Plant–herbivore dynamics is a major topic in ecological research, but empirical knowledge on the ecological effects of different densities of large grazers from fully replicated experiments is rare. Previous studies have focused on grazing vs no grazing, and our understanding of the extent to which different levels of grazing alter vegetation composition, and how quickly such effects can be measured, is therefore limited. We performed a fully replicated, short‐term (four‐year) experiment using large enclosures (each ~0.3 km2) with three different sheep densities (no grazing, low grazing and high grazing, respectively) in an alpine environment with summer grazing in southern Norway to address these issues. Sheep grazing mainly affected plant species at high densities of sheep as compared to no sheep after a four‐year treatment; few effects of low sheep densities were detectable. Highly selected herbs, herbs suggested vulnerable to trampling, and woody species decreased, while most graminoids, one ruderal, one prostrate species and two bryophyte taxa increased at high sheep densities. We found contrasting responses within main functional groups highlighting that fine details of plant life histories need to be known for responses to grazing to be successfully predicted. Vascular plant cover and bare soil responded to sheep density after two years of treatment, but only for one of the species was frequency change observed at this stage. Overall, plants in low grazing plots were found to be almost unaffected. Changes in abundance were mainly found at the no grazing and high grazing treatments. Plant species that decreased at high grazing generally increased at no grazing and vice versa, suggesting a response to both cessation of grazing and enhanced grazing respectively. Our study demonstrates, beyond a simple comparison of heavily grazed and non‐grazed sites, that herbivore effects on plants are typically non‐linearly related to herbivore density, and that the speed of plant responses will depend both on the plant property examined and the grazing pressure.  相似文献

We recorded the body axis orientation of free-living black wildebeest relative to incident solar radiation and wind. Observations were made on three consecutive days, on six occasions over the course of 1 year, in a treeless, predominantly cloudless habitat. Frequency of orientation parallel to incident solar radiation increased, and perpendicular to incident solar radiation decreased, as ambient dry-bulb temperature or solar radiation intensity increased, or wind speed decreased. We believe these changes were mediated via their effect on skin temperature. Parallel orientation behavior was more prominent when the wildebeest were standing without feeding than it was when they were feeding. We calculate that a black wildebeest adopting parallel orientation throughout the diurnal period would absorb 30% less radiant heat than the same animal adopting perpendicular orientation. Parallel orientation was reduced at times when water was freely available, possibly reflecting a shift from behavioral to autonomic thermoregulatory mechanisms. The use of orientation behavior by black wildebeest is well developed and forms part of the suite of adaptations that help them to maintain heat balance while living in a shadeless, often hot, environment.  相似文献

Stomata of barley (Hordeum vulgare L.) and wheat (Triticum aestivum L.) leaves failed to open in the light and close in the dark or respond to changes in the CO2 concentration of the atmosphere in either light or dark when the leaves were in an O2-free atmosphere. In contrast, the expected responses to environmental changes were found in atmospheres containing 1.5% O2. Phospholipase D (PLD) genes are members of a superfamily that is defined by several highly conserved motifs. PLD in mammals has been proposed to play a role in membrane vesicular trafficking and signal transduction. Using site-directed mutagenesis, 25 point mutants have been made in human PLD1 (hPLD1) and characterized. We find that a motif (HxKxxxxD) and a serine/threonine conserved in all members of the PLD superfamily are critical for PLD biochemical activity, suggesting a possible catalytic mechanism. Functional analysis of catalytically inactive point mutants for yeast PLD demonstrates that the meiotic phenotype ensuing from PLD deficiency in yeast derives from a loss of enzymatic activity. Finally, mutation of an HxKxxxxD motif found in a vaccinia viral protein expressed in the Golgi complex results in loss of efficient vaccinia virus cell-to-cell spreading, implicating the viral protein as a member of the superfamily and suggesting that it encodes a lipid modifying or binding activity. The results suggest that vaccinia virus and hPLD1 may act through analogous mechanisms to effect viral cellular egress and vesicular trafficking, respectively.  相似文献

