With the objective to develop a practical method of screening potato for drought tolerance, shoot and root growth in plantlets
raised in vitro (from nodal cuttings drawn from in vivo as well as in vitro grown plantlets) were studied in three genotypes with known root mass production under field conditions. Different levels
of water stress were induced using five concentrations of agar in MS (Murashige and Skoog in Physiol Plant 15:473–497, 1962)
medium. Water potential of various media ranged from −0.70 MPa to −0.98 MPa. Water stress in culture adversely affected plantlet
growth, and the responses varied with genotype and explant source. Genotype IWA-1 was less affected than Konafubuki and Norin-1.
In the experiment with explants from in vivo grown plants, the time to rooting was considerably delayed in Konafubuki and Norin-1 by an increase in agar concentration,
but no such effect was observed in IWA-1. In all media, the mean number of roots and root length was greater in IWA-1 than
Konafubuki and Norin-1, and the latter two genotypes were at par. At 10 gl−1 agar, IWA-1 had taller plantlets, heavier foliage dry weight, root volume, as well as root dry weight than Konafubuki and
Norin-1, whereas the latter two genotypes were at par for all these characteristics. This pattern was similar to the reported
pattern of these genotypes for root dry weight under field conditions. However, such similarity in the in vitro and field behavior of the tested genotypes was not observed when nodal cuttings drawn from in vitro plantlets were used as explants. It is concluded that in vitro screening of potato under specific and limited water stress conditions by raising plantlets from nodal cuttings drawn from
in vivo grown plants may provide a system for effectively differentiating the genotypes for their expected root mass production under

In an effort to prepare Co(II)-substituted metallo-beta-lactamase L1 from Stenotrophomonas maltophilia for future spectroscopic and mechanistic studies, two methods for the preparation of Co(II)-L1 were tested. Method A involved adding CoCl2 directly to apo-L1 under anaerobic conditions. The resulting enzyme contained 1.9 moles of cobalt and exhibited very little activity, and UV-Vis, 1H NMR, and EPR studies indicated that most of the cobalt in this sample was Co(III). Method B involved reducing the single and unique disulfide bridge in L1 with tris(carboxyethyl)phosphine prior to adding CoCl2. The resulting enzyme was pink, contained 2.5 moles of cobalt per mole of enzyme, and exhibited kcat and Km values of 18+1 s(-1) and 10+/-1 microM, respectively, when using nitrocefin as the substrate. UV-Vis, 1H NMR, and EPR studies revealed that this enzyme sample contained high-spin Co(II). The UV-Vis spectra also provided evidence for Co(II) bound to one or both of the reduced cysteines. Efforts to block this non-specific Co(II) binding site using a chemical modification agent or site-directed mutagenesis were unsuccessful. The data presented here demonstrate the problem of solvent-exposed disulfides when preparing Co(II)-substituted enzymes and offers a convenient method to circumvent the problem. 相似文献

During a two-year investigation of freshwater microcrustaceans in Thailand, an undescribed species of fairy shrimp of the genus Branchinella Sayce, 1903 in the family Thamnocephalidae, was discovered. This is the first record of the genus and the family from Southeast Asia and the second anostracan reported from Thailand. The new species, Branchinella thailandensis n. sp. appears to be morphologically intermediate between B. kugenumaensis (Ishikawa, 1895) from Japan and east China, and B. madurai Raj, 1951 from India and Pakistan. B. thailandensis n. sp. was collected from several temporary ponds in 11 provinces of the northeast and the central Thailand, often co-occurring with Streptocephalus sirindhornae Sanoamuang, Murugan, Weekers & Dumont, 2000. 相似文献

