The optimal growth of mesophilic methanotrophic bacteria (collection strains of the genera Methylocystis, Methylomonas, Methylosinus, and Methylobacter) occurred within temperature ranges of 31-34 degrees C and 23-25 degrees C. None of the strains studied were able to grow at 1.5 or 4 degrees C. Representatives of six methanotrophic species (strains Mcs. echinoides 2, Mm. methanica 12, Mb. bovis 89, Mcs. pyriformis 14, Mb. chroococcum 90, and Mb. vinelandii 87) could grow at 10 degrees C (with a low specific growth rate). The results obtained suggest that some mesophilic methane-oxidizing bacteria display psychrotolerant (psychrotrophic) but not psychrophilic properties. In general, the Rosso model, which describes bacterial growth rate as a function of temperature, fits well the experimental data, although, for most methanotrophs, with symmetrical approximations for optimal temperature.  相似文献

A method for isolation of rat hepatocytes using liver perfusion by ethylenediamine tetra-acetic acid (EDTA)-containing sucrose solution and mechanical tissue disaggregation by controlled vibration (MVD) is described. The yields of hepatocytes produced by this method were similar to those obtained using collagenase perfusion. The cells had well preserved membrane integrity as judged by the trypan blue staining test (91 +/- 4%), ATP contents, rates of endogenous respiration and enzyme leakage that indicated they were functional cells. There was little evidence of expression of latent damage when the cells were stored either at 37 degrees C (by pre-incubation) or at 4 degrees C. This method can be used to isolate high yields of functional cells from rat liver if the collagenase perfusion technique is not available.  相似文献

Homogenate respiratory activity was studied after different storage terms of the whole rat liver at 4 degrees C in sucrose-based solution and following normothermic reperfusion. Preservation of homogenate respiratory activity in all metabolic states after normothermic reperfusion of the control liver (60 min, 4 degrees C) is shown. Further storage (6 and 24 hrs) of isolated liver under the mentioned above conditions strengthens the substrate respiration of homogenate both after storage and after normothermic reperfusion. At the same time oxidative phosphorylation does not practically change. No change was noted in respiratory activity in the states 3, 4ADP and 3DNP after 24 hrs of liver storage in respect of a previous term. Following normothermic liver reperfusion contributes to a statistically true reduction of mentioned parameters of respiration, that correlates with a decrease in the degree of respiration and phosphorylation coupled of the studied system.  相似文献

Using of isolated hepatocytes for investigation of the effects of hypothermia, it has been demonstrated that sucrose-base solution provides of maintenance of the energetic parameters (level of ATP, glucose synthesis, rate of gluconeogenesis) within 48 hrs of storage at 4 degrees C. It efficiency was compared with effect on the energetic status of isolated hepatocytes widely used preservation solution--solution of University Wisconsin (UW). After long-term of cold storage of isolated hepatocytes (72 hrs) at 4 degrees C in both solutions, it has been shown sharp decrease of ATP level (on two time). Viability of the liver cells (in both cases) was practically without change.  相似文献

The results of the experimental study of the environment modification--the emergence of Liesegang rings around a Dictyostelium discoideum population are presented. The formation of Liesegang rings induced by D. discoideum cells is observed on addition of glucose into the semi-solid nutrient medium (agar concentration 0.5-1.5%). We show that the emergence of Liesegang rings is attended with a redistribution of folic acid in the nutrient substrate. A pH decrease in the course of D. discoideum cultivation is shown to be a factor inducing the redistribution of folic acid. The mechanisms of structural modification of the D. discoideum environment are discussed.  相似文献

The transformation of the spatial structure of a Dictyostelium discoideum population in response to environmental changes induced by this population was investigated. A comparative analysis of the spatial and temporal characteristics of the D. discoideum colony is given for two cases: (a) when the colony is cultivated on a bacterial lawn, i.e. under conditions close to natural, and (b) in the absence of the bacterial lawn when the colony grows on the nutrient substrate enriched with folic acid. It is shown that the environmental changes induced by cell metabolism modify the spatial structure of the D. discoideum population first, the rate of population propagation falls drastically, which correlates with a decrease in the substrate pH; second, the spatial redistribution of the D. discoideum cell density correlates with the redistribution of folic acid in the substrate. The mechanism of the environment impact on the D. discoideum colony transformation is discussed.  相似文献

A theoretical study of the phenomenon of Liesegang structure formation induced by the Dictyostelium discoideum population in a medium containing folic acid was carried out. Using a "reaction-diffusion" model proposed in this work, it was shown that the formation of Liesegang structures around the Dictyostelium discoideum population depends on two competing processes: (a) inactivation of folic acid by vegetative amoebae and (b) the chemical reaction of folic acid with the products of amoeba metabolism, which results in the formation of insoluble sediment. The dependence of the model solutions on the geometric and functional parameters was studied. The results are in good agreement with experimental data.  相似文献

The necessity and effectiveness of introducing the program of the exchange of syringes for intravenous drug users as a preventive and antiepidemic measure are discussed. Preventive and antiepidemic measures within the concept of harm reduction are described. The comparative analysis of materials obtained as a result of two sociological questionings, carried out in 1997-1998 among the representatives of the community (random nonrepeating choice) and in the middle of 1999 among the participants of the program revealed that the participants of the program had statistically significant changes in risky behavior patterns during the use of injection drugs.  相似文献
