The Cerrado is the second largest Brazilian biome and contains the headwaters of three major hydrological basins in Brazil. In spite of the biological and ecological relevance of this biome, there is little information about how land use changes affect the chemistry of low-order streams in the Cerrado. To evaluate these effects streams that drain areas under natural, rural, and urban land cover were sampled near Brasília, Brazil. Water samples were collected between September 2004 and December 2006. Chemical concentrations generally followed the pattern of Urban > Rural > Natural. Median conductivity of stream water of 21.6 (interquartile: 22.7) ??S/cm in urban streams was three and five-fold greater relative to rural and natural areas, respectively. In the wet season, despite of increasing discharge, concentration of many solutes were higher, particularly in rural and natural streams. Streams also presented higher total dissolved N (TDN) loads from natural to rural and urban although DIN:DON ratios did not differ significantly. In natural and urban streams TDN was 80 and 77% dissolved organic N, respectively. These results indicate that alterations in land cover from natural to rural and urban are changing stream water chemistry in the Cerrado with increasing solute concentrations, in addition to increased TDN output in areas under urban cover, with potential effects on ecosystem function.  相似文献

The transfer of carbon (C) from Amazon forests to aquatic ecosystems as CO2 supersaturated in groundwater that outgases to the atmosphere after it reaches small streams has been postulated to be an important component of terrestrial ecosystem C budgets. We measured C losses as soil respiration and methane (CH4) flux, direct CO2 and CH4 fluxes from the stream surface and fluvial export of dissolved inorganic C (DIC), dissolved organic C (DOC), and particulate C over an annual hydrologic cycle from a 1,319-ha forested Amazon perennial first-order headwater watershed at Tanguro Ranch in the southern Amazon state of Mato Grosso. Stream pCO2 concentrations ranged from 6,491 to 14,976 ??atm and directly-measured stream CO2 outgassing flux was 5,994 ± 677 g C m?2 y?1 of stream surface. Stream pCH4 concentrations ranged from 291 to 438 ??atm and measured stream CH4 outgassing flux was 987 ± 221 g C m?2 y?1. Despite high flux rates from the stream surface, the small area of stream itself (970 m2, or 0.007% of watershed area) led to small directly-measured annual fluxes of CO2 (0.44 ± 0.05 g C m2 y?1) and CH4 (0.07 ± 0.02 g C m2 y?1) per unit watershed land area. Measured fluvial export of DIC (0.78 ± 0.04 g C m?2 y?1), DOC (0.16 ± 0.03 g C m?2 y?1) and coarse plus fine particulate C (0.001 ± 0.001 g C m?2 y?1) per unit watershed land area were also small. However, stream discharge accounted for only 12% of the modeled annual watershed water output because deep groundwater flows dominated total runoff from the watershed. When C in this bypassing groundwater was included, total watershed export was 10.83 g C m?2 y?1 as CO2 outgassing, 11.29 g C m?2 y?1 as fluvial DIC and 0.64 g C m?2 y?1 as fluvial DOC. Outgassing fluxes were somewhat lower than the 40?C50 g C m?2 y?1 reported from other Amazon watersheds and may result in part from lower annual rainfall at Tanguro. Total stream-associated gaseous C losses were two orders of magnitude less than soil respiration (696 ± 147 g C m?2 y?1), but total losses of C transported by water comprised up to about 20% of the ± 150 g C m?2 (±1.5 Mg C ha?1) that is exchanged annually across Amazon tropical forest canopies.  相似文献

以大田栽培的苎麻植株为材料,用TPS-2便携式光合作用测定系统测定自然条件下生长的苎麻叶片的光合气体交换参数,以及光响应曲线和CO2响应曲线,并通过回归和相关法分析探讨净光合速率与主要生理、生态因子间的关系.结果表明:(1)苎麻叶片的净光合速率(Pn)日变化曲线呈现双峰型,2个光合峰高度接近,其净光合速率具有典型的午休"现象;蒸腾速率(Tr)日变化曲线呈现单峰型,其走势与气孔导度(Gs)日变化一致.(2)苎麻叶片光合作用的光饱和点(LSP)为1 568.5μmol?m-2?s-1,光补偿点(LCP)为54.18μmol?m-2?s-1,表观量子效率(AQY)为0.025 8 mol?mol-1;而其CO2补偿点(CCP)、饱和点(CSP)和羧化效率(CE)分别为49.25、1 746.9μmol?mol-1和0.045;因此,苎麻属于喜光性阳生植物,且对强光有一定的耐受能力.(3)苎麻叶片Pn日变化的主要限制因子是胞间CO2浓度(Ci),主要决定生理因子是气孔导度(Gs).  相似文献

