Summary Samples of leaves and fruits exploited as food items byTrachypithecus auratus sondaicus were analysed for nitrogen content, acid detergent fibre (ADF), pepsin cellulase digestibility (CDIG), condensed tannins (CT), total phenolics (TP) and protein precipitation capacity (PP) and compared with fruits and leaves not eaten. Differences in chemical measures for items eaten and not eaten were not statistically significant but trends indicate that leaves may have been selected for their lower fibre content and greater digestibility. Fruits eaten also had a higher mean level of CDIG and lower mean level of ADF than fruits not eaten but these measures are not considered to be of major importance in fruit selection as CDIG was lower and ADF higher in fruits eaten than in leaves eaten. Levels of CT, TP and PP capacity were higher in fruits eaten than in fruits not eaten but lower in leaves eaten than in leaves not eaten. The role of tannins and phenolics in food selection is discussed. Leaves (and fruits) were not strongly selected on the basis of protein content. Approximately half the dietary intake ofT. auratus sondaicus was leaves, a protein-rich food source. Possibly, protein levels in foliage at Pangandaran were sufficient that selection for this nutrient was not required. A nutrient other than protein (for example, soluble carbohydrates) may have been maximised through food selection. The protein/ADF ratio may provide an indicator of the acceptability of foliage in a habitat as potential food for a colobine. However, this ratio did not govern food selection byT. auratus sondaicus at Pangandaran.  相似文献

Precedemment signalée chez Kerforneidictyum (Pleurodictyforme dévonien), la rare disposition des rides septales suivant un mode tétracoralliaire est décrite chez un Palaeack sp. du Viséen d'Algérie et chez P. cuneiformis subsp. A du Viseen d'Australie. Chez Palaeacis sp. sont mises en evidence une ride septale cardinale forte en position externe opposée à une ride antipode déterminant un plan de symétrie bilatérale; de part et d'autre une zone systématiquement confuse marque l'emplacement des rides alaires. La reconnaissance des périantipodes prête à discussion et ce secteur présente une originalité certaine par rapport à celui des Tétracoralliaires. Chez P. cuneiformis subsp. A, une coupe transversale montre la présence de quatre rides proéminentes qui fusionnent dans I'axe du corallite et I'existence d'une symétrie bilatérale. La position systématique du genre Palaeacis est discutée: son attribution aux Tétracoralliaires est exclue, ancun caractére n'est incompatible avec le statut de Tabulé, la création d'une unité taxon-omique nouvelle regroupant Palaeacis, Trachypsammia et les Pleurodictyformes est envisageable mais actuellement prématurée. ? Palaeacis, Tabulata, Rugosa, rides septales, systématique, relations deparenré, Viséen. The tetracoral pattern of septal ridges described in Kerforneidictyum (Devonian Pleurodictyum-like coral) is scar'ce in the Tabulata, but has been found in Palaeacis sp. from the Visean of Algeria and in P. cuneiformis subsp. A from the Visean of Australia. Palaeacis sp. shows a strong cardinal septal ridge in peripheral position, just opposite the counter ridge. thus creating a plane of bilateral symmetry: an area on both sides. which is always blurred, indicates the position of the alar ridges. counter-lateral ridges are not easily identified and yield differing interpretation. Compared with the Rugosa this area presents distinctive features. In P. cuneiformis subsp. A, a transverse section exhibits four well-developed septal ridges that coalesce in the axis of the corallite with obvious bilateral symmetry. The systematic position of the genus Palaeacis is questioned: Rugosa it is not; it may belong to Tabulata: however, erecting a new higher taxon for Palaeacis, Trachypsammia and other Pleurodictyum-like corals is premature at our present state of knowledge. ? We have determined the nucleotide sequences of two regions within the A+T-rich region of mitochondrial DNA (mtDNA) in the siIII type of Drosophila simulans and the maI type of D. mauritiana. The sequences of the two regions in siIII and maI are almost identical. The sequences include elements corresponding to the type I and type II repeats elements and the T-stretches as reported in D. melanogaster; an approximately 340-bp region (A region) adjacent to the tRNA(Ile) gene includes a part of the type II repeat element, and an approximately 440- bp region (B region) includes a central portion of the A+T-rich region between the type I and type II repeat arrays. Each sequence of the two species was compared with those of D. melanogaster and D. yakuba. The sequences of the A region are relatively well conserved among the four species. The alignment of the two sequences of the B region with those of D. melanogaster and D. yakuba requires numerous insertions/deletions. For both regions, nucleotide differences between D. simulans or D. mauritiana and D. melanogaster are similar to those between the two and D. yakuba. The tendency is obvious in a subregion within the type II repeat element in the A region. These findings suggest that the rate of nucleotide substitution in the subregion is accelerated in the lineage leading to D. melanogaster. Loss of functional constraint in the stem-loop-forming sequence is proposed for this acceleration.   相似文献

