Glycyrrhetinic acid, the metabolite of the natural product glycyrrhizin, is a well known nonselective inhibitor of 11β-hydroxysteroid dehydrogenase (11β-HSD) type 1 and type 2. Whereas inhibition of 11β-HSD1 is currently under consideration for treatment of metabolic diseases, such as obesity and diabetes, 11β-HSD2 inhibitors may find therapeutic applications in chronic inflammatory diseases and certain forms of cancer. So far, no selective 11β-HSD2 inhibitor has been developed and neither animal studies nor clinical trials have been reported based on 11β-HSD2 inhibition. Starting from the lead compound glycyrrhetinic acid, novel triterpene type derivatives were synthesized and analyzed for their biological activity against overexpressed human 11β-HSD1 and 11β-HSD2 in cell lysates. Several hydroxamic acid derivatives showed high selectivity for 11β-HSD2. The most potent and selective compound is active against human 11β-HSD2 in the low nanomolar range with a 350-fold selectivity over human 11β-HSD1.  相似文献

Suberin is found in a variety of tissues, such as root endoderms and periderms, storage tuber periderms, tree cork layer, and seed coats. It acts as a hydrophobic barrier to control the movement of water, gases, and solutes as well as an antimicrobial barrier. Suberin consists of polymerized phenolics, glycerol, and a variety of fatty acid derivatives, including primary fatty alcohols. We have conducted an in-depth analysis of the distribution of the C18:0 to C22:0 fatty alcohols in Arabidopsis (Arabidopsis thaliana) roots and found that only 20% are part of the root suberin polymer, together representing about 5% of its aliphatic monomer composition, while the remaining 80% are found in the nonpolymeric (soluble) fraction. Down-regulation of Arabidopsis FATTY ACYL REDUCTASE1 (FAR1), FAR4, and FAR5, which collectively produce the fatty alcohols found in suberin, reduced their levels by 70% to 80% in (1) the polymeric and nonpolymeric fractions from roots of tissue culture-grown plants, (2) the suberin-associated root waxes from 7-week-old soil-grown plants, and (3) the seed coat suberin polymer. By contrast, the other main monomers of suberin were not altered, indicating that reduced levels of fatty alcohols did not influence the suberin polymerization process. Nevertheless, the 75% reduction in total fatty alcohol and diol loads in the seed coat resulted in increased permeability to tetrazolium salts and a higher sensitivity to abscisic acid. These results suggest that fatty alcohols and diols play an important role in determining the functional properties of the seed coat suberin barrier.Suberin is a cell wall-linked polymeric barrier that plays a critical role in the survival of plants by protecting them against various biotic and abiotic stresses. It primarily acts as a hydrophobic barrier to control the movement of water, gases, and solutes, but also contributes to the strength of the cell wall (Ranathunge et al., 2011). Suberin is deposited at the inner face of primary cell walls next to the plasma membrane (Kolattukudy, 1980; Franke and Schreiber, 2007). It is typically found as lamellae (alternating dark and light bands when viewed by transmission electron microscopy) in the endodermis, exodermis, and peridermis of roots, as well as in the peridermis of underground storage tubers (Bernards, 2002). Suberin is also found in shoot periderms of trees (i.e. cork layer) and in seed coats (Molina et al., 2006, 2008) and is deposited in response to wounding (Kolattukudy, 2001).Suberin is a polymer consisting of aliphatics (fatty acid derivatives), phenolics, and glycerol. The predominant aliphatic components of suberin are ω-hydroxy fatty acids, α,ω-dicarboxylic acids, very-long-chain fatty acids, and primary fatty alcohols, while the major phenolic components are p-hydroxycinnamic acids, especially ferulic acid (Kolatukudy, 1980; Bernards et al., 1995; Pollard et al., 2008; Ranathunge et al., 2011). In the periderm of underground storage organs, suberin is found in association with waxes, which are isolated either by extensive extraction in solvent (Soliday et al., 1979; Serra et al., 2009) or by brief immersion of tubers in chloroform (Espelie et al., 1980). These suberin-associated waxes consist of linear aliphatics (e.g. alkanes, fatty acids, and fatty alcohols), which are similar to cuticular wax components of aerial tissues but generally of shorter chain lengths (Espelie et al., 1980). In waxes extracted from 3-week-old wounded potato (Solanum