Chemokines are an important group of soluble molecules with specialized functions in inflammation. The roles of many specialized chemokines and their receptors remain poorly understood in the human intervertebral disc. We investigated CXCL16 and its receptor, CXCR6, to determine their immunolocalization in disc tissue and their presence following exposure of cultured human annulus fibrosus cells to proinflammatory cytokines. CXCL16 is a marker for inflammation; it also can induce hypoxia-inducible factor 1α (HIF-1α), which is a phenotypic marker of heathy nucleus pulposus tissue. We found CXCL16 and CXCR6 immunostaining in many cells of the annulus portion of the disc. Molecular studies showed that annulus fibrosus cells exposed to IL-1ß, but not TNF-α, exhibited significant up-regulation of CXCL16 expression vs. control cells. There was no significant difference in the percentage of annulus cells that exhibited immunolocalization of CXCL16 in grade I/II, grade III or grade IV/V specimens. The presence of CXCL16 and its receptor, CXCR6, in the annulus in vivo suggests the need for future research concerning the role of this chemokine in proinflammatory functions, HIF-1α expression and disc vascularization.  相似文献

Woodlands worldwide have been greatly modified by clearing for agriculture, and their conservation and restoration requires understanding of tree recruitment processes. Seed production is one possible point of recruitment failure, and one that the spatial arrangement of trees may affect. We sampled 118 Eucalyptus microcarpa (Myrtaceae) trees to compare and analyse the determinants of seed production in this dominant tree of modified, fragmented temperate grassy woodlands, which extend over much of southeastern Australia. Fecundity was estimated as the seed crop measured on leaf mass and whole tree bases and was compared between categories of tree configuration. We also modelled fecundity using boosted regression trees, a new and flexible tool. Fecundity on a leaf mass basis was predominantly influenced by environmental factors (topographic ‘wetness’, slope, soil type), rather than by local tree density and configuration. Fewer seed per unit leaf mass were produced on flat and topographically wet sites, reflecting poor tolerance of waterlogging by E. microcarpa. By contrast, whole tree fecundity was little influenced by environmental factors. Local tree density and configuration did influence whole tree fecundity, which was high in solitary and woodland‐spaced trees and reduced under high local density. We found little evidence for reduced fecundity of E. microcarpa in solitary trees. This points to the importance of scattered trees as sources of seed for tree recruitment and for natural regeneration of landscape level tree cover. Considerable uncertainty remains in modelled seed supply, and may be reduced with sampling across multiple years and greater environmental and spatial domains.  相似文献

SUMMARY. A ciliate found in the renal organ of the gray garden slug, Deroceras reticulatum , is referred to Tetrahymena (Paraglaucoma) rostrata (Kahl). In the parasitic phase, the ciliate is evidently not histiophagic. It is readily cultivated in bacterized tissue infusions, and has been established in axenic culture in yeast extract.
The number of complete primary ciliary meridians in ciliates of the parasitic phase has been observed to range from 41 to 58; in ciliates from cultures, the number of meridians has been observed to range from 28 to 37. In culture, the form of T. rostrata from D. reticulatum appears to be morphologically identical with a strain (NZ-4) recovered from soil in New Zealand.  相似文献

The nature of the interaction of bacteriophage T4D and the outer cell wall of its host, Escherichia coli B, has been investigated. Bacteria with altered or modified cell walls have been obtained by two different growth procedures: (i) growth in high osmolarity medium or (ii) growth in broth in the presence of divalent heavy metal ions. When these altered host cells were washed and subsequently added to regular growth medium, they interacted with added phage particles, but successful infection did not occur. Most of the phage particles released from these treated cells were observed to have full heads and an altered tail structure. The altered phage tails had contracted sheaths and unusual pieces of the bacterial cell wall attached to the distal portion of the exposed phage tail tube. Phage released from bacteria grown in the high osmolarity medium had attached cell wall pieces of two major types, these pieces being either 40 or 21 nm in diameter. The smaller-type cell wall pieces (21 nm) were formed by three spheres each measuring 7 nm in diameter. Phage particles released from cells previously exposed to the divalent metal ions had only one 7-nm cell wall sphere attached to the distal end of the tail tube. It was found that these 7-nm spheres (i) are normal components of the cell wall and are morphologically similar to endotoxin, (ii) are held in place on the cell wall by a component of the cell wall called protein b, and (iii) are most likely the site of penetration of the phage tail tube through which the phage DNA enters the host cell.  相似文献

