Biochemical evidence indicates that TGF-beta-activated kinase 1 (TAK1), a key modulator of the inflammatory response, exists in a complex with various adaptor proteins including the TAK1 binding protein 1 (TAB1). However, the physiological importance of TAB1 in TAK1 activation, and in the subsequent induction of proinflammatory mediators, remains unclear. In this study, a critical role for TAK1 in IL-1alpha or TNFalpha stimulated MAPK and NFkappaB activation was confirmed by inhibition of the nuclear accumulation of NFkappaB p65 and phosphorylated forms of c-Jun and p38 following siRNA mediated TAK1 silencing. These effects were associated with significant reductions in IL-1alpha stimulated levels of secreted IL-6, IL-8, MCP-1 and GM-CSF. In contrast, IL-1alpha or TNFalpha dependent cellular redistribution of NFkappaB p65 and phosphorylated c-Jun and p38 was not affected by 80% siRNA mediated knockdown of TAB1 protein levels. Interestingly, IL-6, IL-8 and GM-CSF release from TAB1 siRNA transfected cells was significantly reduced following IL-1alpha treatment, but was unchanged after TNFalpha stimulation, suggesting differential roles for TAB1 in IL-1alpha and TNFalpha signalling pathways. These findings may imply an as yet unidentified role for TAB1 in the inflammatory response, which is independent of the activation of classical TAK1 associated signalling cascades. 相似文献

Surveillance of wild vertebrates can be challenging in remote and inaccessible areas such as tropical rainforests. Blood-feeding parasites, such as leeches, can facilitate wild vertebrate monitoring by targeting residual DNA from the animals the leeches feed on. Successes in detecting host DNA from leeches suggest that host viruses may also be detectable. To systematically test this hypothesis, we performed a proof of concept study using quantitative PCR (qPCR) to detect DNA viruses (bovine herpesvirus [BHV], human adenovirus [HAdV]) and RNA viruses (influenza A [InfA] and measles morbillivirus [MeV]) from nucleic acids extracted from medicinal leeches fed with blood spiked with each virus. All viruses except BHV showed a gradual decline in concentration from day 1 to 50, and all except BHV were detectable in at least half of the samples even after 50 days. BHV exhibited a rapid decline at day 27 and was undetectable at day 50. Our findings in medicinal leeches indicate that leeches collected in the wild might be an untapped resource for detecting vertebrate viruses and could provide new opportunities to study wildlife viral diseases of rare species in challenging environments, where capturing and handling of animals is difficult. 相似文献

In the genome of Artiodactyla (cow, sheep, pigs, camels, and whales), a major retroposon group originated from a presumable horizontal transfer of BovB, a retrotransposon-like element retroposon, between 52 and 70 million years ago. Since then, BovB retroposons have proliferated and today occupy a quarter of the cow's genome sequence. The BovB-related short interspersed elements (SINEs) were used for resolving the phylogeny of Bovinae (cows, spiral-horned antelopes, and nilgais) and their relatives. In silico screening of 55,000 intronic retroposon insertions in the cow genome and experimental validation of 126 introns resulted in 29 informative retroposon markers for resolving bovine evolutionary relationships. A transposition-in-transposition analysis identifies three different phases of SINE activity and show how BovB elements have expanded in the cattle genome. 相似文献

This article analyses the large-scale popular urban uprisings that shook Mozambican cities on 1 and 2 September 2010, following the government's announcement of successive rises in the price of public transport fares and basic commodities. Using ethnographic material from the city of Chimoio and the capital Maputo, the following work highlights the organisational character of the ‘strikes’ (greves), as the popular uprisings were called, and explores them as a new form of organising political discontent. Comparing them to other historical and contemporary popular uprisings, this article argues that the strikes violently and rhizomically generated ephemeral and egalitarian forms of political authority and order that simultaneously confronted, replicated and undercut the aspects of Mozambican statehood. Deploying Durkheim's notion of effervescence, the work further argues that the creative fervour, multisemic aspects and festive character of the popular uprisings need to be recognised; thus, this analysis challenges the reductive labelling of these events as ‘riots’ or ‘food riots’. 相似文献

