Although the sodium-calcium exchanger (NCX1) is encoded by a single gene, it is widely expressed in both fetal and adult tissues and functions in many diverse physiological processes to maintain intracellular calcium homeostasis. In order to determine whether NCX1 is also ubiquitously expressed in the early mouse embryo, in situ hybridization and RT-PCR were used to determine the spacio-temporal expression of NCX1. Our results indicate that NCX1 expression is present within the 7.75-8.0 dpc cardiogenic plate before the first heartbeat, and that NCX1 is initially expressed in a heart-restricted pattern within the early mouse embryo. However, in more developed embryos (11.0 dpc and older) NCX1 is expressed in other tissues. 相似文献
Lipid transfer proteins (LTPs) are the key contributor of organelle-specific lipid distribution and cellular lipid homeostasis. Here, we report a novel implication of LTPs in phagocytosis, trogocytosis, pinocytosis, biosynthetic secretion, recycling of pinosomes, and motility of the parasitic protist E. histolytica, the etiological agent of human amoebiasis. We show that two StAR-related lipid transfer (START) domain-containing LTPs (named as EhLTP1 and 3) are involved in these biological pathways in an LTP-specific manner. Our findings provide novel implications of LTPs, which are relevant to the elucidation of pathophysiology of the diseases caused by parasitic protists. 相似文献
Presence of a tumor and its characteristics like location, size and properties are estimated. Estimation is based on the measurement of the skin surface temperature of the breast. Consideration is given to a 2-D breast tissue infected with a tumor. Heat transfer in the breast tissue modeled using the Pennes bioheat equation is solved by the finite volume method. Skin surface temperature profile of the breast is characteristic of the tumor location, its size and grade. In the inverse analysis, the objective function is minimized using the genetic algorithm. Exact estimation is obtained if one parameter is estimated at a time. However, the accuracies are acceptable even when blood perfusion rate, location and size are estimated simultaneously. 相似文献
A molecular ruler, FliK, controls the length of the flagellar hook. FliK measures hook length and catalyses the secretion‐substrate specificity switch from rod‐hook substrate specificity to late substrate secretion, which includes the filament subunits. Here, we show normal hook‐length control and filament assembly in the complete absence of the C‐ring thus refuting the previous ‘cup’ model for hook‐length control. Mutants of C‐ring components, which are reported to produce short hooks, show a reduced rate of hook–basal body assembly thereby allowing for a premature secretion‐substrate specificity switch. Unlike fliK null mutants, hook‐length control in an autocleavage‐defective mutant of flhB, the protein responsible for the switch to late substrate secretion, is completely abolished. FliK deletion variants that retain the ability to measure hook length are secreted thus demonstrating that FliK directly measures rod‐hook length during the secretion process. Finally, we present a unifying model accounting for all published data on hook‐length control in which FliK acts as a molecular ruler that takes measurements of rod‐hook length while being intermittently secreted during the assembly process of the hook–basal body complex. 相似文献
A rapid and sensitive method for quantifying iodine in intact starch granules using gas chromatography is described with detection limits as low as 0.2% (w/w) iodine in starch. Sample preparation includes NaBH4 reduction of the various iodine species associated with starch to the colorless soluble iodide ion, followed by its quantitative derivatization to EtI using in CH2Cl2. Identification and quantification of EtI is carried out by extraction and injection of the EtI so generated in CH2Cl2 into a gas chromatography-mass spectrometer (GC-MS). Routine quantification of EtI was then performed using GC with a flame ionization detector (GC-FID). Results for different iodine:potassium iodide ratios of the initially bound iodine and for seven different starch matrices showed that in all cases regression coefficients for the standards were high (R2 >0.96). 相似文献
This article explores the consequences of rapidly fissuring binary models of community and progress in a national and global context. It suggests that this is a dominant (black/white) paradigm used to map both mainstream and particularly liminal sites of cultural/political articulation. Furthermore, it argues that such models of imputed progress, reason and civility contain the narcissistic residue of an earlier liberal encounter: namely, the fetish quality attached to the black body as abject text and privileged humanist ontology by the twentieth-century European movement of liberal humanism. The article argues that recent attempts to rethink this position vis à vis the European Holocaust have actually rehearsed the logic, both of fetish and binarism. Moreover, that this stasis in liberal thinking on issues of race and violence has heightened the attraction of those political movements characterized by cultural absolutism. The essay suggests that acknowledgement of the 'public secret' which underwrites liberal narrative stasis is the first step towards a more appropriate cultural grammar. 相似文献
Dopamine replacement therapy in Parkinson's disease is associated with several unwanted effects, of which dyskinesia is the most disabling. The development of new therapeutic interventions to reduce the impact of dyskinesia in Parkinson's disease is therefore a priority need. This review summarizes the key molecular mechanisms that underlie dyskinesia. The role of dopamine receptors and their associated signaling mechanisms including dopamine‐cAMP‐regulated neuronal phosphoprotein, extracellular signal‐regulated kinase, mammalian target of rapamycin, mitogen and stress‐activated kinase‐1 and Histone H3 are summarized, along with an evaluation of the role of cannabinoid and nicotinic acetylcholine receptors. The role of synaptic plasticity and animal behavioral results on dyskinesia are also evaluated. The most recent therapeutic advances to treat Parkinson's disease are discussed, with emphasis on the possibilities and limitations of non‐pharmacological interventions such as physical activity, deep brain stimulation, transcranial magnetic field stimulation and cell replacement therapy. The review suggests new prospects for the management of Parkinson's disease‐associated motor symptoms, especially the development of dyskinesia.