tuberosum) periderms, alkyl ferulates (i.e. ferulic acid linked by an ester bond to a C16:0–C32:0 fatty alcohol) represent up to 60% of the total wax load (Schreiber et al., 2005). Root waxes are also found in 6- to 7-week-old mature taproots of Arabidopsis (Arabidopsis thaliana) with a fully developed periderm (Li et al., 2007; Kosma et al., 2012). They are enriched in alkyl hydroxycinnamates (AHCs) made of C18:0 to C22:0 fatty alcohols esterified with coumaric, caffeic, or ferulic acids (Kosma et al., 2012). The monomer composition (in terms of major chemical species and chain length) of both suberin and suberin-associated waxes varies considerably between plant species, tissues, and developmental stages. Aliphatic suberin and suberin-associated waxes are considered the major contributors to the diffusion resistance of suberized cell walls to radial transport of water and solutes (Soliday et al., 1979; Espelie et al., 1980; Zimmermann et al., 2000; Ranathunge and Schreiber, 2011). The organization of suberin components in the lamellated structure as well as how waxes may be associated with the polymer is a matter of debate (Graça and Santos, 2007).Primary fatty alcohols are long-chain hydrocarbons containing a single hydroxyl group at the terminal position. They are ubiquitously detected as components of the suberin polymer, representing 1% to 10% of the total monomer mass recovered after transesterification (Holloway, 1983; Bernards, 2002; Pollard et al., 2008). Primary fatty alcohols are also typical components of suberin-associated waxes, where they can be found either in free form or linked by an ester bond with a hydroxycinnamic acid (i.e. as AHCs; Soliday et al., 1979; Espelie et al., 1980; Bernards and Lewis 1992; Li et al., 2007; Kosma et al., 2012). In mechanically isolated endodermis of soybean (Glycine max) roots, fatty alcohols represent about 1.5% and 0.2% of the total aliphatics found in suberin-associated waxes and suberin polymer, respectively (Thomas et al., 2007). In onion (Allium cepa) root exodermis, fatty alcohols (C14:0–C28:0) account for 7% to 12% of the soluble fraction, while the suberin fraction contains only C22:0 fatty alcohol, which makes up 3% of the suberin fraction across all exodermal maturation zones (Meyer et al., 2011). In suberizing potato periderms 7 d post wounding, C16:0 to C28:0 fatty alcohols represent about 10% and 18% of the total aliphatics in the insoluble poly(aliphatic) domain (suberin polymer) and in the soluble (nonpolymeric) fraction, respectively (Yang and Bernards, 2006). A similar study on native periderms from 21-d-stored potato (Serra et al., 2009) reported that fatty alcohols represent about 20% of the total aliphatic components found in the suberin polyester, while unlinked fatty alcohols and alkyl ferulates accounted for about 23% and 44% of the total aliphatics in the soluble waxes.In Arabidopsis, C18:0, C20:0, and C22:0 fatty alcohols account for slightly less than 3% of the polymerized aliphatics in roots of soil-grown plants (Domergue et al., 2010), but as much as 36% [w/w] of the soluble wax load (Li et al., 2007). Arabidopsis fatty acyl reductases FAR1 (At5g22500), FAR4 (At3g44540), and FAR5 (At3g44550) generate, respectively, the C22:0, C20:0, and C18:0 fatty alcohol present in the suberin of root, seed coat, and wounded leaf tissues (Domergue et al., 2010). These three enzymes also generate the C18:0 to C22:0 fatty alcohol components that make up AHCs of root waxes (Kosma et al., 2012). Although one particular chain length of primary alcohol was reduced in each far single mutant line (C18:0-OH, C20:0-OH, and C22:0-OH in far5, far4, and far1, respectively), the total fatty alcohol load of the suberin polymer and its composition were only slightly affected and mutant plants had no obvious developmental or physiological defects (Domergue et al., 2010). In this study, we report on the distribution of primary fatty alcohols in the soluble (nonpolymeric) and insoluble (suberin polymer) fractions from mature roots of Arabidopsis. We report that far double and triple mutants have highly reduced fatty alcohol levels, in a chain length-specific manner, in both fractions as well as in the seed coat suberin polymer. The significant reductions in total fatty alcohol and diol levels in the seed coat of these mutants lead to increased permeability and higher sensitivity to abscisic acid (ABA), bringing to light insights on the roles of fatty alcohols and diols in determining functional properties of suberin.  相似文献

