Efficacy of enterocin S760 in treatment of mice with anthrax infection due to Bacillus anthracis M-71

The therapeutic efficacy of enterocin S760, a broad spectrum antimicrobial peptide produced by Enterococcus faecium LWP760 was tested on mice infected with Bacillus anthracis M-71 to induce anthrax (second Tsenkovsky's vaccine). Intraperitoneal four-, two- or one-fold administration of the peptide in a dose of 25 mg/kg for 10 days for prophylactic (1 hour after the contamination) and therapeutic (24 hours after the contamination) purposes prevented or cured the infection in 90-100% of the mice versus the 100-percent lethality in the control (untreated animals). The antimicrobial activity of enterocin S760 against B. anthracis M-71 in vivo correlated with activity in vitro. Enterocin S760 is considered a novel promising antimicrobial for the treatment of grampositive and gramnegative infections.     

cDNA Cloning and Characterization ofNpap60:A Novel Rat Nuclear Pore-Associated Protein with an Unusual Subcellular Localization during Male Germ Cell Differentiation

We have cloned and characterized a cDNA,Npap60,encoding a rat nuclear pore-associated protein. The 3-kb cDNA was obtained by antibody screening of a rat testis expression library. The predicted NPAP60 contains 381 amino acids with a composition of 25.6% charged residues and is highly hydrophilic. TheNpap60gene appears to be conserved in mouse, rat, and human. Immunofluorescence studies with anti-NPAP60 fusion protein antibody show that the NPAP60 protein colocalizes with nuclear pore complexes in RAT1A cells. The expression ofNpap60is about 10–20 times higher in rat testis than in somatic tissues. The subcellular localization of NPAP60 protein changes dramatically during male germ cell differentiation, from nuclear pore complex-like staining in spermatocytes to whole nucleus staining in spermatids and finally to a nuclear surface staining in mature spermatozoa. These changes are temporally and spatially related to nuclear reorganization during male germ cell differentiation.     

Effect of Glutamine Synthetase Gene Overexpression in Birch (<Emphasis Type="Italic">Betula pubescens</Emphasis>) Plants on Auxin Content and Rooting in vitro

The effects of transformation of downy birch (Betula pubescens Ehrh.) with the GS1 gene encoding the cytosolic form of glutamine synthetase on the rooting of plants in vitro was studied. The transgenic plants had an elevated content of glutamine as well as glutamic and aspartic acids and rooted more rapidly than the control plants. Rooting on a medium containing the glutamine synthetase inhibitor phosphinothricin prevented the accumulation of auxin in birch plants carrying the GS1 gene, indicating the involvement of this enzyme in raising the level of auxins in the transgenic plants. The correlation between the increase in the auxin levels in the transgenic plants carrying the glutamine synthetase gene and the increase in the rooting rate is shown for the first time.

A study is carried out on the distribution of ABO, MN, Rh, P and Le blood groups systems, haptoglobins, and common anthropogenetic features in the population of two villages of the Samarkand district. The examined samples have appeared to be heterogenous for ABO, Rh and P systems. For MN and Le unification is possible. The summary data on the allotment of phenotypes are as follows: (%) 0-30.2, A-35.3, B-26.2, AB-8.3, M-33.9, N-26.2, MN-39.9, Rh- -1.5, P-78.6, Le(a-b+)-33, Le (a+b-)-22.2, Le(a-b-)-34.2, Le(a+b+)-10.6. Gene frequencies have are calculated. The occurrence with regards to haptoglobins in both the villages is the same. The summary data are as follows: Hp1=1-4.8, Hp1=2-36.2, Hp2=2-59%. Gene frequencies have been calculated. In the population of the village Karakent there is a change of the frequency in many features of dermatoglyphics. The frequencies of anthropogenetic features do not differ between the villages. The summary data are the following: the frequency of theelbow type of hand is 86.9%, the right type of "hand clasping"-55.0%, the right type of "arm folding"-50.4%, righthandedness-99.7%. The occurence of square lobe of the ear is 21%, spliced one is 26.2%, the hanging down is 52.7%. The frequency of the gene insensitivity for PTC taste is 0.47 in one village and 0.53 in the other.

Small doses of X-ray irradiation activate the NO-synthase component of the nitric oxide cycle

It has been shown that chronic X-ray irradiation, (CRI), activates the formation of NO in rats. This is apparent in the increase in the level of NO2- in the blood plasma from 12.59 +/- 1.7 to 39.79 +/- 2.9 nmol/ml after 10 days of irradiation. On the 20 and 30 day of CRI, the level of NO2(-)- was 21.05 +/- 1.2 and 30.73 +/- 1.9 nmol/ml respectively. The changes in the NO-synthase component of the NO cycle were accompanied by a decrease in the osmotic resistance of the erythrocytes and the nitritreductase activity of hemoglobin.     

Changes in fatty acids contents and growth characteristics in transformed oilseed rape (Brassica napus)

Spring oilseed rapeBrassica napus L. ssp.oleifera cv. HM-81 was transformed with T_L-DNA of the Ri plasmid of the agropine strainAgrobacterium rhizogenes 15834. Selfed progenies (R₂ and R₃ generations) were studied for changes in values of growth characteristics and fatty acids contents. Transformants are ‘homozygous’ for T_L-DNA. Both generations of transformants differed significantly from the nontransformed control plants in reduced length, lower number of pods per plant, lower total mass of seeds and the higher number of branches. The contents of palmitic, linoleic and linolenic acids were significantly higher in transformants when compared with the control. On the contrary, the contents of both stearic and oleic acids were in most of transformants significantly lower. Only traces of erucic acid (less than 0.05 % ) were found, both in transformed and nontransformed plants.