Lactobacillus amylophilus GV6 fermented a variety of pure and natural starches directly to L(+) lactic acid. Starch to lactic acid conversion efficiency was more than 90% by strain GV6 at low substrate concentrations with all starches. The strain GV6 produced high yields of lactic acid per g of substrate utilized with pure starches such as soluble starch, corn starch, and potato starch, yielding 92–96% at low substrate concentrations in 2 days and 78–89% at high substrate (10%) concentrations in 4–6 days. Strain GV6 also produced high yields of lactic acid per g of substrate utilized with crude starchy substrates such as wheat flour, sorghum flour, cassava flour, rice flour and barley flour yielding 90–93% at low substrate concentrations in 2 days and 80% or more at high substrate concentrations in 6–7 days. Lactic acid yields by L. amylophilus GV6 with pure starches were comparable when low cost crude starchy substrates were used. The genus Isoetes L. in India is represented by 14species, of which eight species are recognized as being local endemics [confinedto one particular phyto-geographical division (PGD)] while the remaining sixoccur in more than one PGD and are described as endemic to a wider range. Thelocal endemic species are Isoetes dixitei,Isoetes panchganiensis and Isoetessahyadriensis in Western Ghats region; Isoetespantii, Isoetes bilaspurensis, Isoetesreticulata and Isoetes tuberculata inChotanagpur Malwa Vindhya Plateau and Isoetes debii innortheastern India. The wider endemic species are Isoetespanchananii, Isoetes sampathkumaranii,Isoetes rajasthanensis, Isoetesmahadevensis, Isoetes indica andIsoetes coromandelina. Our studies on the patterns ofendemism suggest that the radiation of quillworts advanced from dry lowlandareas to the rainy uplands and mountains. Isoetescoromandelina is the first Indian quillwort to colonize in lowlandsof the coastal zone (Coromandel), from where it spread to different parts of thesub-continent and gave rise to new species. Thus this species is a key Indianspecies which has played an important role in the radiation of quillwort in thecountry and appeared as the connecting link among the quillwort flora of variousPGDs. Lysine biosynthesis is crucial for cell-wall formation in bacteria. Enzymes involved in lysine biosynthesis are thus potential targets for anti-microbial therapeutics. Dihydrodipicolinate synthase (DHDPS) catalyzes the first step of this pathway. Unlike its homologues, Staphylococcus aureus DHDPS is a dimer both in solution and in the crystal and is not feedback inhibited by lysine. The crystal structure of S. aureus DHDPS in the free and substrate bound forms provides a structural rationale for its catalytic mechanism. The structure also reveals unique conformational features of the S. aureus enzyme that could be crucial for the design of specific non-competitive inhibitors. 相似文献

The present study was aimed to find out the protective effect of ethanolic extract of E. ribes fruits on homocysteine, lactate dehydrogenase (LDH) and lipid profile in serum, lipid peroxidation (LPO) and non-enzymatic antioxidant glutathione (GSH) levels in brain homogenates and histopathological examination of brain tissue in methionine (1 g/kg body weight, orally for 30 days) induced hyperhomocysteinemic rats. A significant increase in homocysteine, LDH, total cholesterol, triglycerides, low density lipoprotein (LDL-C) and very low density lipoprotein (VLDL-C) levels was observed in serum. Increased LPO levels in brain homogenates with reduced serum high density lipoprotein (HDL-C) levels and decreased GSH content were other salient features observed in methionine treated pathogenic control rats. Administration of ethanolic E. ribes extract (100 mg/kg body weight, orally) for 30 days to methionine-induced hyperhomocysteinemic rats produced a significant decrease in the levels of homocysteine, LDH, total cholesterol, triglycerides, LDL-C, VLDL-C in serum and LPO levels in brain homogenates with significant increase in serum HDL-C levels and GSH content in brain homogenates, when compared with pathogenic control rats. Biochemical observations were further substantiated with histological examination of brain. Degenerative changes of neuronal cells in methionine treated rats were minimized to near normal morphology by ethanolic E. ribes extract administration as evident by histopathological examination. The results provide clear evidence for the first time, that ethanolic E. ribes extract treatment enhances the antioxidant defense against methionine-induced hyperhomocysteinemia and oxidative stress in brain. 相似文献

