Amplitude characteristics and kinetics of laser-induced oxidation of high-potential cytochrome CH by a photosynthetic reaction center (RC) were investigated in Ectothiorhodospira shaposhnikovii chromatophore preparations of various humidity. It is shown that the diminuition of the amount of oxidized cytochrome and the decrease of the rate of the reaction on lowering the preparation humidity can be explained in terms of the concept of conformation-controlled electron transfer within the CH-RC complex. A model is suggested which predicts that the reversible transition of the complex from one conformational state which allows electron transfer ("contact" state) to the other in which the transfer is impossible ("non-contact" state) is the result of drying (or low temperature) induced changes in the electron tunnelling path in the region of "contact" of the cytochrome CH and RC protein globules.  相似文献

It is shown that the activity of enzymes participating in renin-angiotensin and brain kinin systems' metabolism depends on functional state of hypothalamo-pituitary-adrenocortical system. Under experimental hypocorticism the activity of angiotensin-converting enzyme and kininase I in the hypothalamus, hippocamp, corpus striatum and rat pituitary decreases; the renin-like enzyme activity decreases in the corpus striatum but increases in the hypothalamus and hippocamp. After hydrocortisone administration to adrenalectomized rats the angiotensin-converting enzyme activity of the hippocamp and pituitary is shown to be normalized as well as renin-like enzyme and kininase I of the hippocamp and corpus striatum. The activity of the studied enzymes in the hypothalamus decreases in this case.  相似文献

While studying the effect of peroral captopril injections on the activity of renin-angiotensin system (RAS), the activity of angiotensin-converting enzyme (ACE) and angiotensin II content from anterior and mediobasal hypothalamus, medulla and adenohypophysis of intact rats has been established to decrease. Captopril administration decreases ACE activity which increases after hydrocortisone injection in rat medulla and striatum. Captopril results in no potentiation of hormonal effect in hypothalamus and in adenohypophysis where ACE activity decreases following hydrocortisone injection. A decrease in the RAS activity of brain structures and adenohypophysis induced by captopril administration to rats is accompanied by the inhibition of the activity in the pituitary-adrenal system. A decrease in ACTH level and in 11-hydroxycorticosteroids of the rat blood plasma has been determined after single captopril injection in the dosage of 10 and 50 mg/kg of body weight. Duration of the effect depends on the captopril dosage.  相似文献

Parthenocarpy and fruit malformations are common among independent transgenic tomato lines, expressing genes encoding different pathogenesis-related (PR) protein and antimicrobal peptides. Abnormal phenotype developed independently of the expression and type of target genes, but distinctive features during flower and fruit development were detected in each transgenic line. We analyzed the morphology, anatomy, and cytoembryology of abnormal flowers and fruits from these transgenic tomato lines and compared them with flowers and fruits of wild tomatoes, line YaLF used for transformation, and transgenic plants with normal phenotype. We confirmed that the main cause of abnormal flower and fruit development was the alterations of determinate growth of generative meristem. These alterations triggered different types of anomalous growth, affecting the number of growing ectopic shoots and formation of new flowers. Investigation of the ovule ontogenesis did not show anomalies in embryo sac development, but fertilization did not occur and embryo sac degenerated. Nevertheless, the ovule continued to differentiate due to proliferation of endothelium cells. The latter substituted embryo sac and formed pseudoembryonic tissue. This process imitated embryogenesis and stimulated ovary growth, leading to the development of parthenocarpic fruit. We demonstrated that failed fertilization occurred due to defective male gametophyte formation, which was manifested in blocked division of the nucleus in the microspore and arrest of vegetative and generative cell formation. Maturing pollen grains were overgrown microspores, not competent for fertilization but capable to induce proliferation of endothelium and development of parthenocarpic ovary. Thus, our study provided new data on the structural transformations of reproductive organs during development of parthenocarpic fruits in transgenic tomato.  相似文献

We apply a theoretical approach developed earlier. The interaction ofions that permeate a channel with slowly relaxing charged channel-forminggroups (ion-conformational interaction – ICI) is addressed by thisapproach. One can describe the ion concentration influence (ion regulation)on channel functioning in this manner. A patch-clamp method in awhole-cell configuration is used to study the ICI. For this purpose theinfluence of an external concentration of potassium ions on thepotential-dependent potassium current (IA) in the externalmembrane of GH3 cells was studied. The increase of[K+ out] from 5 mM to 100 mM causes anon-monotonous shift of current-voltage dependencies. The dependence of bothan activation time constant tgrn and a steady-state activation(n) on [K+]out have a minimum andmaximum respectively. The analysis of the results suggests that the observedeffects are caused by ICI. A physical model is developed to describe thedependence of the potassium channel kinetics on the external concentrationof the ions and the membrane potential. The deformation of the closedstate of the gate and the corresponding energy shifts cause the observednon-monotonous dependencies due to ICI. Thus, the general theoreticalapproach has an experimental confirmation and is applied to concreteexamples. Formulas for concentrational dependencies of the channel kineticsare given for practical uses.  相似文献

A neural network that uses the basic Hebbian learning rule and the Bayesian combination function is defined. Analogously to Hopfield's neural network, the convergence for the Bayesian neural network that asynchronously updates its neurons' states is proved. The performance of the Bayesian neural network in four medical domains is compared with various classification methods. The Bayesian neural network uses more sophisticated combination function than Hopfield's neural network and uses more economically the available information. The naive Bayesian classifier typically outperforms the basic Bayesian neural network since iterations in network make too many mistakes. By restricting the number of iterations and increasing the number of fixed points the network performs better than the naive Bayesian classifier. The Bayesian neural network is designed to learn very quickly and incrementally.  相似文献

It has been established that hydrocortisone administration increased the amount of total, free, bound and synaptosomal GABA in the hypothalamus, glutamate decarboxylase activity in the homogenate and synaptosomes and time of the mediator turnover. ACTH administration increased the GABA content and glutamate decarboxylase activity in synaptosomes. The total amino acid content and time of its turnover got higher only with single hormone administration. In the hippocamp hydrocortisone administration increased the total and free GABA contents, its turnover time, glutamate decarboxylase activity in the homogenate and decreased GABA-aminotransferase activity in the homogenate and synaptosomes. The GABA level in synaptosomes grew only with multiple hormone administration. Single administration of ACTH decreased the total GABA content, glutamate decarboxylase activity in the homogenate, while its multiple administration increased the GABA level in synaptosomes followed by a decrease of GABA-aminotransferase activity in the homogenate and synaptosomes. The GABA turnover time fell with single hormone administration and grew with the multiple one. Adrenalectomy induced no changes in the GABA content and activity of its metabolism enzymes in the hypothalamus, however the bound GABA level decreased, while the turnover time increased. In the hippocamp adrenalectomy decreased total, free and synaptosomal GABA contents, glutamate decarboxylase activity in a homogenate and turnover time. Subsequent hydrocortisone administration only partly normalized the revealed changes of the GABA metabolism in the brain structures under adrenalectomy.  相似文献

Using voltage clamping, changes in inward calcium current arising after the occurrence of postsynaptic currents were investigated in isolated neurons ofHelix lucorum. Inward calcium current was reversibly reduced during the formation of inhibitory or excitatory postsynaptic current, produced either by spontaneous activation of an unidentified interneuron or by stimulating the anal nerve. Application of serotonin to the neuronal cell body also reversibly suppressed calcium current.
