Recently, we isolated from a raw vegetable processing line a Serratia strain with strong biofilm-forming capacity and which produced N-acyl-L-homoserine lactones (AHLs). Within the Enterobacteriaceae, strains of the genus Serratia are a frequent cause of human nosocomial infections; in addition, biofilm formation is often associated with persistent infections. In the current report, we describe the detailed characterization of the isolate using a variety of genotypic and phenotypic criteria. Although the strain was identified as Serratia plymuthica on the basis of its small subunit ribosomal RNA (16S rRNA) gene sequence, it differed from the S. plymuthica type strain in production of pigment and antibacterial compounds, and in AHL production profile. Nevertheless, the identification as S. plymuthica could be confirmed by gyrB phylogeny and DNA:DNA hybridization.  相似文献

The eukaryotic porin or voltage-dependent anion-selective channel (VDAC1) is a pore-forming protein discovered twenty five years ago in the mitochondrial outer membrane. Its gene in eukaryotes is known, but its tertiary structure has never been solved. Structure predictions highlight the presence of several amphipathic beta-strands possibly organised in a beta-barrel. VDAC1 has recently been described as being a NADH:ferricyanide reductase in the plasma membrane. There it affects the regulation of cell growth and death. Physiological cell death (apoptosis) has become a major research focus of biomedical research. Regulation of the enzyme will have impacts on cancer and autoimmune diseases (insufficient apoptosis) as well as neurodegenerative diseases (excessive apoptosis). VDAC1 in the plasma membrane establishes a novel level of apoptosis regulation putatively via its redox activity.  相似文献

In the yeast Saccharomyces cerevisiae, PKA and Sch9 exert similar physiological roles in response to nutrient availability. However, their functional redundancy complicates to distinguish properly the target genes for both kinases. In this article, we analysed different phenotypic read-outs. The data unequivocally showed that both kinases act through separate signalling cascades. In addition, genome-wide expression analysis under conditions and with strains in which either PKA and/or Sch9 signalling was specifically affected, demonstrated that both kinases synergistically or oppositely regulate given gene targets. Unlike PKA, which negatively regulates stress-responsive element (STRE)- and post-diauxic shift (PDS)-driven gene expression, Sch9 appears to exert additional positive control on the Rim15-effector Gis1 to regulate PDS-driven gene expression. The data presented are consistent with a cyclic AMP (cAMP)-gating phenomenon recognized in higher eukaryotes consisting of a main gatekeeper, the protein kinase PKA, switching on or off the activities and signals transmitted through primary pathways such as, in case of yeast, the Sch9-controlled signalling route. This mechanism allows fine-tuning various nutritional responses in yeast cells, allowing them to adapt metabolism and growth appropriately.  相似文献

To re-enact the long way to the origin of life with today's chemical methods, many steps have to be investigated in the light of a primordial scenario deduced from geochemical research. After the formation of our planet and its atmosphere, prebiotic chemical evolution started its course with the formation of the first building blocks for the formation of biomolecules. In the case of proteins, those building blocks were amino acids that had to be formed in the primitive atmosphere, and then had to react to peptides and proteins as the main pillar of first life. In this paper, we describe the processes in the primordial atmosphere according to contemporary geochemical knowledge leading to the synthesis of amino acids until the formation of homochiral peptides, and, thus, show a plausible pathway towards the origin of life.  相似文献

Abstract

The aim of the present study was to evaluate the efficacy of Elastoguard? silver-releasing rubber in preventing Pseudomonas aeruginosa biofilm formation in water. Biofilm formation by P. aeruginosa under various conditions in an in vitro model system was compared for silver-releasing and conventional rubber. Under most conditions tested, the numbers of sessile cells attached to silver-releasing rubber were considerably lower with reference to conventional rubber, although the effect diminished with increasing volumes. The release of silver also resulted in a decrease in planktonic cells. By exposing both materials simultaneously to conditions for biofilm growth, it became obvious that the antibiofilm effect was due to a reduction in the number of planktonic cells, rather than to contact-dependent killing of sessile cells. The data demonstrate that the use of silver-releasing rubber reduces P. aeruginosa biofilm in water and reduces the number of planktonic cells present in the surrounding solution.  相似文献

The number of studies dealing with tertiary ozonation to remove trace organic contaminants (TrOCs) in effluents originating from wastewater treatment plants (WWTPs) is increasing due to the need for upgrading the WWTPs overall performance. To follow-up TrOCs removal in real-time during ozone-based treatment, online surrogate measurements are necessary, of which mainly spectroscopic surrogates (i.e. UV–VIS absorbance and fluorescence) are the emerging techniques in literature. This paper summarizes and reflects on the state-of-the-art as retrieved from more than 100 peer-reviewed studies published between January 2007 and December 2020 and dealing with (1) surrogate correlation models for the prediction of TrOCs removal in secondary effluent and (2) control strategies to adjust the ozone dose during (full-scale) operation. Next to the flow and load proportional ozone dosing strategies, controlling the ozone dose solely based on the characteristics of the effluent entering the ozonation unit, also a differential control strategy based on the change in characteristics due to ozonation of the WWTP effluent is highlighted. The latter seems the best option as flow and load proportional ozone dosing do not consider the amount and/or reactivity of the matrix constituents. The presence of organic and inorganic scavengers of ozone and radicals in the effluent matrix has a significant impact on the TrOCs removal efficiency. This effluent quality can differ in time and between WWTPs, hence the surrogate correlation models should be widely applicable. At the end of the review, recommendations are made for future research and implementation of an effective control strategy for (full-scale) applications.

The purpose of this study was to determine the effect of load on lower extremity biomechanics during the pull phase of the clean. Kinematic and kinetic data of the 3 joints of the lower extremity were collected while participants performed multiple sets of cleans at 3 percentages: 65, 75, and 85% of 1 repetition maximum (1RM). General linear models with repeated measures were used to assess the influence of load on angular velocities, net torques, powers, and rates of torque development at the ankle, knee, and hip joint. The results suggest that the biomechanical demands required from the lower extremities change with the lifted load and to an extent depend on the respective joint. Most notably, the hip and knee extended significantly faster than the ankle independent of load, whereas the hip and ankle generally produced significantly higher torques than the knee did. Torque, rate of torque development (RTD), and power were maximimal at 85% of 1RM for the ankle joint and at 75% of 1RM for the knee joint. Torque and RTD at the hip were maximal at loads >75% of 1RM. This study provides important novel information about the mechanical demands of a weightlifting exercise and should be heeded in the design of resistance training programs.  相似文献