Split-thickness skin grafts remain central to the strategy of burn wound treatment. The dressing used to cover the donor wound site has a significant effect on healing parameters. The purpose of this study was to compare split-thickness skin graft donor site reepithelialization under Xeroform and Jelonet dressings. A dermatome was used to cut two consecutive strips of skin from 25 paired donor sites on the thigh, calf, or back of 19 participants. Standardization of the harvest method was achieved by using the same surgeon to harvest the compared skin graft strips, with attention to consistency of dermatome skin-thickness setting, downward pressure, and angle of dermatome approach. A strip of Xeroform or Jelonet was applied to one of each pair of wounds. Epidermal and dermal thickness was measured from biopsy specimens cut at the midpoint of each split-thickness graft strip. The day of final dressing separation was declared the day of complete donor reepithelialization (healing). The mean healing time for Xeroform and Jelonet was 10.4 +/- 2.6 days (n = 25) and 10.6 +/- 2.8 days (n = 25) (p = 0.76) at sites cut to a mean depth of 0.23 +/- 0.08 mm and 0.23 +/- 0.09 mm (p = 0.89), respectively. There was no correlation between graft thickness and healing time for sites dressed with Xeroform (r = 0.17) or Jelonet (r = 0.02). Donors sites reharvested 10 to 21 days after a prior harvest healed an average of 3.1 days earlier than virgin sites (8.4 +/- 1.6 versus 11.5 +/- 2.6 days, p < 0.001), although reharvested grafts were on average 0.05 mm thicker (p = 0.10). The mean thickness of reepithelialized donor-site epidermis (0.13 +/- 0.04 mm, n = 30) was found to be twice the thickness of virgin epidermis from the same sites (0.06 +/- 0.02 mm, n = 38, p < 0.001). Thirty-six grafts harvested with dermatomes set to cut 8/1000 inch (0.20 mm) deep ranged from 0.12 to 0.42 mm thick, with only eight of these grafts measuring within +/-10 percent of the desired thickness setting. Before donor dressing separation, Xeroform and Jelonet dressings were judged to be more comfortable by nine patients and one patient, respectively, whereas no difference was detected by six patients. The authors now use Xeroform as the preferred donor dressing.  相似文献

The membrane fusion activities of the isolated single-envelope intracellular form of vaccinia virus (INV) and the double-envelope extracellular (EEV) form were studied by using a lipid-mixing assay based on the dilution of a fluorescent probe. Fluorescently labeled INV and EEV from both the IHD-J and WR strains of vaccinia virus fused with HeLa cells at neutral pH, suggesting that fusion occurs with the plasma membrane during virus entry. EEV fused more efficiently and with faster kinetics than INV: approximately 50% of bound EEV particles fused over the course of 1 h, compared with only 25% of the INV particles. Fusion of INV and EEV was strongly temperature dependent, being decreased by 50% at 34 degrees C and by 90% at 28 degrees C. A monoclonal antibody to a 14-kilodalton envelope protein of INV that has been implicated in the fusion reaction (J. F. Rodriguez, E. Paez, and M. Esteban, J. Virol. 61:395-404, 1987) completely suppressed the initial rate of fusion of INV but had no effect on the fusion activity of EEV, suggesting that vaccinia virus encodes two or more membrane fusion proteins. Finally, cells infected with the WR strain of vaccinia virus formed syncytia when briefly incubated at pH 6.4 or below, indicating that an acid-activated viral fusion protein is expressed on the cell surface. However, WR INV and EEV did not display increased fusion activity at acid pH, suggesting that the acid-dependent fusion factor is not incorporated into virions or that its activity there is masked.  相似文献

Investigations of cross-sectional geometry in nonhuman primate limb bones typically attribute shape ratios to qualitative behavioral characterizations, e.g., leaper, slow climber, brachiator, or terrestrial vs. arboreal quadruped. Quantitative positional behavioral data, however, have yet to be used in a rigorous evaluation of such shape-behavior connections. African apes represent an ideal population for such an investigation because their relatedness minimizes phylogenetic inertia, they exhibit diverse behavioral repertoires, and their locomotor behaviors are known from multiple studies. Cross-sectional data from femoral and humeral diaphyses were collected for 222 wild-shot specimens, encompassing Pan paniscus and all commonly recognized African ape subspecies. Digital representations of diaphyseal cross sections were acquired via computed tomography at three locations per diaphysis. Locomotor behaviors were pooled broadly into arboreal and terrestrial categories, then partitioned into quadrupedal walking, quadrumanous climbing, scrambling, and suspensory categories. Sex-specific taxonomic differences in ratios of principal moments of area (PMA) were statistically significant more often in the femoral diaphysis than the humeral diaphysis. While it appears difficult to relate a measure of shape (e.g., PMA ratio) to individual locomotor modes, general locomotor differences (e.g., percentage arboreal vs. terrestrial locomotion) are discerned more easily. As percentage of arboreal locomotion for a group increases, average cross sections appear more circular. Associations between PMA ratio and specific locomotor behaviors are less straightforward. Individual behaviors that integrate eccentric limb positions (e.g., arboreal scrambling) may not engender more circular cross sections than behaviors that incorporate repetitive sagittal movements (e.g., quadrupedal walking) in a straightforward manner.  相似文献
