In order to know the contribution of adrenal and gonadal androgens to the development of the side gland of Suncus murinus, we studied the effects of gonadectomy and adrenalectomy on gland thickness and the plasma levels of testosterone, androstenedione (delta 4-dione) and dehydroepiandrosterone (DHA). In males, castration decreased gland thickness to 71% of the control. The plasma levels of delta 4-dione and testosterone were also decreased from 4.16 +/- 0.50 and 0.65 +/- 0.10 ng/ml to 1.44 +/- 0.17 and 0.12 +/- 0.02 ng/ml, respectively. Adrenalectomy following castration caused no notable additional decrease in gland thickness, although the plasma levels of delta 4-dione and DHA were further decreased by this treatment. In females, ovariectomy affected neither gland thickness nor plasma androgen levels, except for a peculiar rise in the plasma concentration of delta 4-dione. In contrast, adrenalectomy in addition to ovariectomy decreased gland thickness to 63% of the control and the plasma concentrations of delta 4-dione and DHA from 1.43 +/- 0.26 and 0.43 +/- 0.05 ng/ml to 0.37 +/- 0.11 and 0.10 +/- 0.04 ng/ml, respectively. Therefore, testicular androgens are required for the male side gland to fully develop, whereas in the female adrenal androgens are important for the maintenance of sebaceous gland activity and delta 4-dione is quantitatively more important than DHA. One hour after the intraperitoneal administration of [3H]delta 4-dione, dihydrotestosterone was found to be the major androgen bound to nuclei of the side gland. Frozen bone-patellar tendon bone allografts are useful in anterior cruciate ligament reconstruction as the freezing procedure
kills tissue cells, thereby reducing immunogenicity of the grafts. However, a small portion of cells in human femoral heads
treated by standard bone-bank freezing procedures survive, thus limiting the effectiveness of allografts. Here, we characterized
the survival rates and mechanisms of cells isolated from rat bones and tendons that were subjected to freeze–thaw treatments,
and evaluated the influence of these treatments on the mechanical properties of tendons. After a single freeze–thaw cycle,
most cells isolated from frozen bone appeared morphologically as osteocytes and expressed both osteoblast- and osteocyte-related
genes. Transmission electron microscopic observation of frozen cells using freeze-substitution revealed that a small number
of osteocytes maintained large nuclei with intact double membranes, indicating that these osteocytes in bone matrix were resistant
to ice crystal formation. We found that tendon cells were completely killed by a single freeze–thaw cycle, whereas bone cells
exhibited a relatively high survival rate, although survival was significantly reduced after three freeze–thaw cycles. In
patella tendons, the ultimate stress, Young’s modulus, and strain at failure showed no significant differences between untreated
tendons and those subjected to five freeze–thaw cycles. In conclusion, we identified that cells surviving after freeze–thaw
treatment of rat bones were predominantly osteocytes. We propose that repeated freeze–thaw cycles could be applied for processing
bone-tendon constructs prior to grafting as the treatment did not affect the mechanical property of tendons and drastically

Platinum was determined by the inductively coupled plasma mass spectrometry (ICP-MS) in the intervertebral discs and vertebrae of ovarian tumor bearing patients treated withcis-diamminedichloro-platinum (II) (cisplatin). Platinum was 0.05 ng/mL at the absolute detection limit, and platinum was undetectable in the intervertebral discs and vertebrae of human specimens without cisplatin treatments. On the other hand, platinum was detected in the intervertebral discs and vertebrae of patients administered cisplatin, and platinum concentration was at levels of 1.06–10.31 μg/g dry tissue in the intervertebral discs and 0.60–1.28 μg/g dry tissue in the vertebrae, respectively. The platinum level of intervertebral discs was 4.3-fold higher than that of the vertebrae. The 465,813 base pair sequence corresponding to the 40.150.0min region on the genetic map of Escherichia coli K-12 (W3110)was determined. Analysis of the sequence revealed that thisregion contained at least 466 potential open reading frames,of which 187 (40%) were previously reported, 105 (23%) werehomologous to other known genes, 103 (22%) were identical orsimilar to hypothetical genes registered in databases, and theremaining 71 (15%) did not show a significant similarity toany other gene. At the 45.246.0 min region, we founda very large cluster of about 30 genes, whose functions areinvolved in the biosynthesis of polysaccharides as the componentsof outer membranes.
