Growing hyphae of Gelasinospora reticulispora required a continuousdark period prior to photoinduction of perithecia. The inductivedark period was interrupted by brief exposure of the hyphaeto white light so that the formation of perithecia no longertook place. Photosensitivity of the hyphae in terms of the light-breakeffect gradually changed during the inductive dark period. Sensitivityreached its maximum at the 18th hr of the dark period when anirradiation of 1?105 ergs cm–2 of near-UV light or 4?104ergs cm–2 of blue-light was sufficient for the light-break.Red and far-red light had no effect at all. The light-breakeffect was limited to the irradiated portion of the hyphae anddid not affect any unirradiated portions. Inhibitory effecton perithecial formation of continuous white light could betotally replaced for several days with intermittent irradiationof near-UV or blue light if given for 5 min every 4 hr. Hyphae of Gelasinospora reticulispora growing on corn meal agarat 25?C required light of form perithecia. This response tolight was highly correlated to the length of the preliminarydark period, i.e. the photoinduction of perithecia never occurredin cultures grown in the dark for periods of 27 hr or less,whereas hyphae became photosensitive if incubated for 30 hror longer in the dark. The perithecia became simultaneouslyvisible with the 30 to 48 hr dark-grown hyphae irrespectiveof the different dark periods, but time courses for cultureshaving 54 hr or longer of darkness were dependent upon the timewhen light was given. Light induced perithecia only in the dark-grown portions ofthe hyphae. The longer the dark period, the more the sensitivityto light increased when the most effective range of wavelenghtswas shifted from near-ultraviolet to blue. Light of green andlonger wavelenghts was not at all effective irrespective ofthe duration of darkness. The photoinduced stimulus was notmovable from the irradiated to the unirradiated portions. The reductive carboxylation of α-ketoglutarate by purified NADP+-isocitrate dehydrogenase (EC 1.1.1.42) from maturing castor bean seeds ( Ricinus communis L. ) has been characterized. The optimum pH for the reaction was 6.5, whereas pH 8.5 was optimum for oxidation of isocitrate (forward reaction). The enzyme utilized NADH as well as NADPH as the reducing agent in the reverse reaction, but only NADP+ in the forward reaction. The Km values for NADPH and NADH were 0.044 and 2.8 m M respectively, and for α-ketoglutarate and HCO3 4.1 and 3.7 m M. The enzyme was activated by various cations including Mg2+, Mn2+, Co2+, Zn2+, Ni2+ and Co2+. Km values for Mg2+ Mn2+, Co2+ and Zn2+ were 12, 34, 37 and 49μ M respectively.  相似文献

The level of butanol-soluble cytokinin in potato tubers (Solanumtuberosum L.) decreased greatly after harvest of the tubers,remained low during rest-period and then began to increase priorto the end of the period. In warm-stored tubers (25?C) the levelrose faster than in cold-stored ones (4?C). On the other hand,the level of water-soluble cytokinin, which was high duringthe rest-period, decreased after the rest was broken. Wounding the tubers caused an increase in butanol-soluble cytokinin.The increase in cytokinin within the first 24 hr after woundingwas smaller in warm-stored tubers than in cold-stored ones andwas decreased with increasing storage time of the tubers. Thelevel of water-soluble cytokinin decreased by wounding. Theseresults suggest that water-soluble cytokinin is a storage formand that the level of butanol-soluble cytokinin is regulatedby an interconversion between the two. By incorporation of 3H2O into the fatty acid chain in the presenceof unlabeled precursor, we showed that fatty acids are synthesizedfrom PGA, PEP and pyruvate by intact spinach chloroplasts inthe light. 13C-tracer experiments confirmed that 1-C of pyruvateis decarboxylated and 2-C is incorporated into fatty acids bythe chloroplasts. The patterns of fatty acids synthesized fromPGA and pyruvate were the same as that from acetate. The highestrate of fatty acid synthesis was reached at the physiologicalconcentration of PGA (3 mM) and pyruvate (1 mM). These resultsindicate the operation of the following path in the chloroplastsin light: PGA-PEPPyruvateacetylCoAfatty acids. Since citrateand OAA were much less active and malate and glyoxylate wereinert as precursors for fatty acid synthesis, PEP or pyruvatecarboxylation, citrate lyase reaction and malate synthetasereaction are not involved in the formation of acetylCoA andfatty acids. Since pyruvate was much more effective as a substratefor fatty acid synthesis than lactate, acetaldehyde or acetate,direct, decarboxylation path is considered to be the primarypath from pyruvate to acetylCoA. The insignificant effect ofchloroplast-washing on fatty acid synthesis from PGA and pyruvateindicates that the glycolytic path from PGA to pyruvate is associatedwith the chloroplasts. Since pyruvate was more effectively incorporatedinto fatty acids than acetylCoA, it is unlikely that pyruvatedecarboxylation to acetylCoA is due to mitochondria contaminatingthe chloroplast preparation. On the basis of measurements of 3H2O incorporation in the lightand dark, the activity of fatty acid synthesis in spinach leavesappears to be shared by the activities in chloroplasts (87%)and other organelles (13%). 1 Present address: Radioisotope Centre, University of Tokyo,Yayoi, Bunkyo 113, Japan. Feeding selectivities of Daphnia galeata were measured using various density combinations of 14C-labeled Chlamydomonas and 3H-labeled natural bacteria. In all food combinations, D. galeata preferred algae. Selectivity coefficient (bacterial clearance rate: algal clearance rate) was 0.35 at the level of incipient food limitation (1.5 × 106 µm3 ml–1).However, it increased up to 0.9 with increasing total food concentration. A simple mechanical sieving model alone cannot explain such a change in the selectivity coefficient. A mechanism of clogging suggestively explain the increase of bacterial ingestion efficiency by D. galeata, especially at high food concentrations. The change of selectivity coefficient related to food condition suggests that bacterial feeding by the Daphnia is incidental event associated with the algal feeding.  相似文献