The efficacy of Withania somnifera (Ws) to limit myocardial injury after ischemia and reperfusion was explored and compared to that of Vit E, a reference standard known to reduce mortality and infarct size due to myocardial infarction. Wistar rats (150–200 g) were divided into six groups and received orally saline (sham, control group), Ws-50/kg (Ws control and treated group) and Vit E-100 mg/kg (Vit E control and treated group) respectively for 1 month. On the 31st day, rats of the control, Vit E and Ws treated groups were anesthetized and subjected to 45 min occlusion of the LAD coronary artery followed by 60 min reperfusion. Hemodynamic parameters: systolic, diastolic and mean arterial pressure (SAP, DAP, MAP), heart rate (HR), left ventricular end diastolic pressure (LVEDP), left ventricular peak (+) LVdP/dt and (–) LVdP/dt were monitored. Hearts were removed and processed for histopathological and biochemical studies: Myocardial enzyme viz, creatin phosphokinase (CPK), and antioxidant parameters: malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSHPx) were estimated. Postischemic reperfusion produced significant cardiac necrosis, depression of left ventricular functions (MAP, LVEDP, (+) and (–) LVdP/dt) and a significant fall in GSH (p < 0.01), SOD, CAT(p < 0.05), LDH and CPK (p < 0.01) as well as an increase in MDA level (p < 0.05) in the control group rats as compared to sham group. The changes in levels of protein and GPx was however, not significant. Ws and Vit E favorably modulated most of the hemodynamic, biochemical and histopathological parameters though no significant restoration in GSH, MAP (with Vit E) were observed. Ws on chronic administration markedly augmented antioxidants (GSH, GSHPx, SOD, CAT) while Vit E did not stimulate the synthesis of endogenous antioxidants compared to sham. Results indicate that Ws significantly reduced myocardial injury and emphasize the beneficial action of Ws as a cardioprotective agent. 相似文献

A 2-year long study was carried out to isolate and characterise various bacterial species present in the biofilm formed on titanium surfaces exposed to seawater and to assess the manganese oxidizing potential of the marine isolates. The amount of manganese present in the biofilm was also measured using atomic absorption spectrometry (AAS). The results showed that titanium was susceptible to biofouling. More than 50% of the culturable marine bacterial isolates were capable of bringing about oxidation of Mn(II). All these manganese oxidizing bacteria were heterotrophic. Autotrophic manganese oxidizing bacteria such as Leptothrix was not isolated in the present study. The AAS results confirmed that the manganese content in the biofilms increased with increasing exposure time. Hence, the study indicates that the titanium surfaces when exposed to seawater were colonised by a large number of heterotrophic bacteria, which have the ability of bringing about biomineralisation of manganese. 相似文献

Three years of studies on some Indian Himalayan rivers in Jammu & Kashmir, Himachal Pradesh, and Sikkim revealed the presence
of Didymosphenia geminata (Lyngbye) M. Schmidt, a large diatom, hitherto unreported from these areas. This is first report on the occurrence of this
diatom in some Indian Himalayan rivers: Kishanganga in Jammu & Kashmir, Ravi in Himachal Pradesh in Western Himalaya and in
Teesta river and its two tributaries, Lachen Chhu and Lachung Chhu from Sikkim in the Eastern Himalaya. D. geminata is the most abundant taxon in both suspended and benthic algal communities in these rivers. The species exhibits wide ecological
amplitude, allowing it to inhabit both cold and warm waters. We also recorded the exotic brown trout, Salmo
trutta
fario in these Himalayan rivers. However, in Central Himalayan rivers, Alaknanda, Bhilangana, Mandakini and Dhauliganga in Uttarakhand,
where this diatom was absent the trout were absent too. Our studies indicate that D. geminata may extend its distribution range to other Himalayan rivers, hitherto uninhabited by the diatom, depending on the mode of
introduction of exotic trout in these rivers. 相似文献
