The synthesis of two C-1 analogues of pancratistatin has been accomplished in 17 steps from bromobenzene. The key steps involved the enzymatic dihydroxylation, regioselective opening of epoxyaziridine 9 with the alane derived from 8, a solid-state silica-gel-catalyzed intramolecular opening of aziridine to produce phenanthrene 13 whose oxidative cleavage and recyclization provided the full skeleton of the Amaryllidaceae constituents. The new analogues 5 and 6 exhibited promising activity in several human cancer cell lines.  相似文献

In order to confirm the taxonomic position of environmental strains determined based on their biochemical, cultural, and morphological characteristics, molecular genetic identification was carried out. A number of problems in identification of microorganisms were shown to be associated with contamination of the cultures in the course of isolation. Advantages of a comprehensive approach—combining 16S rRNA gene sequencing with a set of biochemical, cultural, and morphological parameters—for identification of microorganisms isolated from environmental objects and clinical samples are discussed.  相似文献

A comprehensive study with the assessment of reactive responses to motor tasks was performed in nine patients with a tumor localized in the frontal divisions of the brain using two methodological approaches: functional magnetic resonance imaging (fMRI) and EEG. The data obtained were compared to the results of a similar study on 12 healthy subjects. It was established that cerebral pathology was associated with disorders of functional specialization and an increase in the diffuse component of reactivity. The fMRI responses were characterized by greater intactness compared to the EEG parameters of reactive changes. These features are especially marked when an afferent stimulus is sent to the damaged hemisphere. The characteristics of the involvement of individual EEG bands in the formation of motor responses and changes in the fMRI response topography are determined by the degree of cerebral dysfunction reflected by the pattern of baseline EEG reorganization and the severity of the motor defect. The predominant increase in the coherence of slow rhythms in the damaged hemisphere irrespective of the target of the afferent stimulus in patients with severe cerebral dysfunction reflects the dominant formation of a pathological focus and is indicative of a greater, compared to healthy subjects, involvement of deep brain structures in the reactive process, which is confirmed by the fMRI data.  相似文献

This work is devoted to personal identification of a pilot who perished during World War II in 1941. As a result of search works near the village of Sambek, participants of the search association Mius-Front found remains of a downed plane. We carried out complex genetic analysis of the mitochondrial and autosomal DNA isolated from the remains found among the plane fragments. For personal identification of the remains, recent relatives of crew members of the plane were found, who donated the biological material for comparative research. Owing to the analysis of DNA of the native niece of one of the pilots and the remains of the unknown person, it was possible to establish that these remains belong to Junior Sergeant Dzyuba Vladimir Romanovich, who was considered to be missing in action since October 17, 1941.  相似文献

Contact traces on a human body are a widespread object investigated in cases involving criminal acts against a person. In such cases, interpretation of the results of DNA analysis, as the most evident method, is accompanied by considerable difficulties. This paper shows the possibility of detecting genetic material in the sweat and grease deposits left by the donor on the skin surface of a living recipient. The comparative study of DNA amount in the sweat and grease substance on the surface of the skin of different areas of the body of men and women is carried out. Typing of microsatellite loci of autosomal DNA and Y chromosome in swabs from places of contact of donor males with integuments of recipients of both sexes is carried out. No statistically significant differences in the DNA amount of sweat and grease substance depending on its localization on human body are revealed. The DNA study of sweat and grease substance left by donor men on the skin of both men and women in half of cases reveals allelic combinations inherent in both the donor and the recipient. The results obtained indicate equal chances of detecting the DNA of contacting individuals.  相似文献

This report describes the development, optimization, and implementation of a miniaturized cell-based assay for the identification of small-molecule insulin mimetics and potentiators. Cell-based assays are attractive formats for compound screening because they present the molecular targets in their cellular environment. A fluorescence resonance energy transfer (FRET) cell-based assay that measures the insulin-dependent colocalization of Akt2 fused with either cyan fluorescent protein or yellow fluorescent protein to the cellular membrane was developed. This ratiometric FRET assay was miniaturized into a robust, yet sensitive 3456-well nanoplate assay with Z' factors of approximately 0.6 despite a very small assay window (less than twofold full activation with insulin). The FRET assay was used for primary screening of a large compound collection for insulin-receptor agonists and potentiators. To prioritize compounds for further development, primary hits were tested in two additional assays, a biochemical time-resolved fluorescence resonance energy transfer assay to measure insulin-receptor phosphorylation and a translocation-based imaging assay. Results from the three assays were combined to yield 11 compounds as potential leads for the development of insulin mimetics or potentiators.  相似文献

The D4oop of mtDNA is a noncoding locus actively used as an individualizing marker in molecular genetic research. Uneven distribution of SNPs in the D-loop suggests that the functional load within this region is irregular. The nucleotide sequence analysis was used to evaluate the structural and functional role of various D-loop sites of a single individual’s mtDNA an d their importance in terms of phylogenetic conservatism. The role of duplication of various D-loop elements (TAS, ETAS, CSB elements) in increasing the reliability of the mtDNA replication initiation and termination is discussed.  相似文献
