Spinach leaf phosphoribulokinase is sensitive to modification by ATP analogs that react with lysine residues. The 2',3'-dialdehyde derivative of ATP (oATP) inactivates enzyme in a slow, time-dependent fashion. The process follows first-order kinetics (kinact = 0.07 min-1), and the concentration dependence of inactivation indicates tight inhibitor binding (Ki = 106 microM). ATP offers good protection against inactivation (Kd = 67 microM), suggesting that oATP is directed toward the catalytic site. This conclusion is supported by the fact that oATP functions as an alternate substrate (Km = 0.55 mM). Inactivation of phosphoribulokinase by [14C]oATP results in a modification stoichiometry of 0.7/site. The 14C-labeled enzyme is stable to dialysis, suggesting that the covalent adduct formed between protein and oATP is not a simple Schiff's base. Adenosine di- and triphosphopyridoxals (Ado-P2-Pl and Ado-P3-Pl, respectively) also inhibit spinach phosphoribulokinase in a time-dependent fashion. In this case, activity loss is reversible unless the inhibited species is borohydride-reduced, suggesting that Ado-P2-Pl and Ado-P3-Pl form Schiff's bases with an amino group on the enzyme. Protection is afforded by the substrate ATP, suggesting that modification is active site-directed. Prolonged incubation of enzyme with these inhibitors does not result in complete inactivation of phosphoribulokinase. Residual activity is dependent on inhibitor concentration, as would be expected if equilibrium is established between the noncovalent E.I complex and the covalent (Schiff's base) E-I species. Kinetic data analysis indicates Ki values of 175 and 11 microM for Ado-P2-Pl and Ado-P3-Pl, respectively. Thus, the ATP-binding domain can easily accommodate the pyridoxal moiety which is tethered to the polyphosphate chain. The phosphorylated ATP analogs employed in this study exhibit substantially tighter binding to phosphoribulokinase than does fluorosulfonyl-benzoyladenosine (Ki = 4.8 mM), which we have previously demonstrated to be useful in selectively modifying the ATP-binding domain (Krieger, T. J., and Miziorko, H. M. (1986) Biochemistry 25, 3496-3501; Krieger, T. J., Mende-Mueller, L. M., and Miziorko, H. M. (1987) Biochim. Biophys. Acta 915, 112-119). Although the adduct formed between oATP and enzyme was unsuitable for structural analysis, borohydride reduction of the Schiff's base formed between enzyme and Ado-P3-[3H]Pl produced a species useful for investigation by protein chemistry techniques. A radiolabeled tryptic peptide was prepared, isolated, and sequenced; the data indicate that lysine 68 is the residue modified by Ado-P3-[3H]Pl. 相似文献

In normal human subjects under basal conditions, we have reported that molar concentrations of immunoreactive β-lipotropin (IR-β-LPH) are approximately threefold greater than those of IR-β-endorphin (β-Ep). Following acute stimulation, there is a further two- to threefold disproportionate rise in plasma concentrations of IR-β-LPH as compared to those of IR-β-Ep. To begin to assess the possible factors involved in such altered IR-β-LPH/IR-β-Ep ratios in plasma, the metabolic clearance rate (MCR), volume of distribution (Vd), fractional rate of disappearance (Kd), and half-life () of these peptides were determined by means of bolus injection of highly purified human β-LPH and synthetic human β-Ep in normal human subjects. β-Ep was found to have an MCR and a Kd greater than that of β-LPH, and a shorter . These differences, however, although they may in part be contributory, cannot solely account for the greater ratio of IR-β-LPH to IR-β-Ep in plasma, or for the disproportionate rise in plasma concentrations of these peptides after acute stimulation. 相似文献

The efficiency of strict reverse isolation and antimicrobial decontamination in remission induction therapy of acute leukemia was studied retrospectively in 47 patients who were treated with a standardized aggressive chemotherapy of daunorubicin and cytosine arabinoside. Twenty-two patients were treated in strict reverse isolation with antimicrobial decontamination and 25 patients in the open ward without any measures against infections. In the patients in isolation the incidence of new infections per patient was 0.77 compared to 1.42 in the control group. Previous studies have shown that the cholesteryl ester core of plasma low density lipoprotein (LDL) can be extracted with heptane and replaced with a variety of hydrophobic molecules. In the present report we use this reconstitution technique to incorporate two fluorescent probes, 3-pyrenemethyl-23, 24-dinor-5-cholen-22-oate-3β-yl oleate (PMCA oleate) and dioleyl fluorescein, into heptane-extracted LDL. Both fluorescent lipoprotein preparations were shown to be useful probes for visualizing the receptor-mediated endocytosis of LDL in cultured human fibroblasts. When normal fibroblasts were incubated at 37°C with either of the fluorescent LDL preparations, fluorescent granules accumulated in the perinuclear region of the cell. In contrast, fibroblasts from patients with the homozygous form of familial hypercholesterolemia (FH) that lack functional LDL receptors did not accumulate visible fluorescent granules when incubated with the fluorescent reconstituted LDL. A fluorescence-activated cell sorter was used to quantify the fluorescence intensity of individual cells that had been incubated with LDL reconstituted with dioleyl fluorescein. With this technique a population of normal fibroblasts could be distinguished from a population of FH fibroblasts. The current studies demonstrate the feasibility of using fluorescent reconstituted LDL in conjunction with the cell sorter to isolate mutant cells lacking functional LDL receptors. 相似文献

