Possibility of reversing the action of the membranotropic antibiotic, gramicidin S, on bacteria

The study on the possibility of eliminating gramicidin S from the bacterial cells which had adsorbed it showed that a part of the labeled antibiotic bound by the bacteria may be washed out with buffer or salines. When the cells which had adsorbed gramicidin S were treated with lecithin emulsion, a significant part of the bound antibiotic was transferred to the lecithin liposomes. This turned the gramicidin S effect to the cells: significant but not complete reduction of the membrane barrier properties and dehydrogenase reactivation. Elimination of gramicidin S also reduced the colony forming capacity in a part of the cells.

Use of bromthymol blue for studying the thermotropic properties of the liposomes from egg lecithin

The dependence of the turbidity changes at 615 nm monobilayers liposomes from egg yolk lecithin in the presence of bromothymol blue on temperature and storage conditions has been investigated. It is established that the thermotropic properties of liposomes change irregularly and depend on the storage conditions. Sharp release of the bound dye at temperature above 35-37 degrees C is associated with thermotropic change in liposomes and the detected effects, with the change of orientation of the phosphorylcholine group.     

A new method of global search for functional DNA segments using a fractal representation of nucleotide texts]

New global method for computer prediction of functional sites in nucleotide sequences, based on the fractal representation, is presented. Fractal representation of set of sequences (FRS) provides simple way for generating recognitions matrix of functionally similar sequences and simple estimations of its efficiency for searching homologous regions in new sequences. Other advantages of the method are absence of the necessity of sequences alignment during generating based set and searching new homologous regions and small CPU time. Usage of the method illustrated for searching globin and histone genes, for ALU repeats in human genome and long terminal repeats in virus genome.     

The effect of him mutations characterized by enhanced induced mutagenesis on the spontaneous mitotic recombination in the ADE2 gene in Saccharomyces cerevisiae

We have studied the influence of him1, him2, him3 and himX mutations on the frequency of spontaneous mitotic gene conversion in the yeast Saccharomyces cerevisiae using the set of heteroallelic combinations in the ADE2 gene. Data obtained on the HIM/HIM, him/him homozygotes and HIM/him heterozygotes indicate that the him1 mutation is recessive with respect to conversion, whereas the him2, him3 and himX mutations are semidominant. Gene conversion was increased in the majority of heteroalleles of mutant diploids him1/him1. On the contrary, the him2, him3 and himX mutants have hypo-rec phenotypes on mitotic conversion. The him mutations do not affect some heteroalleles, moreover, for some heteroalleles, the effects of the him mutations was opposite. On the basis of the sum of genetical data and, particularly, of conversion event pattern in the him mutants, we suggest that him mutations analysed affect the repair pathway for mismatch correction.     

Analysis of the mechanism of the fixation of membrane structures using osmium tetroxide. II. An electron microscopic and x-ray structural study of myelin frozen and lyophilized at low temperature]

Comparative electron microscope and X-ray studies were made on the frog sciatic nerve myelin after freeze-drying technique. The specimens were fixed with OsO4 before and after freeze-drying. In the latter case, osmium was used as a hydrophobic solution (OsO4 in CCl4), or in the high vacuum during osmium sublimation. The results obtained in this study do not fit in the accepted mechanism operating during osmium fixation of membranes. Another mechanism is proposed by the authors, and the problem of osmium localization within the space of the myelin repeated unit is discussed.     

Parameters characterizing the productivity of an inoculum of the producer of oleandomycin

The possibility of using different criteria for estimation of the productivity of the vegetative inoculum of the oleandomycin-producing organism was studied with a purpose of providing maximum levels of the biosynthetic process. The use of the criteria of the culture respiration intensity and accumulation of the physiologically active mycelium as the main ones was shown to be advisable. The quantity and distribution of the mycelium in samples and the time course of changes in active acidity of the medium during the inoculum growth may be used as auxiliary criteria.