Hyaluronan, a major macropolysaccharide in the extracellular matrix of connective tissues, is intimately involved in the biology of cancer. Hyaluronan accumulates into the stroma of various human tumors and modulates intracellular signaling pathways, cell proliferation, motility and invasive properties of malignant cells. Experimental and clinicopathological evidence highlights the importance of hyaluronan in tumor growth and metastasis. A high stromal hyaluronan content is associated with poorly differentiated tumors and aggressive clinical behavior in human adenocarcinomas. Instead, the squamous cell carcinomas and malignant melanomas tend to have a reduced hyaluronan content. In addition to the stroma–cancer cell interaction, hyaluronan can influence stromal cell recruitment, tumor angiogenesis and epithelial–mesenchymal transition. Hyaluronan receptors, hyaluronan synthases and hyaluronan degrading enzymes, hyaluronidases, are involved in the modulation of cancer progression, depending on the tumor type. Furthermore, intracellular signaling and angiogenesis are affected by the degradation products of hyaluronan. Hyaluronan has also therapeutic implications since it is involved in multidrug resistance.  相似文献

Introduction

In the last decade, (18)F-fluorodeoxyglucose (18F-FDG) positron emission tomography (PET and PET/CT) has become one of the major diagnostic tools used in oncology. A significant number of patients who undergo this procedure, due to non-thyroidal reasons, present incidental uptake of (18F-FDG) in the thyroid. The aim of the study was to compare the SUVmax (standardized uptake value) of thyroid focal lesions, which were incidentally found on PET/CT, in relation to the results of thyroid fine-needle aspiration biopsy (FNAB) and/or histopathological evaluation.

Materials and Methods

Patients referred for PET/CT examination, due to non-thyroidal illness, presented focal 18F-FDG uptake in the thyroid and were advised to undergo ultrasonography (US), hormonal evaluation, FNAB and/or total thyroidectomy at our institution.

Results

6614 PET/CT examinations performed in 5520 patients were analyzed. Of the 122 patients with focal thyroid 18F-FDG activity, 82 patients (67.2%) underwent further thyroid evaluation using FNAB. Benign lesions were diagnosed in 46 patients, malignant - in 19 patients (confirmed by post-surgical histopathology), while 17 patients had inconclusive results of cytological assessment. Mean SUVmax of benign lesions was 3.2±2.8 (median = 2.4), while the mean SUVmax value for malignant lesions was 7.1±8.2 (median = 3.5). The risk of malignancy was 16.7% for lesions with a SUVmax under 3, 43.8% for lesions with a SUVmax between 3 and 6, and 54.6% for lesions with a SUVmax over 6. In the group of malignant lesions, a positive correlation between the lesion’s diameter and SUVmax was observed (p = 0.03, r = 0.57).

Conclusions

Subjects with incidental focal uptake of 18F-FDG in thyroid are at a high risk of thyroid malignancy. A high value of SUVmax further increases the risk of malignancy, indicating the necessity for further cytological or histological evaluation. However, as SUVmax correlated with the diameter of malignant lesions, small lesions with focal uptake of 18F-FDG should be interpreted cautiously.  相似文献