Geoducks (Panopea abrupta) as isotopic bioarchives of seasonality in the climate of British Columbia
Geochemical analyses of geoduck shells (Panopea abrupta) have yielded new high-resolution isotope records of individual growth increments. The geoducks, which were collected from Parry Passage at Langara Island, British Columbia, are the most northern specimens that have yet been analyzed for stable oxygen and carbon isotopes. The isotope records represent the entire shell growth during the calendar years 1918, 1987, and 1991. The analyses showed that values of δ18Oshell and δ13Cshell vary within their growth increments. Temperature derived from δ18Oshell is consistent with modern instrumental data of sea-surface temperature. High-resolution δ18O records from individual growth increments substantiate the assumption that they are formed annually. The instrumental data indicate that the growth season was from May through October/November. The sea temperature became gradually warmer during the beginning of the growth season, and decreased steeply during autumn. Similar subannual trends can be recognized in shells growing almost a century ago. Correlation between δ18Oshell and δ13Cshell is negative for 1917–1919 and 1986–1988, whereas the correlation is positive for 1990–1992. Differences in correlation may be related to the composition of available food particles, i.e., algal blooms and plant debris. The food composition is partially affected by the Pacific Decadal Oscillation (PDO). These findings have major implications for the understanding of the interpretation of carbon isotopes in marine bivalves. Because of their spatial and temporal distribution, the geoduck shells are highly suitable for oceanographic reconstructions using stable-isotope analysis and sclerochronology. Furthermore, the large shell size and thickness and their deeply buried life position suggest a high preservation potential in ancient deposits. In a mass balance of 15N-labelled nitrate added to soil grown with pea or barley, denitrification estimates using the acetylene-inhibition technique
were compared with unaccounted for 15N. During the growth season of 1989, which was drier than average, N losses due to denitrification estimated by the acetylene-inhibition
technique were negligible. A substantial amount of fertilizer N was unaccounted for by the 15N mass balance, especially in the pea plots. The loss took place during the period of grain-filling in which no leaching occurred,
and was accompanied by a decrease in 15N content of the plants. Volatilization of ammonia from the aerial parts of the plants is a possible explanation of the observed

Late onset Alzheimer’s disease (LOAD) is a genetically complex and clinically heterogeneous disease. Recent large-scale genome wide association studies (GWAS) have identified more than twenty loci that modify risk for AD. Despite the identification of these loci, little progress has been made in identifying the functional variants that explain the association with AD risk. Thus, we sought to determine whether the novel LOAD GWAS single nucleotide polymorphisms (SNPs) alter expression of LOAD GWAS genes and whether expression of these genes is altered in AD brains. The majority of LOAD GWAS SNPs occur in gene dense regions under large linkage disequilibrium (LD) blocks, making it unclear which gene(s) are modified by the SNP. Thus, we tested for brain expression quantitative trait loci (eQTLs) between LOAD GWAS SNPs and SNPs in high LD with the LOAD GWAS SNPs in all of the genes within the GWAS loci. We found a significant eQTL between rs1476679 and PILRB and GATS, which occurs within the ZCWPW1 locus. PILRB and GATS expression levels, within the ZCWPW1 locus, were also associated with AD status. Rs7120548 was associated with MTCH2 expression, which occurs within the CELF1 locus. Additionally, expression of several genes within the CELF1 locus, including MTCH2, were highly correlated with one another and were associated with AD status. We further demonstrate that PILRB, as well as other genes within the GWAS loci, are most highly expressed in microglia. These findings together with the function of PILRB as a DAP12 receptor supports the critical role of microglia and neuroinflammation in AD risk. 相似文献

The milk protein β-lactoglobulin (βLG) dominates the properties of whey aggregates in food products. Here we use spectroscopic and calorimetric techniques to elucidate how anionic, cationic and non-ionic surfactants interact with bovine βLG and modulate its heat-induced aggregation. Alkyl trimethyl ammonium chlorides (xTAC) strongly promote aggregation, while sodium alkyl sulfates (SxS) and alkyl maltopyranosides (xM) reduce aggregation. Sodium dodecyl sulfate (SDS) binds to non-aggregated βLG in several steps, but reduction of aggregation was associated with the first binding step, which occurs far below the critical micelle concentration. In contrast, micellar concentrations of xMs are required to reduce aggregation. The ranking order for reduction of aggregation (normalized to their tendency to self-associate) was C10-C12>C8>C14 for SxS and C8>C10>C12>C14>C16 for xM. xTAC promote aggregation in the same ranking order as xM reduce it. We conclude that SxS reduce aggregation by stabilizing the protein's ligand-bound state (the melting temperature t(m) increases by up to 10°C) and altering its charge potential. xM monomers also stabilize the protein's ligand-bound state (increasing t(m) up to 6°C) but in the absence of charged head groups this is not sufficient by itself to prevent aggregation. Although micelles of both anionic and non-ionic surfactants destabilize βLG, they also solubilize unfolded protein monomers, leaving them unavailable for protein-protein association and thus inhibiting aggregation. Cationic surfactants promote aggregation by a combination of destabilization and charge neutralization. The food compatible surfactant sodium dodecanoate also inhibited aggregation well below the cmc, suggesting that surfactants may be a practical way to modulate whey protein properties. 相似文献
