The mitochondrial biogenesis and energy expenditure regulator, PGC-1α, has been previously reported to be induced in the white adipose tissue (WAT) and liver of mice overexpressing spermidine/spermine N (1)-acetyltransferase (SSAT). The activation of PGC-1α in these mouse lines leads to increased number of mitochondria, improved glucose homeostasis, reduced WAT mass and elevated basal metabolic rate. The constant activation of polyamine catabolism produces a futile cycle that greatly reduces the ATP pools and induces 5'-AMP-activated protein kinase (AMPK), which in turn activates PGC-1α in WAT. In this study, we have investigated the effects of activated polyamine catabolism on the glucose and energy metabolisms when targeted to specific tissues. For that we used a mouse line overexpressing SSAT under the endogenous SSAT promoter, an inducible SSAT overexpressing mouse model using the metallothionein I promoter (MT-SSAT), and a mouse model with WAT-specific SSAT overexpression (aP2-SSAT). The results demonstrated that WAT-specific SSAT overexpression was sufficient to increase the number of mitochondria, reduce WAT mass and protect the mice from high-fat diet-induced obesity. However, the improvement in the glucose homeostasis is achieved only when polyamine catabolism is enhanced at the same time in the liver and skeletal muscle. Our results suggest that the tissue-specific targeting of activated polyamine catabolism may reveal new possibilities for the development of drugs boosting mitochondrial metabolism and eventually for treatment of obesity and type 2 diabetes.  相似文献

Excitotoxicity through stimulation of N-methyl-d-aspartate (NMDA) receptors contributes to neuronal death in brain injuries, including stroke. Several lines of evidence suggest a role for protein kinase C (PKC) isoforms in NMDA excitotoxicity. We have used specific peptide inhibitors of classical PKCs (alpha, beta, and gamma), novel PKCs delta and epsilon, and an atypical PKCzeta in order to delineate which subspecies are involved in NMDA-induced cell death. Neuronal cell cultures were prepared from 15-day-old mouse embryos and plated onto the astrocytic monolayer. After 2 weeks in vitro the neurons were exposed to 100 micro m NMDA for 5 min, and 24 h later the cell viability was examined by measuring the lactate dehydrogenase release and bis-benzimide staining. While inhibitors directed to classical (alpha, beta, and gamma) or novel PKCs (delta or epsilon) had no effect, the PKCzeta inhibitor completely prevented the NMDA-induced necrotic neuronal death. Confocal microscopy confirmed that NMDA induced PKCzeta translocation, which was blocked by the PKCzeta inhibitor. The NMDA-induced changes in intracellular free Ca2+ were not affected by the peptides. In situ hybridization experiments demonstrated that PKCzeta mRNA is induced in the cortex after focal brain ischemia. Altogether, the results indicate that PKCzeta activation is a downstream signal in NMDA-induced death of cortical neurons.  相似文献

Significant seasonal differences in intra- and interorgan prevalence, abundance and distribution of rodlet cells (RC) were observed in freshwater bream caught in May and August 1997 from a PCB-polluted lake, Lake Kernaala, Finland. In May, the majority of RCs in liver were seen in blood vessel endothelium, while in August most cells were located in the epithelium of the intrahepatic biliary ducts. In kidney, a significant increase in RCs in the columnar epithelium of the collecting ducts was observed from May to August. RCs in spleen were extremely rare in May, but were seen frequently in the endothelium of the splenic blood vessels in August. In gill and intestine, RCs were found in every tissue sample studied, with no statistical differences in their abundance or distribution between seasons. No RCs were seen in gonads. Total numbers of RCs increased dramatically from May to August in kidney and in spleen. The frequency of RCs corresponded strongly with the current phase of parasitic infections in liver, kidney, and spleen. It is concluded that RCs play a role in host defence mechanisms in fish, displaying induced expression at the times of peak parasitic prevalences.  相似文献

Based on a model of light limited growth, Huisman and Weissing found that in a well mixed water column with constant light supply (energy reaching the water surface), equilibrium growth and competition of phytoplankton for light can be characterised by a critical light intensity at the base of the column (I*out). The present study attempts to give a further insight into this model. We first analyse the dependence of the critical light intensity on four parameters: initial slope of the photosynthesis-intensity (p-I) curve, maximal photosynthetic rate, the light-saturated parameter Ikand specific carbon loss rate. Increases in the first two parameters tend to reduce the critical light intensity and increases in the last two tend to increase the critical light intensity. Then we analyse the performance of a model under variable light supply with a time-scale of 1 day (24 hr). Within this time-scale, the critical light intensity changes with time. However, the equilibrium growth and the outcome of competition for light can be adequately characterised by critical light extinction defined as the upper limit of total light extinction due to both biomass and non-living matter in the water column. Under constant light supply, a critical light intensity uniquely corresponds to a critical light extinction. Therefore, critical light extinction can be utilised to predict the equilibrium growth and the outcome of competition under both constant and variable light supply. By changing the maximal light supply at noon, seasonal succession of species composition of communities is investigated. The possible effect of two typical photoresponses, photoadaptation and photoinhibition, on growth and competiton are discussed. Copyright 1999 Academic Press.  相似文献

