The method is proposed for calculation of the most important parameters of the two-stage enzymatic or transport process--modification factors alpha and beta (which characterize the effector action mechanism) as well as the inhibition constant K(i) or activation constant K(a) (characterize the effector affinity for protein). The method was derived as based on the analysis of kinetic regularities of the action of reversible effectors (inhibitors and activators) on the catalytic (transport) activity of proteins. The method is based on the titration of enzymatic (transport) protein by the substrate with the absence and with presence of the effector taken in one of concentrations as well as determination (under the fixed substrate concentration) of the inhibition coefficient i(0.5) (in case of the inhibitor action) or the activation coefficient a(0.5) (in case of the activator action). Practical use of the method has been demonstrated on the example of reversible inhibition to eosine Y (2', 4', 5', 7' - tetrabromofluorescein) ofthe reaction of enzymatic hydrolysis of ATP catalyzed by highly purified transport Ca2+, Mg(2+)-ATPase isolated from the smooth-muscle sarcolemma. In this case the inhibitory effect is characterized by the following parameters: alpha = 6-8 > 1; beta = 0.50-0.53 < 1; inhibition constant K(i) = 10(-9) - 10(-8) M. Consequently, judging from the values of alpha and beta, the eosine Y effect on the analyzed Ca2+, Mg(2+)-dependent ATP-hydrolase enzymatic reaction is based on the mechanism of the mixed inhibition (one can observe the inhibition of the both stages of enzymatic transformation--the substrate binding with the enzyme and decomposition of "Michaelis complex" in the direction of formation of the reaction products). The inhibitor itself, in correspondence with K(ij) values is characterized by rather high affinity for Ca2+, Mg(2+)-ATPase. It is supposed that the proposed approach can be useful when identifying the type of the reversible effector action on the enzymatic (transport) activity of proteins, estimation of real affinity of the inhibitors and activators for the latter.  相似文献

In experiments carried out with the use of the radioactive label (45Ca2+) on suspension of the rat uterus myocytes processed by digitonin solution (0.1 mg/ml), influence of spermine and cyclosporin A on Mg2+, ATP-dependent Ca2+ transport in mitochondria at different Mg2+ concentration were investigated. Ca2+ accumulation in mitochondria was tested as such which was not sensitive to thapsigargin (100 nM) and was blocked by ruthenium red (10 microM). It has been shown, that spermine (1 mM) stimulates Mg2+, ATP-dependent Ca2+ accumulation in mitochondria irrespective of Mg2+ concentration (3 or 7 mM) in the incubation medium. At the same time cyclosporin A (5 microM) effects on Ca2+ accumulation in mitochondria depend on Mg2+ concentration in the incubation medium: at 3 mM Mg2+ the stimulating effect was observed, and at 7 mM Mg2+ - the inhibitory one. In conditions which led to the increase of nonspecific mitochondrial permeability and, accordingly, to dissipation of electrochemical potential (it was reached by 5 min. preincubation of myocytes suspension in the medium that contained 10 microM Ca2+, 2 mM phosphate and 3 or 7 mM Mg2+, but not ATP) significant inhibition of Mg2+, ATP-dependent Ca2+ accumulation in mitochondria was observed. The inhibition to the greater degree was observed when medium ATP and Mg2+ were absent simultaneously in the preincubation. Thus the quality of spermine effects on Ca2+ accumulation was kept: stimulation in the presence both of 3 mM and 7 mM Mg2+. Ca2+ accumulation did not reach the control level when 3 mM Mg2+ and 1 mM spermine was present and ATP absent in the preincubation medium. However, in the presence of 7 mM Mg2+ and 1 mM spermine practically full restoration (up to a control level) of Ca2+ accumulation was observed. At the same time with other things being equal such restoration was not observed at simultaneous absence of ATP and Mg2+ in the preincubation medium. The quality of cyclosporin A effects on Ca2+ accumulation in mitochondria was also kept: stimulation - in the presence of 3 mM Mg2+, inhibition - in the presence of 7 mM Mg2+ in the preincubation medium. And, at last, in the presence of cyclosporin A irrespective of the fact which preincubation medium was used, Ca2+ accumulation level practically did not depend on Mg2+ concentration.  相似文献

Evidence on the influence of Ca2+ on the binding of neutral red by plasma membranes of skeletal muscle is dealt with. It is suggested that the binding of Ca2+ occurs according to the principle of bimolecular binding. It is suggested that the Ca2+-binding sites with high affinity may control the process of calcium transport by plasma membranes.  相似文献

The supramolecular compound calix[4]arene C-90 (5,11,17,23-tetra(trifluoro)methyl(phenylsulfonylimino)-methylamino-25,26,27,28-tetrapropoxycalix[4]arene) is shown to efficiently inhibit the ATP hydrolase activity of Ca2+,Mg2+-ATPase in the myometrium cell plasma membrane fraction and also in a preparation of the purified enzyme solubilized from this subcellular fraction. The inhibition coefficient I 0.5 values were 20.2 ± 0.5 and 58.5 ± 6.4 μM for the membrane fraction and the solubilized enzyme, respectively. The inhibitory effect of calix[4]arene C-90 was selective comparatively to other ATPases localized in the plasma membrane: calix[4]arene C-90 did not influence the activities of Na+,K+-ATPase and “basal” Mg2+-ATPase. The inhibitory effect of calix[4]arene C-90 on the Ca2+,Mg2+-ATPase activity was associated with the cooperative action of four trifluoromethylphenyl sulfonylimine (sulfonylamidine) groups oriented similarly on the upper rim of the calix[4]arene macrocycle (the calix[4]arene “bowl”). The experimental findings seem to be of importance for studies, using calix[4]arene C-90, of membrane mechanisms of regulation of calcium homeostasis in smooth muscle cells and also for investigation of the participation of the plasma membrane Ca2+-pump in control of electro- and pharmacomechanical coupling in myocytes.  相似文献

