kinetin (KIN). Among all isolated embryos, approximately 46.1% were between 1.0–1.4 mm, while the smallest group of embryos (7.1%) were those <0.5 mm. The ability of haploid embryos to germinate varied depending on oat genotypes and the size of embryos. Haploid embryos <0.5 mm were globular and did not germinate, whereas embryos ≥1.5 mm had clearly visible coleoptiles, radicles, and scutella, and were able to germinate. Germination of oat haploid embryos varied depending on growth regulators in the regeneration medium. Most haploid embryos germinated on medium with 0.5 mg L?1 NAA and 0.5 mg L?1 KIN, while the fewest germinated on medium with 1 mg L?1 DIC, 1 mg L?1 PIC, and 0.5 mg L?1 KIN. One hundred thirty germinated haploid embryos converted into haploid plants. Fifty oat DH lines were obtained after colchicine treatment.  相似文献

The aim of this study was to evaluate the impact that 6‐O‐(3″, 4″‐di‐Otrans‐cinnamoyl)‐α‐ l ‐rhamnopyranosylcatalpol (Dicinn) and verbascoside (Verb), two compounds simultaneously reported in Verbascum ovalifolium, have on tumor cell viability, apoptosis, cell cycle kinetics, and intracellular reactive oxygen species (ROS) level. At 100 µg/mL and 48 hours incubation time, Dicinn and Verb produced good cytotoxic effects in A549, HT‐29, and MCF‐7 cells. Dicinn induced cell‐cycle arrest at the G0/G1 phase and apoptosis, whereas Verb increased the population of subG1 cells and cell apoptosis rates. Furthermore, the two compounds exhibited time‐dependent ROS generating effects in tumor cells (1‐24 hours). Importantly, no cytotoxic effects were induced in nontumor MCF‐10A cells by the two compounds up to 100 µg/mL. Overall, the effects exhibited by Verb in tumor cells were more potent, which can be correlated with its structural features, such as the presence of phenolic hydroxyl groups.  相似文献

The phenotypic differentiation of relic P. sylvestris in southern Europe and southwestern Asia was verified using thirty-two populations sampled from the Iberian Peninsula, Massif Central, Balkan Peninsula, Crimea and Anatolia. Twenty-one morphological and anatomical needle traits and 18 cone morphological characteristics were examined to describe the population diversity and differentiation. The needle characters were not correlated to those of cone. The differences between regions were significant based on 12 needle and 9 cone characteristics, suggesting spatial isolation. The differentiation between the Iberian and Anatolian populations was the highest, which indicates the isolation by distance. The high level of morphological differentiation was also found among Iberian populations, supporting the already known complex history of the species in that region. Populations within other regions were differentiated at lower levels; however, the West Anatolian populations differed morphologically from the eastern ones. The described pattern of morphological differentiation supports the idea of the long-lasting existence of P. sylvestris in the south-European and Anatolian mountain regions. To conserve this variation, seed transfer between regions in the forest economy should be restricted.  相似文献

The expression of transmembrane transporter multidrug resistance-associated protein 1 (MRP1) confers the multidrug-resistant phenotype (MDR) on cancer cells. Since the activity of the other MDR transporter, P-glycoprotein, is sensitive to membrane perturbation, we aimed to check whether the changes in lipid bilayer properties induced by flavones (apigenin, acacetin) and flavonols (morin, myricetin) were related to their MRP1 inhibitory activity. All the flavonoids inhibited the efflux of MRP1 fluorescent substrate from human erythrocytes and breast cancer cells. Morin was also found to stimulate the ATPase activity of erythrocyte ghosts. All flavonoids intercalated into phosphatidylcholine bilayers as judged by differential scanning calorimetry and fluorescence spectroscopy with the use of two carbocyanine dyes. The model of an intramembrane localization for flavones and flavonols was proposed. No clear relationship was found between the membrane-perturbing activity of flavonoids and their potency to inhibit MRP1. We concluded that mechanisms other than perturbation of the lipid phase of membranes were responsible for inhibition of MRP1 by the flavonoids.  相似文献

Application of somatic embryogenesis to Pinus strobus clonal propagation and genetic improvement was hampered by the difficulty in achieving synchronous maturation of a large number of somatic embryos that would germinate and produce plants. Media containing abscisic acid (80 μ M ) and osmotic agents such as sucrose, polyethylene glycol and/or dextran did not sustain development of mature somatic embryos from plated embryonal masses. This indicated that factors other than osmotic agents might be involved in sustaining development of Pinus strobus somatic embryos to maturity. It was subsequently found that media lacking osmotica but containing a high concentration of gellan gum (1%) induced significant improvement in the development of mature somatic embryos in the presence of 80 or 120 μ M abscisic acid. This positive effect was independent of the genotype and all four tested lines displayed similar responses. Media containing gellan gum at concentrations from 0.4 to 1.2% formed gels that varied in their strength. Gel strength was proportional to the concentration of gellan gum in the specific medium but varied depending on the medium formulation. Gel strength increased with the duration of storage of the culture medium by 46% (SD 14) after 14 days of storage. Preliminary results showed that embryos matured on high gellan gum media displayed improved germination frequencies. These results indicate that in Pinus strobus the water status and possibly other medium characteristics that are influenced by increased concentration of gelling agent have stimulatory effects on maturation of somatic embryos.  相似文献