Amyloid formation is an ordered aggregation process, where β-sheet rich polymers are assembled from unstructured or partially folded monomers. We examined how two Escherichia coli cytosolic chaperones, DnaK and Hsp33, and a more recently characterized periplasmic chaperone, Spy, modulate the aggregation of a functional amyloid protein, CsgA. We found that DnaK, the Hsp70 homolog in E. coli, and Hsp33, a redox-regulated holdase, potently inhibited CsgA amyloidogenesis. The Hsp33 anti-amyloidogenesis activity was oxidation dependent, as oxidized Hsp33 was significantly more efficient than reduced Hsp33 at preventing CsgA aggregation. When soluble CsgA was seeded with preformed amyloid fibers, neither Hsp33 nor DnaK were able to efficiently prevent soluble CsgA from adopting the amyloid conformation. Moreover, both DnaK and Hsp33 increased the time that CsgA was reactive with the amyloid oligomer conformation-specific A11 antibody. Since CsgA must also pass through the periplasm during secretion, we assessed the ability of the periplasmic chaperone Spy to inhibit CsgA polymerization. Like DnaK and Hsp33, Spy also inhibited CsgA polymerization in vitro. Overexpression of Spy resulted in increased chaperone activity in periplasmic extracts and in reduced curli biogenesis in vivo. We propose that DnaK, Hsp33 and Spy exert their effects during the nucleation stages of CsgA fibrillation. Thus, both housekeeping and stress induced cytosolic and periplasmic chaperones may be involved in discouraging premature CsgA interactions during curli biogenesis.Key words: chaperone, curli, functional amyloid, CsgA, DnaK, Hsp33, Spy  相似文献

目的:克隆核心组蛋白H2A、H2B、H3和H4的基因,表达并纯化组蛋白与谷胱甘肽S-转移酶(GST)的融合蛋白。方法:用PCR方法从乳腺文库中扩增核心组蛋白H2A、H2B、H3和H4的编码序列,分别将其以正确相位与pGEX-KG载体中的GST编码序列融合,得到重组质粒pGST-H2A、pGST-H2B、pGST-H3和pGST-H4,分别转化大肠杆菌BL21,表达融合蛋白GST-H2A、GST-H2B、GST-H3和GST-H4;用谷胱甘肽-Sepharose 4B亲和纯化融合蛋白;用Western印迹检测融合蛋白的表达及纯化。结果:分别构建了核心组蛋白H2A、H2B、H3和H4的融合表达载体;Western印迹检测表明,融合蛋白GST-H2A、GST-H2B、GST-H3和GST-H4获得表达及纯化。结论:表达并纯化了H2A、H2B、H3和H4的融合蛋白,为进一步研究核心组蛋白的功能奠定了基础。  相似文献

Functional super-resolution (fSR) microscopy is based on the automated toponome imaging system (TIS). fSR-TIS provides insight into the myriad of different cellular functionalities by direct imaging of large subcellular protein networks in morphologically intact cells and tissues, referred to as the toponome. By cyclical fluorescence imaging of at least 100 molecular cell components, fSR-TIS overcomes the spectral limitations of fluorescence microscopy, which is the essential condition for the detection of protein network structures in situ/in vivo. The resulting data sets precisely discriminate between cell types, subcellular structures, cell states and diseases (fSR). With up to 16 bits per protein, the power of combinatorial molecular discrimination (PCMD) is at least 2(100) per subcellular data point. It provides the dimensionality necessary to uncover thousands of distinct protein clusters including their subcellular hierarchies controlling protein network topology and function in the one cell or tissue section. Here we review the technology and findings showing that functional protein networks of the cell surface in different cancers encompass the same hierarchical and spatial coding principle, but express cancer-specific toponome codes within that scheme (referred to as TIS codes). Findings suggest that TIS codes, extracted from large-scale toponome data, have the potential to be next-generation biomarkers because of their cell type and disease specificity. This is functionally substantiated by the observation that blocking toponome-specific lead proteins results in disassembly of molecular networks and loss of function.  相似文献

Anthropogenic N inputs and riverine export were determined for a meso-scale river basin in one of the most developed and economically important regions of South America. The Piracicaba River basin is located in southeastern Brazil and drains into a tributary of the Paraná River. The basin supports over 3 million people (about 2% of the population of Brazil) with intensive agricultural and industrial activities. During two years from 1995 to 1997, biweekly samples were collected at 10 stations along the Piracicaba River and its tributaries for analyses of dissolved and particulate N. The average annual flux of dissolved inorganic N and total N increased by a factor of 15 and 20 times, respectively, from the headwaters to the lower reaches of the main channel, whereas discharge increased by only 7 times. On a per area basis, the export of TN varied according to land use and was significantly correlated to the net input of anthropogenic N. Among 10 sub-catchments composing the basin, areas mostly covered by pasture and forest had the lowest export, whereas more agricultural and urban areas had higher export. The amount of N exported from each sub-catchment varied widely, but inputs were consistently higher than fluvial outputs. Losses and retention of N occurred throughout the basin but were especially high in the sub-catchment with a main-stem reservoir, suggesting that aquatic processing plays an important role in controlling riverine N export. Total net anthropogenic input to the Piracicaba River basin was 4,500 (± 900) kg N km–2 yr–1 of which about 40% was exported via fluvial outputs.  相似文献

A new species of Gesneriaceae from Honghe County, Southeastern Yunnan, China, Tremacron hongheense WH. Chen & YM. Shui, is described and illustrated. The new species is similar to Tremacron forrestii Craib, but differs by its leaf blade adaxially sparsely long setose (vs. densely white appressed pubescent and sparsely rusty brown villous), corolla tube outside short white glandular (vs. nearly glabrous), corolla lobes red and thickening at apex, especially adaxial lip (vs. yellow and not thickening), stamens 16-18cm long (vs. 04-12cm long), staminode 05-14cm long (vs. 02-04cm long).  相似文献