The genetic load disclosed by inbreeding has been analyzed in a multiple regression model for a population involving several localities in the state of Espírito Santo, Brazil. The inbreeding load has been estimated for number of pregnancies, abortions, stillbirths, children born alive, anomalies in general, sex ratio, infant mortality, post-infant mortality, and sterility and infertility of the couple. There was no evidence of either maternal or paternal inbreeding effects on the variables analyzed. The effect of inbreeding of the zygote was significant only for anomalies in general (B = 2.29 +/- 0.45) and infant mortality (B = 3.19 +/- 1.39). The latter result must be accepted with caution because of the many environmental causes affecting infant mortality. The B/A ratio suggested a predominantly mutational load for anomalies in general (B/A = 25), but with respect to infant mortality (B/A = 6), the ratio is regarded as an underestimate because of the environmental contribution to A and therefore not supportive of the segregational interpretation. 相似文献

The a priori identification of induced pluripotent stem cells remains a challenge. Being able to quickly identify the most embryonic stem cell-similar induced pluripotent stem cells when validating results could help to reduce costs and save time. In this context, tools based on non-classic logic can be useful in creating aid-systems based on visual criteria. True colonies when viewed at 100x magnification have been found to have the following 3 characteristics: a high degree of border delineation, a more uniform texture, and the absence of a cracked texture. These visual criteria were used for fuzzy logic modeling. We investigated the possibility of predicting the presence of alkaline phosphatase activity, typical of true induced pluripotent stem cell colonies, after 25 individuals, with varying degrees of experience in working with murine iPS cells, categorized the images of 136 colonies based on visual criteria. Intriguingly, the performance evaluation by area under the ROC curve (16 individuals with satisfactory performance), Spearman correlation (all statistically significant), and Cohen''s Kappa agreement analysis (all statistically significant) demonstrates that the discriminatory capacity of different evaluators are similar, even those who have never cultivated cells. Thus, we report on a new system to facilitate visual identification of murine- induced pluripotent stem cell colonies that can be useful for staff training and opens the possibility of exploring visual characteristics of induced pluripotent stem cell colonies with their functional peculiarities. The protozoan parasite Trypanosoma cruzi displays similarities to fungi in terms of its sterol lipid biosynthesis, as ergosterol and other 24-alkylated sterols are its principal endogenous sterols. The sterol pathway is thus a potential drug target for the treatment of Chagas disease. We describe here a comparative study of the growth inhibition, ultrastructural and physiological changes leading to the death of T. cruzi cells following treatment with the sterol biosynthesis inhibitors (SBIs) ketoconazole and lovastatin. We first calculated the drug concentration inhibiting epimastigote growth by 50% (EC50/72 h) or killing all cells within 24 hours (EC100/24 h). Incubation with inhibitors at the EC50/72 h resulted in interesting morphological changes: intense proliferation of the inner mitochondrial membrane, which was corroborated by flow cytometry and confocal microscopy of the parasites stained with rhodamine 123, and strong swelling of the reservosomes, which was confirmed by acridine orange staining. These changes to the mitochondria and reservosomes may reflect the involvement of these organelles in ergosterol biosynthesis or the progressive autophagic process culminating in cell lysis after 6 to 7 days of treatment with SBIs at the EC50/72 h. By contrast, treatment with SBIs at the EC100/24 h resulted in rapid cell death with a necrotic phenotype: time-dependent cytosolic calcium overload, mitochondrial depolarization and reservosome membrane permeabilization (RMP), culminating in cell lysis after a few hours of drug exposure. We provide the first demonstration that RMP constitutes the “point of no return” in the cell death cascade, and propose a model for the necrotic cell death of T. cruzi. Thus, SBIs trigger cell death by different mechanisms, depending on the dose used, in T. cruzi. These findings shed new light on ergosterol biosynthesis and the mechanisms of programmed cell death in this ancient protozoan parasite. 相似文献
