Background and aims
Sufficient soil phosphorus (P) is important for achieving optimal crop production, but excessive soil P levels may create a risk of P losses and associated eutrophication of surface waters. The aim of this study was to determine critical soil P levels for achieving optimal crop yields and minimal P losses in common soil types and dominant cropping systems in China.Methods
Four long-term experiment sites were selected in China. The critical level of soil Olsen-P for crop yield was determined using the linear-plateau model. The relationships between the soil total P, Olsen-P and CaCl2-P were evaluated using two-segment linear model to determine the soil P fertility rate and leaching change-point.Results
The critical levels of soil Olsen-P for optimal crop yield ranged from 10.9 mg kg?1 to 21.4 mg kg?1, above which crop yield response less to the increasing of soil Olsen-P. The P leaching change-points of Olsen-P ranged from 39.9 mg kg?1 to 90.2 mg kg?1, above which soil CaCl2-P greatly increasing with increasing soil Olsen-P. Similar change-point was found between soil total P and Olsen-P. Overall, the change-point ranged from 4.6 mg kg?1 to 71.8 mg kg?1 among all the four sites. These change-points were highly affected by crop specie, soil type, pH and soil organic matter content.Conclusions
The three response curves could be used to access the soil Olsen-P status for crop yield, soil P fertility rate and soil P leaching risk for a sustainable soil P management in field. 相似文献82.

Functional recovery is usually poor following peripheral nerve injury when reinnervation is delayed. Early innervation by sensory nerve has been indicated to prevent atrophy of the denervated muscle. It is hypothesized that early protection with sensory axons is adequate to improve functional recovery of skeletal muscle following prolonged denervation of mixed nerve injury. In this study, four groups of rats received surgical denervation of the tibial nerve. The proximal and distal stumps of the tibial nerve were ligated in all animals except for those in the immediate repair group. The experimental groups underwent denervation with nerve protection of peroneal nerve (mixed protection) or sural nerve (sensory protection). The experimental and unprotected groups had a stage II surgery in which the trimmed proximal and distal tibial nerve stumps were sutured together. After 3 months of recovery, electrophysiological, histological and morphometric parameters were assessed. It was detected that the significant muscle atrophy and a good preserved structure of the muscle were observed in the unprotected and protective experimental groups, respectively. Significantly fewer numbers of regenerated myelinated axons were observed in the sensory-protected group. Enhanced recovery in the mixed protection group was indicated by the results of the muscle contraction force tests, regenerated myelinated fiber, and the results of the histological analysis. Our results suggest that early axons protection by mixed nerve may complement sensory axons which are required for promoting functional recovery of the denervated muscle natively innervated by mixed nerve. 相似文献

Acute graft-versus-host disease(a GVHD) is caused by allo-activated donor T cells infiltrating target organs. As a regulator of immune function, granulocyte colony-stimulating factor(G-CSF) has been demonstrated to relieve the a GVHD reaction.However, the role of G-CSF-primed donor Tcells in specific target organs is still unknown. In this study, we employed a classical MHC-mismatched transplantation mouse model(C57BL/6 into BALB/c) and found that recipient mice transplanted with GCSF-primed T cells exhibited prolonged survival compared with that of the PBS-treated group. This protective function against GVHD mediated by G-CSF-primed donor T cells was further confirmed by decreased clinical and pathological scores in this a GVHD mouse model, especially in the lung and gut. Moreover, we found that Tcells polarized towards Th2 cells and regulatory T cells were increased in specific target organs. In addition, G-CSF treatment inhibited inducible co-stimulator(ICOS) expression and increased the expression of tolerance-related genes in recipient mice. Our study provides new insight into the immune regulatory effects of G-CSF on T cell-mediated a GVHD, especially for its precise regulation in GVHD target organs. 相似文献

Embryonic stem (ES) cells are of significant interest either as an in vitro model recapitulating early embryonic development or as a renewable source of therapeutically useful cells. ES cells aggregation is important for embryoid bodies (EBs) formation and the subsequent generation of ES cell derivatives. This study was conducted to describe scalable production of EBs by the rotary cell culture system (RCCS, STLV type) and estimate the feasibility of constructing engineered cardiac tissue (ECT). In comparison with suspension culture in a Petri dish, the efficiency of the dynamic process was analyzed with respect to the yield of EB formation and their cardiomyocyte differentiation. Cardiomyocyte differentiation was evaluated by immunohistochemical analysis. After the elementary enrichment by gradient percoll, ES cell-derived cardiomyocytes were applied to construct ECT. Cell gross morphology, spatial distribution, and ultrastructure were evaluated by using histological analysis, confocal laser scanning microscopy, and transmission electron microscopy. Results showed that EB efficiencies in STLV were nearly 1.5-2.0 times higher than that of liquid suspension cultures, and cardiomyocyte differentiation of EBs progressed in a normal course after the dynamic cultivation in STLV. Additionally, the differentiated cultures could be enriched elementarily by gradient percoll. Once cast into the collagen strand, cells grew well and became more matured in Petri dishes. Synchronous contraction of the cell cluster was observed on the surface of the ECT, and cell connection was also established. It was the first report to have beating ES-derived cardiomyocytes on a 3-D collagen scaffold, which might provide a promising model for physiological and pharmacological studies and tissue replacement therapy. 相似文献

