A new ceramiaceous alga, Sciurothamnion stegengae De Clerck et Kraft, gen. et sp. nov., is described from the western Indian Ocean and the Philippines. Sciurothamnion appears related to the tribe Callithamnieae on the basis of the position and composition of its procarps and by the majority of post‐fertilization events. It differs, however, from all current members of the tribe by the presence of two periaxial cells bearing determinate laterals per axial cell. Additionally, unlike any present representative of the subfamily Callithamnioideae, no intercalary foot cell is formed after diploidization of the paired auxiliary cells. The genus is characterized by a terminal foot cell (“disposal cell”), which segregates the haploid nuclei of the diploidized auxiliary cell from the diploid zygote nucleus. The nature of three types of foot cells reported in the Ceramiaceae (intercalary foot cells containing only haploid nuclei, intercalary foot cells containing haploid nuclei and a diploid nucleus, and terminal foot cells containing only haploid nuclei) is discussed.  相似文献

T-cell subsets were studied by fluorescence-activated cell sorter analysis in 57 feline immunodeficiency virus (FIV)-seropositive cats with naturally acquired FIV infection to see whether CD4(+)-CD8+ alterations were comparable to those observed in human immunodeficiency virus-infected patients. CD4+ values were decreased and CD8+ values were increased. The CD4+/CD8+ ratio was reduced to 1.6, compared with 3.3 in 33 FIV-seronegative control cats. Variance analysis of data showed a significant influence of FIV seropositivity, sex, and spaying of female cats on CD4+ values. CD8+ values were significantly influenced by FIV seropositivity, age, and breed. These findings indicate a similarity between FIV and human immunodeficiency virus infections, as far as alterations of T-cell subsets are concerned.  相似文献

Several southern Australian red algae of the family Halymeniaceae (Cryptonemiales) are differentiated into hard, massive stalks and considerably softer laminar blades or phyllodes. The taxonomy, morphology and pit-connection ultrastructure of one such species, Cryptonemia kallymenioides (Harvey) Kraft comb. nov., are compared to C. undulata Sonder, which lacks massive stalks. In both species there is extensive periodic secondary cortication of the stalks, resulting in the formation of distinct “growth rings.” The blades of C. kallymenioides appear to be seasonal and its stalks perennial, while plants of C. undulata are apparently perennial but shorter lived than C. kallymenioides. As a result, stalks in the latter can reach 2–3 cm in diameter with up to 18 growth rings, compared to the 1–2 mm diameters and up to 6 rings within the stalks of C. undulata. Heavy secondary thickening of cortical cell walls occurs in both species and confers a “woody” texture to the stalks of C. kallymeniodes. Regardless of the large differences in average stalk diameters between the two species, the pit-connection ultrastructure from cortex to medulla shows much the same sequence of morphological modification. Pit-connections are standard red algal structures in the outer cortex, but become increasingly convoluted on the membrane-bound surfaces abutting cytoplasm and develop wider apertures and less dense cores with increasing distance from the stalk surface. In occasional medullary cells of C. kallymenioides, the cytoplasm disintegrates, leaving cell walls and pit-connections to play an apparently structural role which has not been reported in other red algae. It is suggested that the increase in aperture size and surface areas of pit-connections is compatible with their playing a role in the intercellular transport of solutes towards the inner cell layers which may, in C. kallymenioides, lie many millimeters distant.  相似文献

Summary Trirhabda bacharidis (Weber) (Chrysomelidae), a univoltine, monophagous beetle is the dominant herbivore on Baccharis halimifolia (Compositae), a woody, perennial shrub that leafs out in early spring and retains its leaves into November. Available plant biomass increases during the season but T. bacharidis feeds only during spring and early summer. During the remainder of the growing season, there are no major herbivores feeding on B. halimifolia. B. halimifolia leaves increase in toughness and thickness and decrease in moisture and nutrients as the season progresses. In feeding preference tests, T. bacharidis larvae preferred young leaves over leaves of older age classes. When reared on leaves of different ages, larvae fed on young leaves, weighed more, pupated earlier and had greater survivorship. T. bacharidis larvae showed no significant feeding preference for similarly tough B. halimifolia leaves painted with differing concentrations of late season acetone leaf extract. Elm leaves painted with the same leaf extracts were avoided by larvae of the non-adapted specialist Pyrrhalta luteola (Mueller) (Chrysomelidae).These results suggest that the adapted specialist, T. bacharidis, is not deterred by the B. halimifolia acetone soluble secondary chemical which increases in amount over the season. Decreasing leaf nitrogen (perhaps in concert with increasing leaf toughness) seems to be the primary factor that dissuades its feeding. However, acetone soluble secondary chemicals in the leaves of B. halimifolia may be effective in preventing herbivory from non-adapted insects.  相似文献

