Oviductal tissue from fertile and infertile turkey breeder hens was stained immuno-histochemically to test for the presence of antibody positive cells. Relatively infertile turkey hens (< 50% fertility) were found to have antibody positive cells within the uterovaginal sperm storage tubule epithelium, while fertile turkey hens (> 90% fertility) had no antibody positive cells. Data suggest a local immune response to spermatozoa exists in the uterovaginal sperm storage tubules of the turkey hen which may have a detrimental effect on fertility. 相似文献

Pestiviruses, which belong to the Flaviviridae family of RNA viruses, are important agents of veterinary diseases causing substantial economical losses in animal farming worldwide. Pestivirus particles display three envelope glycoproteins at their surface: E(rns), E1, and E2. We report here the crystal structure of the catalytic domain of E(rns), the ribonucleolytic activity of which is believed to counteract the innate immunity of the host. The structure reveals a three-dimensional fold corresponding to T2 ribonucleases from plants and fungi. Cocrystallization experiments with mono- and oligonucleotides revealed the structural basis for substrate recognition at two binding sites previously identified for T2 RNases. A detailed analysis of poly-U cleavage products using (31)P-NMR and size exclusion chromatography, together with molecular docking studies, provides a comprehensive mechanistic picture of E(rns) activity on its substrates and reveals the presence of at least one additional nucleotide binding site. 相似文献

Nitric oxide (NO) is now recognized as an important signaling molecule and there has been an increasing bulk of studies regarding the various functions of NO in plants exposed to environmental stimulus. There is also emerging evidence, although not extensive, that NO plays systemic signaling roles during the establishment of salt tolerance in many plant species. In this mini-review, we highlight several candidate mechanisms as being functional in this NO systemic signaling action. This paper examines the hypothesis that testosterone (T) produces its differ-entiative effect on the neonatal rat brain after undergoing conversion in situ to estradiol-17β (E2). We examined the abilities of an aromatizing enzyme inhibitor, (1,4,6-androstatriene-3,17-dione) (ATD), and an anti-estrogen, CI628, to inhibit sexual differentiation. Male and female rats were treated during the first few days of postnatal life with ATD or CI628, and females were treated on the following day with T in Silastic capsules or its propionate (TP) in oil. ATD and ATD+T females were normal with respect to time of vaginal opening, ovarian weight, ability to demonstrate an LH surge, and lordosis behavior. T and TP females were masculinized with respect to all these measures. CI628 and CI628+TP females had impaired ovarian function and intermediate lordosis quotients (LQs) compared to controls, though they were higher in both measures than T females. ATD males demonstrated high LQs in response to estradiol benzoate (EB) + progesterone, comparable to those of control females. CI628 males had intermediate LQs, which were significantly higher than those of control males. These results indicate that ATD can substantially protect the neonatal rat brain from the differentiative effects of exogenous or endogenous T, probably by blocking aromatization. CI628 affords only partial protection against T and produces a weak differentiative effect due to its own weak estrogenicity. Thus, aromatization of T in newborn rat brains appears to be essential if sexual differentiation is to occur. 相似文献

TolR is a 15-kDa inner membrane protein subunit of the Tol-Pal complex in Gram-negative bacteria, and its function is poorly understood. Tol-Pal is recruited to cell division sites where it is involved in maintaining the integrity of the outer membrane. TolR is related to MotB, the peptidoglycan (PG)-binding stator protein from the flagellum, suggesting it might serve a similar role in Tol-Pal. The only structure thus far reported for TolR is of the periplasmic domain from Haemophilus influenzae in which N- and C-terminal residues had been deleted (TolR(62–133), Escherichia coli numbering). H. influenzae TolR(62–133) is a symmetrical dimer with a large deep cleft at the dimer interface. Here, we present the 1.7-Å crystal structure of the intact periplasmic domain of E. coli TolR (TolR(36–142)). E. coli TolR(36–142) is also dimeric, but the architecture of the dimer is radically different from that of TolR(62–133) due to the intertwining of its N and C termini. TolR monomers are rotated ∼180° relative to each other as a result of this strand swapping, obliterating the putative PG-binding groove seen in TolR(62–133). We found that removal of the strand-swapped regions (TolR(60–133)) exposes cryptic PG binding activity that is absent in the full-length domain. We conclude that to function as a stator in the Tol-Pal complex dimeric TolR must undergo large scale structural remodeling reminiscent of that proposed for MotB, where the N- and C-terminal sequences unfold in order for the protein to both reach and bind the PG layer ∼90 Å away from the inner membrane. 相似文献
