The effects of the D137L/G126R double mutation in the central part of the tropomyosin α-chain via the simultaneous replacement of two highly conserved non-canonical residues, viz., Asp137 and Gly126, by canonical residues Leu and Arg, respectively, on the properties of the αβ-tropomyosin heterodimer have been studied. It has been shown using circular dichroism that this mutation substantially increases the thermal stability of αβ-tropomyosin heterodimers, which, nevertheless, remains lower than that of αα-tropomyosin homodimers with these mutations in both α-chains. The stability of tropomyosin complexes with F-actin has also been studied by measuring the temperature dependences of their dissociation, which is detected by a decrease in light scattering. It has been revealed that αβ-tropomyosin heterodimers carrying the D137L/G126R mutation in the α-chain dissociate from the surface of actin filaments at a higher temperature than ββ-homodimers but at a lower temperature than αα-homodimers with these mutations in both α-chains. It has also been shown using the in vitro motility assay that D137L/G126R substitution in the α-chain increases the sliding velocity of regulated actin filaments in the case of αα-homodimers, while it noticeably decreases the velocity in the case of αβ-tropomyosin heterodimers. Thus, we can conclude that mutations in one of the chains of the tropomyosin dimeric molecule may have different effects on the properties of tropomyosin homodimers and heterodimers.  相似文献

The influence of the hypertonic (3%) solution of mannitol on the process of the excitability recovery (in the near necrotic zone) appeared as a result of the necroses tissue effect on the ventricle has been investigated in the experiments on the isolated frog's heart. The analogous perfusion was conducted by Ringer solution in the control experiments. The process of isopotential phase S-T recovery was practically the same at heart perfusion with Ringer solution or with mannitol. Simultaneous registration of ventricular mechanogram showed the pronounced negative inotropic effect of mannitol. It is suggested that the positive effect of mannitol which has been noted under clinic conditions during the treatment of ischemic disease, doesn't connect with the effect of hypertonic solution on the cells excitability of periinfarcted (near necrotic) zone.  相似文献

Exposure to cocaine generates silent synapses in the nucleus accumbens (NAc), whose eventual unsilencing/maturation by recruitment of calcium‐permeable AMPA‐type glutamate receptors (CP‐AMPARs) after drug withdrawal results in profound remodeling of NAc neuro‐circuits. Silent synapse‐based NAc remodeling was shown to be critical for several drug‐induced behaviors, but its role in acquisition and retention of the association between drug rewarding effects and drug‐associated contexts has remained unclear. Here, we find that the postsynaptic proteins PSD‐93, PSD‐95, and SAP102 differentially regulate excitatory synapse properties in the NAc. Mice deficient for either of these scaffold proteins exhibit distinct maturation patterns of silent synapses and thus provided instructive animal models to examine the role of NAc silent synapse maturation in cocaine‐conditioned place preference (CPP). Wild‐type and knockout mice alike all acquired cocaine‐CPP and exhibited increased levels of silent synapses after drug‐context conditioning. However, the mice differed in CPP retention and CP‐AMPAR incorporation. Collectively, our results indicate that CP‐AMPAR‐mediated maturation of silent synapses in the NAc is a signature of drug–context association, but this maturation is not required for establishing or retaining cocaine‐CPP.  相似文献

The distribution of the glyprolines Pro-Gly-Pro and Thr-Lys-Pro-Arg-Pro-Gly-Pro (Selanc) was analyzed and compared in tissues of rat organs after different ways of their administration using the peptides uniformly labeled with tritium. Comparative data on changes in concentrations of the peptides in the rat organs after their intraperitoneal, intranasal, intragastric, and intravenous administration are given. The intranasal administration of both peptides was shown to be optimal for the delivery of glyproline molecules in the CNS. A high affinity of the studied glyprolines for gastric tissues was found for all the ways of their administration. We suggest that a high efficiency of action of glyprolines on homeostasis of the gastric mucous tunic was partially provided by accumulation of these peptides (to high concentrations) in gastric tissues.  相似文献

The problem of an electric signal propagating over a thin layer modeling the cell membrane is considered in the electrostatic approximation. At both sides of the membrane there are bulk conductors with different properties. Various types of boundary conditions are examined. Solutions are obtained for a cylindrical fiber and planar layer and compared with the relationships used in the classical Hodgkin-Huxley theory. It is shown that agreement with the cable theory is obtained only if the system is fully symmetrical and the near-membrane layers are thin.  相似文献

Skeletal muscles power body movement by converting free energy of ATP hydrolysis into mechanical work. During the landing phase of running or jumping some activated skeletal muscles are subjected to stretch. Upon stretch they absorb body energy quickly and effectively thus protecting joints and bones from impact damage. This is achieved because during lengthening, skeletal muscle bears higher force and has higher instantaneous stiffness than during isometric contraction, and yet consumes very little ATP. We wish to understand how the actomyosin molecules change their structure and interaction to implement these physiologically useful mechanical and thermodynamical properties. We monitored changes in the low angle x-ray diffraction pattern of rabbit skeletal muscle fibers during ramp stretch compared to those during isometric contraction at physiological temperature using synchrotron radiation. The intensities of the off-meridional layer lines and fine interference structure of the meridional M3 myosin x-ray reflection were resolved. Mechanical and structural data show that upon stretch the fraction of actin-bound myosin heads is higher than during isometric contraction. On the other hand, the intensities of the actin layer lines are lower than during isometric contraction. Taken together, these results suggest that during stretch, a significant fraction of actin-bound heads is bound non-stereo-specifically, i.e. they are disordered azimuthally although stiff axially. As the strong or stereo-specific myosin binding to actin is necessary for actin activation of the myosin ATPase, this finding explains the low metabolic cost of energy absorption by muscle during the landing phase of locomotion.  相似文献