Micro-RNAs are small, noncoding RNAs that act as tumor suppressors or oncogenes. MiR-200c is a member of the miR-200 family; it is known to be dysregulated in invasive breast carcinoma. MiR-200c maintains the epithelial-mesenchymal transition and inhibits cell migration and invasion. Recent studies showed that miR-200c regulated steroid hormone receptors, estrogen receptors (ER), and progesterone receptors (PR). The present study aimed to detect miR-200c in 172 invasive breast carcinoma cases selected from a prospective cohort enrolled in Kuopio, Eastern Finland, between 1990 and 1995. MiR-200c expression was determined with relative q-PCR, and results were compared to clinicopathological variables and patient outcome. We found that PR status combined with miR-200c expression was a significant marker of outcome. High miR-200c expression was associated with reduced survival in PR-negative cases (n = 68); low miR-200c expression indicated reduced survival in PR-positive cases (n = 86) (Cox regression: P = 0.002, OR = 3.433; and P = 0.004, OR = 4.176, respectively). In PR-negative cases, high miR-200c expression was associated with shortened relapse-free survival (Cox regression: P = 0.001, OR = 3.613); increased local/distant recurrence (Logistic regression: P = 0.006, OR = 3.965); and more frequent distant metastasis (Logistic regression: P = 0.015, OR = 3.390). We also found that high grade and low stage tumors were positively correlated with high miR-200c expression (Logistic regression for high grade tumors: P = 0.002, OR = 2.791 and for high stage tumors: P = 0.035, OR = 0.285). Our results indicated that miR-200c may play a role in invasive breast carcinoma. Furthermore, miR-200c combined with PR status provided a refined predictor of outcome. In future, a larger study is required to confirm our results. This data may provide a basis for new research target–progesterone receptor–regulated microRNAs in breast cancer.  相似文献

Two β-galactosidases, β-gal I and β-gal II, from Bifidobacterium breve DSM 20213, which was isolated from the intestine of an infant, were overexpressed in Escherichia coli with co-expression of the chaperones GroEL/GroES, purified to electrophoretic homogeneity and biochemically characterized. Both β-gal I and β-gal II belong to glycoside hydrolase family 2 and are homodimers with native molecular masses of 220 and 211 kDa, respectively. The optimum pH and temperature for hydrolysis of the two substrates o-nitrophenyl-β-D-galactopyranoside (oNPG) and lactose were determined at pH 7.0 and 50°C for β-gal I, and at pH 6.5 and 55°C for β-gal II, respectively. The k cat/K m values for oNPG and lactose hydrolysis are 722 and 7.4 mM−1s−1 for β-gal I, and 543 and 25 mM−1s−1 for β-gal II. Both β-gal I and β-gal II are only moderately inhibited by their reaction products D-galactose and D-glucose. Both enzymes were found to be very well suited for the production of galacto-oligosaccharides with total GOS yields of 33% and 44% of total sugars obtained with β-gal I and β-gal II, respectively. The predominant transgalactosylation products are β-D-Galp-(1→6)-D-Glc (allolactose) and β-D-Galp-(1→3)-D-Lac, accounting together for more than 75% and 65% of the GOS formed by transgalactosylation by β-gal I and β-gal II, respectively, indicating that both enzymes have a propensity to synthesize β-(1→6) and β-(1→3)-linked GOS. The resulting GOS mixtures contained relatively high fractions of allolactose, which results from the fact that glucose is a far better acceptor for galactosyl transfer than galactose and lactose, and intramolecular transgalactosylation contributes significantly to the formation of this disaccharide.  相似文献