Summary The cloacinogenic factor Clo DF13 from Enterobacter cloacae has been transferred to the minicell-producing strain P678-54 of Escherichia coli K12. The data presented show that this Clo DF13 factor segregates into minicells of P678-54 (Clo DF13) and that this factor is the only plasmid, present in these minicells. Proteins from purified P678-54 (Clo DF13) and P678-54 minicells, previously labelled with 14C-amino acids, were compared after electrophoresis on SDS-polyacrylamide gels. From this comparison it appeared that a noninduced Clo DF13 factor directs the synthesis of 4 proteins. The molecular weights of these proteins could be estimated to be about 72000, 32000, 18500 and 12000. In P678-54 (Clo DF13) minicells, one additional Clo DF13 protein was found to be unlabelled. Apparently this protein is not synthesized in P678-54 (Clo DF13) minicells, but is segregated into or is attached to the minicells after being synthesized in the P678-54 (Clo DF13) cells. The molecular weight of this protein is about 62000, which corresponds to the molecular weight of cloacin.  相似文献

Background

Medulloblastoma is the most common malignant brain tumor in children. Despite recent improvements in cure rates, prediction of disease outcome remains a major challenge and survivors suffer from serious therapy-related side-effects. Recent data showed that patients with WNT-activated tumors have a favorable prognosis, suggesting that these patients could be treated less intensively, thereby reducing the side-effects. This illustrates the potential benefits of a robust classification of medulloblastoma patients and a detailed knowledge of associated biological mechanisms.

Methods and Findings

To get a better insight into the molecular biology of medulloblastoma we established mRNA expression profiles of 62 medulloblastomas and analyzed 52 of them also by comparative genomic hybridization (CGH) arrays. Five molecular subtypes were identified, characterized by WNT signaling (A; 9 cases), SHH signaling (B; 15 cases), expression of neuronal differentiation genes (C and D; 16 and 11 cases, respectively) or photoreceptor genes (D and E; both 11 cases). Mutations in β-catenin were identified in all 9 type A tumors, but not in any other tumor. PTCH1 mutations were exclusively identified in type B tumors. CGH analysis identified several fully or partly subtype-specific chromosomal aberrations. Monosomy of chromosome 6 occurred only in type A tumors, loss of 9q mostly occurred in type B tumors, whereas chromosome 17 aberrations, most common in medulloblastoma, were strongly associated with type C or D tumors. Loss of the inactivated X-chromosome was highly specific for female cases of type C, D and E tumors. Gene expression levels faithfully reflected the chromosomal copy number changes. Clinicopathological features significantly different between the 5 subtypes included metastatic disease and age at diagnosis and histology. Metastatic disease at diagnosis was significantly associated with subtypes C and D and most strongly with subtype E. Patients below 3 yrs of age had type B, D, or E tumors. Type B included most desmoplastic cases. We validated and confirmed the molecular subtypes and their associated clinicopathological features with expression data from a second independent series of 46 medulloblastomas.

Conclusions

The new medulloblastoma classification presented in this study will greatly enhance the understanding of this heterogeneous disease. It will enable a better selection and evaluation of patients in clinical trials, and it will support the development of new molecular targeted therapies. Ultimately, our results may lead to more individualized therapies with improved cure rates and a better quality of life.  相似文献

We report the development of simple fluorogenic probes that report on the activity of both bacterial and mammalian uracil–DNA glycosylase (UDG) enzymes. The probes are built from short, modified single-stranded oligonucleotides containing natural and unnatural bases. The combination of multiple fluorescent pyrene and/or quinacridone nucleobases yields fluorescence at 480 and 540 nm (excitation 340 nm), with large Stokes shifts of 140–200 nm, considerably greater than previous probes. They are strongly quenched by uracil bases incorporated into the sequence, and they yield light-up signals of up to 40-fold, or ratiometric signals with ratio changes of 82-fold, on enzymatic removal of these quenching uracils. We find that the probes are efficient reporters of bacterial UDG, human UNG2, and human SMUG1 enzymes in vitro, yielding complete signals in minutes. Further experiments establish that a probe can be used to image UDG activity by laser confocal microscopy in bacterial cells and in a human cell line, and that signals from a probe signalling UDG activity in human cells can be quantified by flow cytometry. Such probes may prove generally useful both in basic studies of these enzymes and in biomedical applications as well.  相似文献

The capacity for self-control is critical to adaptive functioning, yet our knowledge of the underlying processes and mechanisms is presently only inchoate. Theoretical work in economics has suggested a model of self-control centering on two key assumptions: (1) a division within the decision-maker between two ‘selves’ with differing preferences; (2) the idea that self-control is intrinsically costly. Neuroscience has recently generated findings supporting the ‘dual-self’ assumption. The idea of self-control costs, in contrast, has remained speculative. We report the first independent evidence for self-control costs. Through a neuroimaging meta-analysis, we establish an anatomical link between self-control and the registration of cognitive effort costs. This link predicts that individuals who strongly avoid cognitive demand should also display poor self-control. To test this, we conducted a behavioral experiment leveraging a measure of demand avoidance along with two measures of self-control. The results obtained provide clear support for the idea of self-control costs.  相似文献