Our objective for this study was to determine the presence and distribution of tenascin in the human intervertebral disc. The tenascins are a family of extracellular matrix proteins with repeated structural domains homologous to epidermal growth factor, fibronectin type III and the fibrinogens. Little is known about the presence of this protein in the disc. Ten normal human discs donated from subjects newborn to 15 years old, 10 control discs from adult donors aged 24-41 years, and 11 surgical disc specimens from patients aged 26-76 years were examined for immunolocalization of tenascin. In young discs, tenascin was localized throughout the annulus; in the nucleus, localization was confined to pericellular matrix. In adult control and degenerating disc specimens, tenascin in the annulus was localized primarily in pericellular matrix regions encircling either single cells or clusters of disc cells; in rare instances localization was more diffuse in the intraterritorial matrix. In young, healthy disc, tenascin was abundant throughout the annulus. In contrast, degenerating discs in adults showed a localization restricted to the pericellular, and rarely, more restricted intraterritorial matrix. These observations indicate that changes in the amount and distribution of tenascin may have a role in disc aging and degeneration, possibly by modulating fibronectin-disc-cell interactions, and causing alterations in the shape of disc cells.  相似文献

The folate compound which is a structural component of the Escherichia coli T-even bacteriophage baseplates, has been identified as the hexaglutamyl form of folic acid using a new chromatographic procedure (Baugh, C.M., Braverman, E. and Nair, M.G. (1974) Biochemistry 13, 4952-4957). It has also been found that the host cell contains a variety of polyglutamyl forms of folic acid. The major form is the triglutamate (about 50%) but small amounts of higher molecular weight folates including the octaglutamate (1.8%) have been identified. Upon infection with wild-type T4D bacteriophage there is a shift in the distribution of the folate compounds so that the folyl polyglutamyl compounds having the higher molecular weights are increased. Infection of E. coli with baseplate mutants of T4D containing an amber mutation in gene 28 resulted in the formation of significant amounts (over 7%) of folate compound(s) of molecular weight much higher than those observed either in uninfected cells or cells infected with wild-type T4D. It is suggested that the T4D gene 28 product functions to cleave glutamate residues from high molecular weight folyl polyglutamates to increase the availability of the folyl hexaglutamate for virus assembly.  相似文献

An assay for folylpolyglutamate synthetase activity in extracts of uninfected and bacteriophage T4D-infected Escherichia coli B has been developed. T4D infection induced the formation of a new synthetase raising the total synthetase activity three-fold. Extracts obtained after infection with T4 gene 51, 27 or 28 amber mutants showed increased synthetase activities while extracts obtained from cells infected with a T4D gene 29 amber mutant did not show any increase in synthetase activity. The phage-induced synthetase was found to copurify with the gene 29 product and a 100-fold purified synthetase of molecular size of 74,000 daltons has been obtained. The purified synthetase has a folate substrate specificity different from the host synthetase since it added glutamate residues to dihydrofolate as well as to the usual tetrahydrofolate substrate.  相似文献

Wahlia pulchella n. sp. is described from the intestine of the aspidochirote holothuroid Stichopus californicus. The new species differs from other members of the genus in having the following combination of features: the pharynx lies at the end of the first quarter or third of the body, the intestine extends anterior to the brain, the testes are only slightly lobed, and the penis stylet is straight and less than half the length of the adult worm. Wahlia pulchella has a "secondary uterus" in which there may be up to 12 egg capsules. The validity of the genera of holothuroid-inhabiting umagillids characterized by 2 ovaries and a secondary uterus is discussed. It is suggested that Ozametra Marcus, 1949, be placed in synonymy under Wahlia Westblad, 1930. New diagnoses of Wahlia and Seritia Cannon, 1982, are presented.  相似文献

A pteroylpolyglutamate has been found to be a constituent of all Escherichia coli T-even bacteriophages and has been characterized with regard to its oxidation state, molecular weight, origin, and location on the phage particle. The phage compound has been shown to be a dihydropteroyl penta- or hexaglutamate on the basis of its chemical and physical properties. Analyses of extracts of uninfected and T2L-infected E. coli have indicated that the phage dihydropteroyl polyglutamate was present only in infected cells. Its synthesis was sensitive to the addition of chloramphenicol before infection, and the compound appeared to be specifically induced by phage infection. Analyses of isolated phage ghosts and tail substructures have shown that each phage particle contains between two and six phage-specific pteroyl derivatives and that the juncture of the phage tail plate with the tail tube is the most likely site of binding of the phage-induced pteroyl compound.  相似文献