 AFLP markers were evaluated for their usefulness in the genetic analysis of sugarbeet and wild Beta species. Accessions of ten different sugarbeet breeding lines and five wild beets were screened using 256 primer combinations. Of the 11 309 bands investigated, 96.4% were polymorphic among the accessions. A strong positive correlation was found between the number of polymorphisms and AT content of the selective bases of the primer combinations. Random subsets of primer combinations were used to produce genetic distance trees. Permutation tests showed that, for the wild beets, 500 AFLP bands sufficed to obtain the best topology of the tree with a probability at any given node of more than 99%. Ten times as many bands were necessary to obtain support values of the same order of magnitude for the sugarbeet lines. The reproducibility of AFLP for seven primer combinations was investigated by repeated analysis of all steps from DNA isolation to data scoring. For 5088 comparisons, the overall reproducibility was 97.6%. Robustness to genotyping errors was investigated by including an artificial F1 (1 : 1 DNA mixture) of two sugarbeet lines in the screen for polymorphisms. For the 3160 cases of polymorphism between the two lines, 0.2% genotyping errors were found. The general reliability and usefulness of AFLP markers are discussed in relation to the results obtained. Penicillin-binding protein 2a (PBP2a) is the primary beta-lactam resistance determinant of methicillin-resistant Staphylococcus aureus (MRSA). MecA, the gene coding for PBP2a, was cloned with the membrane-anchoring region at the N-terminus deleted. The truncated protein (PBP2a) was overexpressed in Escherichia coli mostly in the soluble form accounting for approximately 25% of soluble cell protein and was purified to homogeneity. The purified protein was shown to covalently bind beta-lactams in an 1:1 ratio as determined by electrospray mass spectrometry. A novel method based on HPLC-elctrospray mass spectrometry has been developed to quantitatively determine the formation of the covalent adducts or acyl-PBP2a complexes. By using this method, combined with kinetic techniques including quench flow, we have extensively characterized the interactions between PBP2a and three beta-lactams and determined related kinetic parameters for the first time. The apparent first-order rate constants (ka) of PBP2a acylation by benzylpenicillin showed a hyperbolic dependence on the concentration of benzylpenicillin. This is consistent with the mechanism that the binding of the penicillin to PBP2a consists of reversible formation of a Michaelis complex followed by formation of the penicilloyl-PBP2a adduct, and allowed the determination of the individual kinetic parameters for these two steps, the dissociation constant Kd of 13.3 mM and the first-order rate constant k2 of 0.22 s-1. From these values, the second-order rate constant k2/Kd, the value reflecting the overall binding efficiency of a beta-lactam, of 16.5 M-1 s-1 was obtained. The fairly high Kd value indicates that benzylpenicillin fits rather poorly into the protein active site. Similar studies on the interaction between PBP2a and methicillin revealed k2 of 0.0083 s-1 and Kd of 16.9 mM, resulting in an even smaller k2/Kd value of 0.49 M-1 s-1. The rate constants k3 for deacylation of the acyl-PBP2a complexes, the third step in the interactions, were measured to be <1.5 x 10(-)5 s-1. These results indicate that the resistance of PBP2a to penicillin inactivation is mainly due to the extremely low penicillin acylating rate in addition to the low association affinity, but not to a fast rate of deacylation. Acylation of PBP2a by a high-affinity cephalosporin, Compound 1, also followed a saturation curve of ka versus the compound concentration, from which k2 = 0.39 s-1, Kd = 0.22 mM, and k2/Kd = 1750 M-1 s-1 were obtained. The 100-fold increase in the k2/Kd value as compared with that of benzylpenicillin is mostly attributable to the decreased (60-fold) Kd, indicating that the cephalosporin fits much better to the binding pocket of the protein.  相似文献

BackgroundBoth genetic and lifestyle factors contribute to the risk of type 2 diabetes, but the extent to which there is a synergistic effect of the 2 factors is unclear. The aim of this study was to examine the joint associations of genetic risk and diet quality with incident type 2 diabetes.Methods and findingsWe analyzed data from 35,759 men and women in the United States participating in the Nurses’ Health Study (NHS) I (1986 to 2016) and II (1991 to 2017) and the Health Professionals Follow-up Study (HPFS; 1986 to 2016) with available genetic data and who did not have diabetes, cardiovascular disease, or cancer at baseline. Genetic risk was characterized using both a global polygenic score capturing overall genetic risk and pathway-specific polygenic scores denoting distinct pathophysiological mechanisms. Diet quality was assessed using the Alternate Healthy Eating Index (AHEI). Cox models were used to calculate hazard ratios (HRs) for type 2 diabetes after adjusting for potential confounders. With over 902,386 person-years of follow-up, 4,433 participants were diagnosed with type 2 diabetes. The relative risk of type 2 diabetes was 1.29 (95% confidence interval [CI] 1.25, 1.32; P < 0.001) per standard deviation (SD) increase in global polygenic score and 1.13 (1.09, 1.17; P < 0.001) per 10-unit decrease in AHEI. Irrespective of genetic risk, low diet quality, as compared to high diet quality, was associated with approximately 30% increased risk of type 2 diabetes (Pinteraction = 0.69). The joint association of low diet quality and increased genetic risk was similar to the sum of the risk associated with each factor alone (Pinteraction = 0.30). Limitations of this study include the self-report of diet information and possible bias resulting from inclusion of highly educated participants with available genetic data.ConclusionsThese data provide evidence for the independent associations of genetic risk and diet quality with incident type 2 diabetes and suggest that a healthy diet is associated with lower diabetes risk across all levels of genetic risk.
