Disturbances act as powerful structuring forces on ecosystems. To ask whether environmental microbial communities have capacity to recover after a large disturbance event, we conducted a whole-ecosystem manipulation, during which we imposed an intense disturbance on freshwater microbial communities by artificially mixing a temperate lake during peak summer thermal stratification. We employed environmental sensors and water chemistry analyses to evaluate the physical and chemical responses of the lake, and bar-coded 16S ribosomal RNA gene pyrosequencing and automated ribosomal intergenic spacer analysis (ARISA) to assess the bacterial community responses. The artificial mixing increased mean lake temperature from 14 to 20 °C for seven weeks after mixing ended, and exposed the microorganisms to very different environmental conditions, including increased hypolimnion oxygen and increased epilimnion carbon dioxide concentrations. Though overall ecosystem conditions remained altered (with hypolimnion temperatures elevated from 6 to 20 °C), bacterial communities returned to their pre-manipulation state as some environmental conditions, such as oxygen concentration, recovered. Recovery to pre-disturbance community composition and diversity was observed within 7 (epilimnion) and 11 (hypolimnion) days after mixing. Our results suggest that some microbial communities have capacity to recover after a major disturbance.  相似文献

Thalidomide is emerging as a therapeutic agent with renewed clinical importance, presenting anti-inflammatory, immunomodulatory, and antineoplasic properties. In this work, we studied the complexation of thalidomide with cyclodextrins as a strategy to circumvent the poor aqueous solubility of the drug. Thalidomide–hydroxypropyl-β-cyclodextrin complexes were obtained by kneading method and were characterized by differential scanning calorimetry, powder X-ray diffractometry, and scanning electronic microscopy. The aqueous solubility and in vitro dissolution of thalidomide were significantly improved through the complexation. Physicochemical analysis of the complexes in solid state revealed a decreased crystallinity of the complexed drug in comparison with free thalidomide. Thalidomide was able to dissociate from the complexes and permeates across intestinal epithelial Caco-2 cells with a favorable high permeability profile equivalent to that of the free drug. In summary, the present results suggest that thalidomide–hydroxypropyl-β-cyclodextrin complexes could be regarded as a promising strategy for improving the gastrointestinal absorption of thalidomide.  相似文献

We evaluated several factors influencing the taxonomic richness of macrophytes, benthic invertebrates, snails, and fish in a series of northern Wisconsin lakes. We chose the study lakes to decouple the potential effects of ionic strength of lake water and stream connection, two factors that are usually highly correlated and therefore have been confounded in previous studies. In addition, our study lakes covered a wide range in a variety of characteristics, including residential development, abundance of exotic species, nutrient concentrations, predator abundance, and lake size. Species richness within each of the four taxonomic groups was significantly positively related to ionic strength (as measured by specific conductance); we also found secondary associations with other variables, depending on the specific group of organisms. The relationship between richness and lake area was dependent on the specific conductance of the lake and the vagility of the organisms; less vagile groups of organisms showed stronger and steeper species–area relationships in low-conductivity lakes. Further, after variance owing to specific conductance was removed, the presence of stream connections was positively related to species richness for fish, snails, and macrophytes as well as familial richness in benthic invertebrates. Our results indicate that lakes with relatively more groundwater input have lower extinction rates for all four groups of taxa and that lakes with stream inlets and outlets have enhanced immigration rates for fish, snails, benthic invertebrate families, and macrophytes. These findings link processes of immigration and extinction of four groups of organisms of varying vagility to landscape-level hydrologic characteristics related to the glacial history of the region.  相似文献

An important distinction is frequently made between constitutively expressed housekeeping genes versus regulated genes. Although generally characterized by different DNA elements, chromatin architecture and cofactors, it is not known to what degree promoter classes strictly follow regulatability rules and which molecular mechanisms dictate such differences. We show that SAGA‐dominated/TATA‐box promoters are more responsive to changes in the amount of activator, even compared to TFIID/TATA‐like promoters that depend on the same activator Hsf1. Regulatability is therefore an inherent property of promoter class. Further analyses show that SAGA/TATA‐box promoters are more dynamic because TATA‐binding protein recruitment through SAGA is susceptible to removal by Mot1. In addition, the nucleosome configuration upon activator depletion shifts on SAGA/TATA‐box promoters and seems less amenable to preinitiation complex formation. The results explain the fundamental difference between housekeeping and regulatable genes, revealing an additional facet of combinatorial control: an activator can elicit a different response dependent on core promoter class.  相似文献

