Two methanotrophic bacteria with optimum growth temperatures above 40° C were isolated. Thermotolerant strain LK6 was isolated
from agricultural soil, and the moderately thermophilic strain OR2 was isolated from the effluent of an underground hot spring.
When compared to the described thermophilic methanotrophs Methylococcus capsulatus and Methylococcus thermophilus, these strains are phenotypically similar to Methylococcus thermophilus. However, their 16S rRNA gene sequences are markedly different from the sequence of Methylococcus thermophilus (∼ 8% divergence) and, together with Methylomonas gracilis, they form a distinct, new genus within the γ-subgroup of the Proteobacteria related to extant Type I methanotrophs. phenotypic characterisation showed that the isolates possess particulate methane monooxygenase (pMMO) but do not contain soluble
methane monooxygenase. The nucleotide sequence of a gene encoding pMMO (pmoA) was determined for both isolates and for Methylomonas gracilis. PmoA sequence comparisons confirmed the monophyletic nature of this newly recognised group of thermophilic methanotrophs
and their relationship to previously described Type I methanotrophs. We propose that strains OR2 and LK6, together with the
misclassified thermophilic strains Methylomonas gracilis VKM-14LT and Methylococcus thermophilus IMV-B3122, comprise a new genus of thermophilic methanotrophs, Methylocaldum gen. nov., containing three new species: Methylocaldum szegediense, Methylocaldum tepidum and Methylocaldum gracile.

The possibility has been demonstrated of the use of calibrated melamine-formaldehyde latex for studying phagocytosis as an object not undergoing intracellular digestion. Latex was discovered to be actively phagocytized by macrophages and to exert no toxic action on them. A study was made of the time course of changes in the color and brightness of the fluorescence of the latex phagocytized particles in macrophages intravitally fluorochrominated by acridine orange. These changes were demonstrated to be analogous to those observed previously during phagocytosis of bacteria and other objects. The data obtained demonstrate once more the transition of the fluorescent complex from lysosomes to phagosomes and the lack of the relationship of these changes with intracellular death and digestion of the phagocytized objects. Size and shape of glycine receptor clusters in a central neuron exhibit a somato-dendritic gradient 总被引:4,自引:0,他引:4
The overall surface disposition of receptors for the neurotransmitter glycine was examined in situ on the teleost Mauthner cell. For this purpose, a monoclonal antibody specific for the glycine receptor was used in immunofluorescence experiments in association with the technique of confocal microscopy. Previous work had shown that on the Mauthner cell and other neurons, receptors for this transmitter are concentrated in discrete microdomains apposed to single presynaptic terminals. We now report that the size and the shape of these clusters depend on their cellular location, that is, their area increases regularly from the soma to the tip of the dendrites. Conversely, the number of clusters per unit area declines so that the final proportion of immunoreactive membrane remains constant at any portion of the cell. These results raise the question of the functional consequences of such an organized subcellular distribution of receptors. 相似文献