The genetic basis of nuclear-cytoplasmic incompatibility was examined using the wild pea (Pisum sativum subsp. elatius) accession VIR320. When this accession is used as the female parent in crosses with domesticated peas (Pisum sativum subsp. sativum) the F1 is highly sterile and displays chlorophyll deficiency, chlorophyll variegation, reduction of leaflets and stipulae while the reciprocal cross produces hybrids that appear normal. A mapping recombinant inbred line (RIL) population was established based on a cross in a compatible direction of a tester line WL1238 with VIR320. The ability to cause nuclear-cytoplasmic conflict was analysed by crossing individual RIL plants as pollen parents with VIR320 as donor of cytoplasm and scoring each F1 for major signs of the conflict. It is concluded that two unlinked nuclear genes are involved in the genetic control of the observed incompatibility. One of the genes, denoted as Scs1, is closely linked to the PhlC gene on linkage group III and the other, denoted as Scs2, is closely linked to the gp gene on linkage group V. Alleles of both genes in WL1238 are dominant and appear to be lethal in the homozygous condition in the VIR320 cytoplasm background. The inhibitory action of calixarene C-107 (5,17-diamino(2-pyridyl)methylphosphono- 11,23-di-tret-butyl-26,28-dihydroxy-25,27-dipropoxy-calix[4]arene) on Na+, K(+)-ATPase activity kinetic properties of myometrium perforated plasma membrane was investigated. It has been shown that the calixarene C-107 inhibiting Na+, K(+)-ATPase does not change the kinetic parameters (Km, nH) of reaction velocity dependence on substrate concentration. The constant Ka of enzyme activation by MgCl2 has complex dependence on calixarene C-107 concentration: it increases twice with growth of calixarene concentration up to 50 nM and decreases to the control level with further growth of calixarene concentration. The Hill cooperativity coefficient nH of activation by MgCl2 does not vary in the presence of calixarene C-107. Both ATP and MgCl2 have no influence on Na+, K(+)-ATPase constant of inhibition by calixarene C-107, but an increase of concentration of the mentioned physiological compounds causes the growth of cooperativity coefficient nH of enzymatic reaction inhibition by calixaren C-107.  相似文献

The effect of divalent cations--Co2+, Cu2+, Mn2+ and Ni2+ (5 mM) on the activity of actomyosin complex ATPase and ATPase of subfragment-1 (S1,head) of myosin from smooth muscle of the uterus was studied. It has been shown that Co2+, Mn2+ and Ni2+ inhibited, while Cu2+ activates the enzyme activity of both actomyosin and myosin S1. Mg and Mn ions had practically no effect on the emission intensity of eosin Y associated with actomyosin, while one could observe the most marked suppression of emission of related fluorescent probe in the presence of Cu cations and less pronounced suppression in the presence of Co2+. In the presence of Mn, Co and Ni cations the average hydrodynamic diameter (HD) of actomyosin complex and of subfragment-1 of the smooth muscle of the uterus is virtually identical to the HD in the presence of Mg2+. In the presence of Cu cations there is a considerable (ten-fold) increase in the size of the protein particles that may be a result of their aggregation. The results obtained evidence for the significant changes in the structure and function of the actomyosin complex of the myometrium in the presence of heavy metals and allow us to assume that the target of the effect of these metals on the contractile proteins is a subfragment-1 of myosin, where the active site of ATPase and actin-binding sites are localized.  相似文献

Na+, K(+)-ATPase--a protein complex of plasmatic membrane, which performs the dual function: firstly, it supports the Na+ and K+ homeostasis, and also transmembrane potential gradient, secondly, it serves as the transducer of signals and as the regulator of the expression of many key genes. Endogenous cardiotonic steroids, which are synthesized in the adrenal glands and hypothalamus, serve as the signal molecules. New concepts about the mechanisms of the realization of the Na+, K(+)-ATPase signal function and their connection with cellular functions, apoptosis, and with pathologies of cardiovascular system and water-salt homeostasis are described in the survey.  相似文献

Vanadate (10(-4)-10(-3) M) effectively blocks Mg2+, ATP-dependent Ca2+ transport in sarcolemmal vesicles and induces a slowly tonic contraction of the smooth muscle. This contraction was observed both with and without nifedipine (10(-5) M) evoking complete inhibition of hyperpotassium contracture, the Ca2+ removal from the solution washing the muscular preparation stimulating the tone decrease. There is a close correlation between the dose-dependent effects of vanadate on the Ca pump activity and tension. It is concluded that in smooth muscles, at least in myometrium, the sarcolemmal Ca-pump is involved into the control of the tonic tension.  相似文献