Aims
To clarify the efficacy and safety of adding sitagliptin to insulin therapy in Japanese patients with suboptimally controlled type 2 diabetes (T2DM).Study Design and Methods
This was a 24-week, prospective, randomized, open-labeled, controlled trial. Patients with T2DM who were suboptimally controlled despite receiving at least twice daily injection of insulin were enrolled in the study. The patients were randomized to continuation of insulin treatment (Insulin group) or addition of sitagliptin 50 to 100 mg daily to insulin treatment (Ins+Sita group). The primary outcome was change in HbA1c at week 24.Results
Adding sitagliptin to insulin significantly reduced HbA1c from 7.9 ± 1.0% at baseline to 7.0 ± 0.8% at week 24 (P <0.0001), while there was no significant change in HbA1c in the Insulin group (7.8 ± 0.7% vs. 7.8 ± 1.1%, P = 0.32). The difference in HbA1c reduction between the groups was 0.9% (95% confidence interval, 0.4 to 1.5, P = 0.01). There was no significant weight gain in either group. Incidence of hypoglycemia was significantly reduced in the Ins+Sita group compared with the Insulin group. Treatment satisfaction was improved in the Ins+Sita group. Baseline HbA1c level and beta cell function were associated with the magnitude of reduction in HbA1c in the Ins+Sita group.Conclusion
Adding sitagliptin to insulin reduced HbA1c without weight gain or increase in hypoglycemia, and improved treatment satisfaction in Japanese patients with T2DM who were suboptimally controlled despite at least twice daily injection of insulin.Trial Registration

研究了无芒隐子草 (Cleistogenessongorica)和条叶车前 (Plantago lessingii)种子萌发对不同温度和水分渗透势的响应。温度在 10℃恒温至 5 0 /2 0℃变温范围内设 11个处理 ,水分渗透势在 0~ - 1.8MPa区间以 0 .2 MPa为间隔设 10个处理。结果显示 :无芒隐子草鞘内小穗的种子具有很高的生活力 ,其萌发温度幅较宽 ,在 10~ 5 0 /2 0℃温度区间 ,较高温度促进萌发 ;条叶车前种子萌发温度幅较窄 ,在 10~ 30℃温度区间 ,较低温度促进其萌发。无芒隐子草和条叶车前种子的最佳萌发温度分别为35 /2 0℃变温和 2 0℃恒温 ,在此条件下的发芽率分别达 94 %和 6 1% ,后者发芽率低的原因是种子存在生理休眠。两种植物种子萌发的最低水分阈值为 - 1.6 MPa,发芽率皆随渗透势降低而呈直线下降趋势 ,但条叶车前较无芒隐子草下降缓慢。发芽率 (y)与渗透势 (x)的回归方程 ,无芒隐子草为 y1 =- 10 .976 x1 98.4 (r2 =0 .95 4 ) ,条叶车前为 y2 =- 5 .90 9x2 6 0 .2 (r2 =0 .96 4 )。随着水分胁迫的加剧 ,供试种胚芽长呈逐渐下降趋势 ,但胚根长呈先增加 (无芒隐子草在 - 0 .6 MPa,而条叶车前在 - 0 .4 MPa)而后下降的趋势。讨论了两种植物种子萌发对不同温度和水分响应的生态学意义 相似文献