The history of the generic and sectional concepts in the traditional moss family Mniaceae is summarised. The division of the Mniaceae into nine genera has been generally accepted. A phylogenetic analysis, based on 35 Mniaceae samples as ingroup and one Leptostomataceae and three Bryaceae samples as outgroups was completed. A total of 113 new sequences were obtained and 4139 aligned characters were included in the combined dataset. Bayesian inferences based on the combined data set showed five well supported clades, which, as a whole, form the monophyletic family Mniaceae. The number of genera in the traditional Mniaceae is 9 and the number of species is 74. All the genera appear monophyletic except Plagiomnium; accepting Orthomnion at the generic level makes Plagiomnium paraphyletic. The principle that only monophyletic taxa are allowed and may be named necessitates the uniting of Plagiomnium and Orthomnion. The necessary combinations are made. The taxonomy of the genera and sections is discussed. Rhizomnium section Macromnium T.J.Kop. & Y.Sun, sect. nov. is described and the new combination Rhizomnium chlorophyllosum (Kindb.) T.J.Kop. is made. The controversy of paraphyly – monophyly in accepting and naming taxa is discussed and Plagiomnium versus Orthomnion given as an example. All the present genera and species seem to have diverged in the area of the Laurasian supercontinent and the speciation of most genera took place in the temperate forested areas of western and eastern North America, Europe and south-eastern Asia.  相似文献

That larger areas will typically host more diverse ecological assemblages than small ones has been regarded as one of the few fundamental ‘laws’ in ecology. Yet, area may affect not only species diversity, but also the trophic structure of the local ecological assemblage. In this context, recent theory on trophic island biogeography offers two clear‐cut predictions: that the slope of the species–area relationship should increase with trophic rank, and that food chain length (i.e. the number of trophic levels) should increase with area. These predictions have rarely been verified in terrestrial systems. To offer a stringent test of key theory, we focused on local food chains consisting of trophic specialists: plants, lepidopteran herbivores, and their primary and secondary parasitoids. For each of these four trophic levels, we surveyed species richness across a set of 20 off‐shore continental islands spanning a hundred‐fold range in size. We then tested three specific hypotheses: that species richness is affected by island size, that the slope of the species–area curve is related to trophic rank, and that such differences in slope translate into variation in food chain length with island size. Consistent with these predictions, estimates of the species–area slope steepened from plants through herbivores and primary parasitoids to secondary parasitoids. As a result of the elevated sensitivity of top consumers to island size, food chain length decreased from large to small islands. Since island size did not detectably affect the ratio between generalists and specialists among either herbivores (polyphages vs oligophages) or parasitoids (idiobionts vs koinobionts), the patterns observed seemed more reflective of changes in the overall number of nodes and levels in local food webs than of changes in their linking structure. Overall, our results support the trophic‐level hypothesis of island biogeography. Per extension, they suggest that landscape modification may imperil food web integrity and vital biotic interactions.  相似文献

Objective

To determine whether there are differences in age and sex distribution and presence of comorbidities between participants included in randomized controlled trials of acetylcholinesterase inhibitors and nationwide cohort of persons with Alzheimer’s disease.

Methods

PubMed, Scopus and Cochrane Library databases were searched for original articles from their inception to January 4, 2015. Double-blind randomized controlled trials with donepezil, rivastigmine or galantamine compared to placebo in participants with Alzheimer’s disease were included. Data from a nationwide cohort of persons with clinically verified diagnoses of Alzheimer’s disease was defined as a reference population.

Results

128 full-text articles were assessed for eligibility, 31 of them fulfilled criteria. Mean age of participants in randomized controlled trials (n = 15,032) was 5.8 years lower (95% CI 5.7 to 5.9, P < 0.001), compared to the mean age of 79.7 years in the reference population with Alzheimer’s disease (n = 28,093). Most of the articles did not report age distribution of participants. The proportion of women was 63.2% (9,475/14,991) in randomized controlled trials and 67.8% (19,043/28,093) (P < 0.001) in the reference population. Information on comorbidities and use of concomitant drugs were lacking or poorly reported in most articles.

Conclusions

There is a discrepancy between participants in randomized controlled trials of acetylcholinesterase inhibitors and real-life population with Alzheimer’s disease. Participants in randomized controlled trials were significantly younger. Further, more detailed reporting of age distribution, comorbidities and concomitant drugs would be important information for clinicians when evaluating conclusions from randomized controlled trials to real-life practice. The existing recommendations of inclusion of older people should be followed to ensure safe pharmacotherapy for older people.  相似文献