Chemotactic migration of bacteria—their ability to direct multicellular motion along chemical gradients—is central to processes in agriculture, the environment, and medicine. However, current understanding of migration is based on studies performed in bulk liquid, despite the fact that many bacteria inhabit tight porous media such as soils, sediments, and biological gels. Here, we directly visualize the chemotactic migration of Escherichia coli populations in well-defined 3D porous media in the absence of any other imposed external forcing (e.g., flow). We find that pore-scale confinement is a strong regulator of migration. Strikingly, cells use a different primary mechanism to direct their motion in confinement than in bulk liquid. Furthermore, confinement markedly alters the dynamics and morphology of the migrating population—features that can be described by a continuum model, but only when standard motility parameters are substantially altered from their bulk liquid values to reflect the influence of pore-scale confinement. Our work thus provides a framework to predict and control the migration of bacteria, and active matter in general, in complex environments.  相似文献

The occurrence, intensity, and composition of mutualisms are dependent not only on the co-occurrence of mutualists, but also the broader biotic context in which they are embedded. Here, the influence of the specific nest tree identity of the ant Azteca instabilis (F. Smith) on the density of the green coffee scale (Coccus viridis Green) was studied in a coffee agroecosystem in southern Mexico. The hypothesis that an indirect competitive interaction for ant attendance occurs between a scale species (Octolecanium sp. Kondo) in the canopy of the shade tree Inga micheliana Harms and C. viridis, which inhabits coffee bushes (Coffea arabica) beneath the shade trees was tested. Coffee bushes beneath a different shade tree species (Alchornea latifolia Swartz) were used as an indication of C. viridis density in a noncompetitive environment. Results indicate that C. viridis occurs in significantly lower density adjacent to nests in Inga, supporting the hypothesis of indirect competition. Additional experimentation suggests that there is a mutualism between Azteca and Octolecanium and that this interaction may be mediated by a hierarchy in ant attendance of scale insects. Our results show the importance of considering the biotic context of ant-hemipteran mutualisms. In coffee agroecosystems, consideration of shade tree diversity and species composition may be directly applicable to the biological control of insect pests.  相似文献

Severe and often therapy-limiting side effects are a major obstacle in cancer chemotherapy. New delivery concepts reducing systemic side effects are needed in order to optimize anticancer therapies. Several approaches have been followed, most of them concentrating on macromolecular carriers like liposomes, monoclonal antibodies, serum proteins or polyethylene glycol. We present here a novel type of anthracycline conjugate, using a small carrier peptide derived from the peptide hormone human calcitonin (hCT). The carrier peptide hCT(9-32) has so far been shown to be capable of transporting fluorophores or proteins across cellular membranes. Two different carrier peptide-daunorubicin conjugates were prepared, one with an acid-stable amide bond, the second with an acid-labile hydrazone bond. In vitro studies with daunorubicin linked to the carrier peptide via an acid-labile hydrazone bond demonstrated comparable cytotoxicity to daunorubicin in various daunorubicin sensitive cell lines (neuroblastoma cell lines SK-N-MC and SMS-KAN; HEK 293 T cells). In addition, fluorescence microscopy provided further insight into the mechanism of uptake of the carrier peptide hCT(9-32), indicating that endosomal compartments with reduced pH are involved in the intracellular release of daunorubicin.  相似文献

Background  

The integrity of DNA molecules is fundamental for maintaining life. The DNA repair proteins protect organisms against genetic damage, by removal of DNA lesions or helping to tolerate them. DNA repair genes are best known from the gamma-proteobacterium Escherichia coli, which is the most understood bacterial model. However, genome sequencing raises questions regarding uniformity and ubiquity of these DNA repair genes and pathways, reinforcing the need for identifying genes and proteins, which may respond to DNA damage in other bacteria.  相似文献