Four experiments were conducted to study 1) factors affecting porcine oocyte maturation in culture medium and 2) a new method for oocyte maturation outside the porcine body. In Experiment 1, five groups of oocytes were cultured in m-TCM199 or m-KRB medium for 24 to 28, 32 to 36 or 40 to 42 hours and then were fertilized in vitro. The cleavage rate (two to four-cell stage) of oocytes cultured for 32 to 36 hours was significantly higher than those of the other oocytes. The results indicate that a suitable culture period for the in vitro maturation of porcine oocytes is 32 to 36 hours. In Experiment 2, four groups of oocytes were cultured in m-KRB or m-KRB supplemented with PFF, PMSG or FSH for in vitro maturation, and the cleavage rates of oocytes were 7.94, 22.56, 30.23 and 23.26%, respectively, after in vitro fertilization. The results show that porcine follicular fluid (PFF) and gonadotrophins added to the culture medium promote porcine oocyte maturation in vitro. In Experiment 3, oocytes were cultured in m-KRB or m-TCM199, supplemented with both gonadotrophin and pocine folliclar fluid for maturation in vitro. After fertilization in vitro, the cleavage rates of oocytes were 26.32 and 27.93% for the two media. The results indicate that the difference between m-KRB and m-TCM199 was insignificant when the media were used to culture porcine oocytes. But there was a significant difference when PFF and gonadotrophins were added to the basic media. In Experiment 4, porcine oocytes were transferred into the reproductive tracts of other animals for maturation. After 34 to 36 hours, the oocytes were collected and fertilized in vitro. The cleavage rates of oocytes were 10.42, 28.45, 3.33 and 36.36%, respectively, for the oocytes matured in mouse uterine horns, rat uterine horns, rat oviducts or rabbit oviducts. 林下灌木是亚热带常绿阔叶林重要的构成部分,但林下灌木细根功能性状变异规律及地下生态策略仍不清楚。以福建建瓯万木林自然保护区内9种灌木为研究对象,对细根直径、根长、比根长、组织密度、碳浓度和氮浓度6个细根性状进行研究,采用序级划分法,分析不同树种细根性状序级间的变化特征、常绿和落叶灌木细根性状之间的差异,不同序级细根性状之间的关系以及细根性状变异维度。结果表明:树种和序级对9种灌木细根形态和化学性质有显著影响。直径、根长、根组织密度随着序级的增加而逐渐增加,比根长和氮浓度逐渐减小,碳浓度在序级间的变化趋势不一,未表现出明显的规律。落叶灌木细根直径、根长和氮浓度均显著高于常绿灌木,碳浓度和组织密度显著低于常绿灌木,表明与常绿灌木相比落叶灌木更偏向于资源获取型生态策略,常绿灌木则更偏向于保守型策略。灌木细根在不同序级间的直径与比根长、组织密度,氮浓度与组织密度有较强的相关性,细根其他性状间的关系并不密切或因序级而异。主成分分析结果表明灌木细根性状变异沿一个主成分轴发生变异,该轴表示灌木细根的资源获取和保守的权衡策略。 相似文献

After low temperature (16℃) treatment, the net photosynthetic rate, chlorophyll content and FBPase activity tended to decrease and ratio of chlorophyll a/b, stomatal resistance, sucrose and starch content tended to increase in rice flag leaves at heading stage. However, the Pi content and water potential did not show pronounced change. The flag leaf chloroplasts contained large starch grains after three days at low temperature. The recovery of net photosynthetic rate in the flag leaves which had recieved stress at 16℃ for three days and transferred to normal nature condition was gradually increased. It almost came up to the value of control after five days. However, the decrease in stomatal resistance was rapid. It was almost lowered to the value of control after one day. Under normal nature comdition for five days, the chlorophyll content and FBPase activity reached near to the value of control. Meanwhile, the ratio of chlorophyll a/b, the sucrose and starch content almost declined to the value of control. The large starch grain in flag leaf chloroplast also disappeared. According to the experimental results, we discussed the relationship between the above-mentioned various changes and net photosynthetic rate We suggested that the effect of low temperature on photosynthesis might be one of factors in yield-reduction of rice. 相似文献

The cathepsin E-A-like, also known as ‘similar to nothepsin’, is a new member of the aspartic protease family, which may take part in processing of egg yolk macromolecules, due to it was identified in the chicken egg-yolk. Previously, studies have suggested that the expression of cathepsin E-A-like increased gradually during sexual maturation of pullets, but the exact regulation mechanism is poorly understood. In this study, to gain insight into the function and regulation mechanism of the gene in egg-laying hen, we cloned the cathepsin E-A-like gene and evaluated its evolutionary origin by using both phylogenetic and syntenic methods. The mode of the gene expression regulation was analysed through stimulating juvenile hens with \(17\upbeta \)-estradiol and chicken embryo hepatocytes with \(17\upbeta \)-estradiol combined with oestrogen receptor antagonists including MPP, ICI 182,780 and tamoxifen. Our results showed that cathepsin E-A-like was an orthologoues gene with nothepsin, which is present in birds but not in mammals. The expression of cathepsin E-A-like significantly increased in a dose-dependent manner after the juvenile hens were treated with \(17\upbeta \)-estradiol (\(P~<~0.05\)). Compared with the \(17\upbeta \)-estradiol treatment group, the expression of cathepsin E-A-like was not significantly changed when the hepatocytes were treated with \(17\upbeta \)-estradiol combined with MPP (\(P~<~0.05\)). In contrast, compared with the \(17\upbeta \)-estradiol combined with MPP treatment group, the expression of cathepsin E-A-like was significantly downregulated when the hepatocytes were treated with \(17\upbeta \)-estradiol combined with tamoxifen or ICI 182,780 (\(P~<~0.05\)). These results demonstrated that cathepsin E-A-like shared the same evolutionary origin with nothepsin. The expression of cathepsin E-A-like was regulated by oestrogen, and the regulative effect was predominantly mediated through ER-\(\upbeta \) in liver of chicken. 相似文献