The Gram-positive bacterium Geobacillus stearothermophilus NRS 2004/3a possesses a cell wall containing an oblique surface layer (S-layer) composed of glycoprotein subunits. O-Glycans with the structure [-->2)-alpha-L-Rhap-(1-->3)-beta-L-Rhap-(1-->2)-alpha-L-Rhap-(1-->](n) (= 13-18), a2-O-methyl group capping the terminal repeating unit at the nonreducing end and a -->2)-alpha-L-Rhap-[(1-->3)-alpha-L-Rhap](n) (= 1-2)(1-->3)- adaptor are linked via a beta-D-Galp residue to distinct sites of the S-layer protein SgsE. S-layer glycan biosynthesis is encoded by a polycistronic slg (surface layer glycosylation) gene cluster. Four assigned glycosyltransferases named WsaC-WsaF, were investigated by a combined biochemical and NMR approach, starting from synthetic octyl-linked saccharide precursors. We demonstrate that three of the enzymes are rhamnosyltransferases that are responsible for the transfer of L-rhamnose from a dTDP-beta-L-Rha precursor to the nascent S-layer glycan, catalyzing the formation of the alpha1,3- (WsaC and WsaD) and beta1,2-linkages (WsaF) present in the adaptor saccharide and in the repeating units of the mature S-layer glycan, respectively. These enzymes work in concert with a multifunctional methylrhamnosyltransferase (WsaE). The N-terminal portion of WsaE is responsible for the S-adenosylmethionine-dependent methylation reaction of the terminal alpha1,3-linked L-rhamnose residue, and the central and C-terminal portions are involved in the transfer of L-rhamnose from dTDP-beta-L-rhamnose to the adaptor saccharide to form the alpha1,2- and alpha1,3-linkages during S-layer glycan chain elongation, with the methylation and the glycosylation reactions occurring independently. Characterization of these enzymes thus reveals the complete molecular basis for S-layer glycan biosynthesis.  相似文献

Plant cuticle is an extracellular lipid-based matrix of cutin and waxes, which covers aerial organs and protects them from many forms of environmental stress. We report here the characterization of CER8 / LACS1 , one of nine Arabidopsis long-chain acyl-CoA synthetases thought to activate acyl chains. Mutations in LACS1 reduced the amount of wax in all chemical classes on the stem and leaf, except in the very long-chain fatty acid (VLCFA) class wherein acids longer than 24 carbons (C24) were elevated more than 155%. The C16 cutin monomers on lacs1 were reduced by 37% and 22%, whereas the C18 monomers were increased by 28% and 20% on stem and leaf, respectively. Amounts of wax and cutin on a lacs1-1 lacs2-3 double mutant were much lower than on either parent, and lacs1-1 lacs2-3 had much higher cuticular permeability than either parent. These additive effects indicate that LACS1 and LACS2 have overlapping functions in both wax and cutin synthesis. We demonstrated that LACS1 has synthetase activity for VLCFAs C20–C30, with highest activity for C30 acids. LACS1 thus appears to function as a very long-chain acyl-CoA synthetase in wax metabolism. Since C16 but not C18 cutin monomers are reduced in lacs1 , and C16 acids are the next most preferred acid (behind C30) by LACS1 in our assays, LACS1 also appears to be important for the incorporation of C16 monomers into cutin polyester. As such, LACS1 defines a functionally novel acyl-CoA synthetase that preferentially modifies both VLCFAs for wax synthesis and long-chain (C16) fatty acids for cutin synthesis.  相似文献

Sacculi of Bacillus sphaericus CCM 2177 contain a secondary cell wall polymer which was completely extracted with 48% hydrofluoric acid. Nuclear magnetic resonance analysis showed that the polymer is composed of repeating units, as follows: -->3)-[4, 6-O-(1-carboxyethylidene)]( approximately 0. 5)-beta-D-ManpNAc-(1-->4)-beta-D-GlcpNAc-(1-->. High-resolution structures reveal how a germline antibody can recognize a range of clinically relevant carbohydrate epitopes. The germline response to a carbohydrate immunogen can be critical to survivability, with selection for antibody gene segments that both confer protection against common pathogens and retain the flexibility to adapt to new disease organisms. We show here that antibody S25-2 binds several distinct inner-core epitopes of bacterial lipopolysaccharides (LPSs) by linking an inherited monosaccharide residue binding site with a subset of complementarity-determining regions (CDRs) of limited flexibility positioned to recognize the remainder of an array of different epitopes. This strategy allows germline antibodies to adapt to different epitopes while minimizing entropic penalties associated with the immobilization of labile CDRs upon binding of antigen, and provides insight into the link between the genetic origin of individual CDRs and their respective roles in antigen recognition.  相似文献
