Virulence of parasites in hosts under environmental stress: experiments with anoxia and starvation 总被引:4,自引:0,他引:4
Most environments periodically impose severe stress that may cause high mortality and alter population structure, for example, by removing sick and old individuals. We examined how anoxic conditions and starvation of the host affect virulence of two closely related trematode parasites, Rhipidocotyle campanula and R. fennica . These parasites differ by prevalence of infection and by exploitation rate of individual hosts (freshwater clam, Anodonta piscinalis ). Infection by R. campanula is rare (<5% prevalence of infection) and destroys on average 90% of the gonad tissue of the individual host. Infection by R. fennica is more common (20–60% prevalence of infection) and leads to on average 30% gonad destruction. In the end, both infections lead to host infertility. We predicted that R. campanula induces higher host mortality than R. fennica under host stress. In two laboratory experiments, we exposed naturally-infected and uninfected clams to anoxia and to starvation. Anoxia occasionally takes place during winter in eutrophic lakes, while some degree of starvation should occur seasonally. We found that mortality rate of clams was much higher under anoxia than under starvation, and that infection increased mortality rate under both types of host stress. As predicted, R. campanula induced higher host mortality than R. fennica . Host survival was population-specific, suggesting that clams of different origins carried different amount of energy reserves. Severe environmental perturbation may remove R. campanula infected individuals from the host population, but recolonization from the fish host is likely to prevent extinction of the parasite suprapopulation. The observed high host mortality induced by R. campanula may be one ecological explanation for the consistently lower prevalence of infection of R. campanula when compared to R. fennica . 相似文献

Manganese is essential for normal development and activity of the nervous tissue. Mn2+ ions are involved in protein synthesis and may prevent free radical damage. Since it is now established that alcohol degradation may produce free radicals, we studied the effect of Mn2+ on ethanol induced alterations using cultured nerve cells as an experimental model of the central nervous system. Neurons and glial cells were cultured from rat brain cortex; a tumoral rat glial cell line (C6) was also examined. We measured enzymatic markers of nerve cell maturation (enolase, glutamine synthetase) and superoxide dismutase, a scavenger of free radicals; all these enzymes being activated by Mn2+ ions. Only for the glial cell types an alcohol antagonizing effect was found when Mn2+ was combined with ethanol. Neurons were not sensitive to that Mn2+ effect. 相似文献

In marine ecosystems, the study of trophic relationships has extensively benefited from the development of stable isotope analyses (SIA) as dietary tracers. SIA are particularly useful in elucidating the structure of deep sea food webs given the constraints involved in obtaining gut‐content data from deep trawling. We used carbon and nitrogen stable isotope analyses and Stable Isotope Bayesian Ellipses in R (SIBER) and Stable Isotope Analysis in R (SIAR) routines, to determine the trophic ecology of five deep‐sea fishes from the upper continental slope of the Celtic Sea. SIA made it possible to deduce some general tendencies in food‐web structure and species trophic interactions and confirmed diet determined by gut‐content analysis for the same species, in other ecoregions. More specifically, mixing models revealed that the deep sea species considered are omnivorous and are able to feed on all the sampled taxa. Based on isotopic ratio, no clear differences in fish diet could be detected from one species to another except for rabbit fish, which has benthic affinities. Three species, blackbelly rosefish, greater forkbeard and softhead grenadier showed overlapping isotopic niches. This study is the first attempt to describe the trophic ecology of deep sea species on the Celtic Sea upper continental slope. In the context of the development of ecosystem integrated modeling approaches for managing fisheries in the Celtic sea, and considering the vulnerability of deep‐water species, improving the knowledge on the trophic ecology of these local species is of importance in order to allow their sustainable exploitation. 相似文献

The preparation of crosslinked aggregates of pancreatic porcine lipase (PPL‐CLEA) was systematically studied, evaluating the influence of three precipitants and two crosslinking agents, as well as the use of soy protein as an alternative feeder protein on the catalytic properties and stability of the immobilized PPL. Standard CLEAs showed a global yield (CLEA’ observed activity/offered total activity) of less than 4%, whereas with the addition of soy protein (PPL:soy protein mass ratio of 1:3) the global yield was approximately fivefold higher. The CLEA of PPL prepared with soy protein as feeder (PPL:soy protein mass ratio of 1:3) and glutaraldehyde as crosslinking reagent (10 μmol of aldehyde groups/mg of total protein) was more active mainly because of the reduced enzyme leaching in the washing step. This CLEA, named PPL‐SOY‐CLEA, had an immobilization yield around 60% and an expressed activity around 40%. In the ethanolysis of soybean oil, the PPL‐SOY‐CLEA yielded maximum fatty acid ethyl ester (FAEE) concentration around 12‐fold higher than that achieved using soluble PPL (34 h reaction at 30°C, 300 rpm stirring, soybean oil/ethanol molar ratio of 1:5) with an enzyme load around 2‐fold lower (very likely due to free enzyme inactivation). The operational stability of the PPL‐SOY‐CLEA in the ethanolysis of soybean oil in a vortex flow type reactor showed that FAEE yield was higher than 50% during ten reaction cycles of 24 h. This reactor configuration may be an attractive alternative to the conventional stirred reactors for biotransformations in industrial plants using carrier‐free biocatalysts. © 2018 American Institute of Chemical Engineers Biotechnol. Mammalian cells deploy autophagy to defend their cytosol against bacterial invaders. Anti‐bacterial autophagy relies on the core autophagy machinery, cargo receptors, and “eat‐me” signals such as galectin‐8 and ubiquitin that label bacteria as autophagy cargo. Anti‐bacterial autophagy also requires the kinase TBK1, whose role in autophagy has remained enigmatic. Here we show that recruitment of WIPI2, itself essential for anti‐bacterial autophagy, is dependent on the localization of catalytically active TBK1 to the vicinity of cytosolic bacteria. Experimental manipulation of TBK1 recruitment revealed that engagement of TBK1 with any of a variety of Salmonella‐associated “eat‐me” signals, including host‐derived glycans and K48‐ and K63‐linked ubiquitin chains, suffices to restrict bacterial proliferation. Promiscuity in recruiting TBK1 via independent signals may buffer TBK1 functionality from potential bacterial antagonism and thus be of evolutionary advantage to the host. 相似文献

In aquatic ecosystems invasive species are among the most important threats to biodiversity worldwide. Understanding the dispersal mechanisms of aquatic invaders is very important for protection and management of vulnerable water bodies. Here we ask how recreational boats that are transported overland could contribute to the dispersal of invasive zebra mussels among lakes in Switzerland. Using a questionnaire sent to registered boat owners, we surveyed properties of transported boats and collected information on self-reported mussel fouling and transport activities of boat owners. We also sampled boat hulls at launching ramps and harbors for biofouling invertebrates. Boats that were kept seasonally or year-round in water were found to have high vector potential with mussel fouling rates of more than 40 %. However, only about 6 % of boats belonging to these groups were transported overland to other water bodies. Considering that approximately 100,000 recreational boats are registered in Switzerland, we estimated that every year around 1400 boats fouled with mussels are transported overland. Such boats pose a high risk of distributing zebra mussels between water bodies. Our results suggest that there is a considerable risk that recreational boats may spread new fouling species to all navigable water bodies within the study area. We speculate that one such species could be the quagga mussel, which has not yet invaded lakes in Switzerland. On a more positive note, our study has identified the group of high-risk boats so that possible control measures would only affect a relatively small number of boat owners. 相似文献