Lysophosphatidic acid (LPA) is a bioactive lipid growth factor which is present in high levels in serum and platelets. LPA binds to its specific G-protein-coupled receptors, including LPA1 to LPA6, thereby regulating various physiological functions, including cancer growth, angiogenesis, and lymphangiogenesis. Our previous study showed that LPA promotes the expression of the lymphangiogenic factor vascular endothelial growth factor (VEGF)-C in prostate cancer (PCa) cells. Interestingly, LPA has been shown to regulate the expression of calreticulin (CRT), a multifunctional chaperone protein, but the roles of CRT in PCa progression remain unclear. Here we investigated the involvement of CRT in LPA-mediated VEGF-C expression and lymphangiogenesis in PCa. Knockdown of CRT significantly reduced LPA-induced VEGF-C expression in PC-3 cells. Moreover, LPA promoted CRT expression through LPA receptors LPA1 and LPA3, reactive oxygen species (ROS) production, and phosphorylation of eukaryotic translation initiation factor 2α (eIF2α). Tumor-xenografted mouse experiments further showed that CRT knockdown suppressed tumor growth and lymphangiogenesis. Notably, clinical evidence indicated that the LPA-producing enzyme autotaxin (ATX) is related to CRT and that CRT level is highly associated with lymphatic vessel density and VEGF-C expression. Interestingly, the pharmacological antagonist of LPA receptors significantly reduced the lymphatic vessel density in tumor and lymph node metastasis in tumor-bearing nude mice. Together, our results demonstrated that CRT is critical in PCa progression through the mediation of LPA-induced VEGF-C expression, implying that targeting the LPA signaling axis is a potential therapeutic strategy for PCa.  相似文献

The egg structure and outline of the embryonic development of Metallyticus splendidus of one of the basal Mantodea representatives, Metallyticidae, were described in the present study. The results obtained were compared with those from the previous studies, to reconstruct and discuss the groundplan of Mantodea and Dictyoptera. In M. splendidus, the egg is spheroidal, it has a convex ventral side at the center in which numerous micropyles are grouped, and it possesses a conspicuous hatching line in its anterior half. These are the groundplan features of mantodean eggs and the “grouped micropyles in the ventral side of the egg” are regarded as an apomorphic groundplan feature of Dictyoptera. A small circular embryo is formed by a simple concentration of blastoderm cells, which then undergoes embryogenesis of the typical short germ band type. Blastokinesis is of the “non-reversion type” and the embryo keeps its original superficial position and original orientation throughout embryonic development. During the middle stages of development, the embryo undergoes rotation around the egg's anteroposterior axis. These features are a part of the groundplan of Mantodea. It is uncertain whether sharing of the “non-reversion type” of blastokinesis by Mantodea and blaberoidean Blattodea can be regarded as homology or homoplasy.  相似文献

Extending healthy lifespan is an emerging issue in an aging society. This study was designed to identify a dietary method of extending lifespan, promoting renoprotection, and preventing muscle weakness in aged mice, with a focus on the importance of the balance between dietary essential (EAAs) and nonessential amino acids (NEAAs) on the dietary restriction (DR)‐induced antiaging effect. Groups of aged mice were fed ad libitum, a simple DR, or a DR with recovering NEAAs or EAAs. Simple DR significantly extended lifespan and ameliorated age‐related kidney injury; however, the beneficial effects of DR were canceled by recovering dietary EAA but not NEAA. Simple DR prevented the age‐dependent decrease in slow‐twitch muscle fiber function but reduced absolute fast‐twitch muscle fiber function. DR‐induced fast‐twitch muscle fiber dysfunction was improved by recovering either dietary NEAAs or EAAs. In the ad libitum‐fed and the DR plus EAA groups, the renal content of methionine, an EAA, was significantly higher, accompanied by lower renal production of hydrogen sulfide (H2S), an endogenous antioxidant. Finally, removal of methionine from the dietary EAA supplement diminished the adverse effects of dietary EAA on lifespan and kidney injury in the diet‐restricted aged mice, which were accompanied by a recovery in H2S production capacity and lower oxidative stress. These data imply that a dietary approach could combat kidney aging and prolong lifespan, while preventing muscle weakness, and suggest that renal methionine metabolism and the trans‐sulfuration pathway could be therapeutic targets for preventing kidney aging and subsequently promoting healthy aging.  相似文献

In maturing endosperms of a variety of sugary mutants of rice, phytoglycogen-like polysaccharides with highly branched a -glucans were accumulated instead of amylopectin. while the amylose content greatly decreased. Measurement of activities per endosperm of the 10 major enzymes involved in starch and sucrose metabolism revealed that the activity of starch debranching enzyme (R-enzyme) was specifically reduced in the sugary mutants. The activity of starch branching enzyme I (Q-enzyme I) was also significantly decreased, but less so than the R-enzyme, in the mutants, suggesting some coordination of the expression of the genes coding for R-enzyme and Q-enzyme I. Western blot analysis showed that the sugary mutations of rice resulted in a decrease in the amount of R-enzyme protein, but not in major modification of the enzyme. These findings strongly suggest that R-enzyme plays a critical role in determining the amylopectin fine structure, since at the extremely low level of R-enzyme activity as compared with Q-enzyme activity, as found in sugary mutants, the rice endosperm produced phytoglycogen. We hypothesize that balance of activities or interaction between Q-enzyme and R-enzyme may be responsible for the fine structure of a -polyglucans in plant tissues.  相似文献
