The time course of production, specificity, and analytical potential of antizearalenone polyclonal rabbit antibody synthesized by formaldehyde condensation and conjugated to bovine serum albumin were investigated. The relative cross-reactivities with natural analogues were: zearalenone, 100%; -zearalenole, 0.15%; and -zearalenole, <0.02%. With synthetic analogues: zearalanone, 31.7% and -zearalanone, 0.12%. An enzyme immunoassay for zearalenone with a sensitivity of 0.1 ng/ml was developed on the basis of these antibodies and solid-phase conjugates homologous to the immunogen in the method of synthesis. 相似文献

Equilibrium maintenance during standing in humans was investigated with a 3-joint (ankle, knee and hip) sagittal model of
body movement. The experimental paradigm consisted of sudden perturbations of humans in quiet stance by backward displacements
of the support platform. Data analysis was performed using eigenvectors of motion equation. The results supported three conclusions.
First, independent feedback control of movements along eigenvectors (eigenmovements) can adequately describe human postural
responses to stance perturbations. This conclusion is consistent with previous observations (Alexandrov et al., 2001b) that
these same eigenmovements are also independently controlled in a feed-forward manner during voluntary upper-trunk bending.
Second, independent feedback control of each eigenmovement is sufficient to provide its stability. Third, the feedback loop

A study was made of the effect of adrenalectomy and substitution therapy in the absence of adrenals on the cAMP content in the hypothalamus, hippocamp and striate body of the rat brain. It was shown that in sham adrenalectomized animals, the concentration of cAMP in the brain structures under study did not differ from the characteristics recorded in the control group. Adrenalectomy in rats was followed by a significant reduction of the nucleotide in all the structures, namely in the hypothalamus by 89.5%, and in the hippocamp and striate body by 30.3 and 30.5%, respectively. Application of substitution therapy (5 mg hydrocortisone per 100 mg body mass) prevented the decreased content of cAMP seen in the adrenalectomized rats, with its magnitude being within the limits recorded in the intact animals. Peculiarities of aggregation in the samples of high density serum lipoproteins LHD2 and LHD3 obtained from healthy donors and patients with ishaemic heart disease were studied under isothermal exposition. It was found that lipoprotein samples from defective serum were characterized by chemical and structural inhomogeneity. It is reflected in the formation of spherolites of various types and of well-grown dendrite forms. Growth of stepped lens-shaped crystals of cholesterol takes place in the normal serum LHD3 preparations after long exposition. Crystal growth is thought to begin according to dislocational mechanism on linear defects existing in the liquid crystalline phase; then transformation to kinetically rough surface and to normal growth of the face occurs with the concentration increase. 相似文献

Formation kinetics, specificity, and analytical potential of polyclonal antibodies raised in rabbits against BSA conjugates of zearalanone carboxymethyloxime (CMO-ZAN) and zearalenone carboxymethyloxime (CMO-ZEN) have been studied. Preparation of the conjugates involved conversion of CMO-ZAN and CMO-ZEN into activated esters or carbodiimide condensation. Human artificial chromosome (HAC)-based vectors represent an alternative technology for gene delivery and expression with a potential to overcome the problems caused by virus-based vectors. The recently developed alphoidtetO-HAC has an advantage over other HAC vectors because it can be easily eliminated from cells by inactivation of the HAC kinetochore via binding of chromatin modifiers, tTA or tTS, to its centromeric tetO sequences. This provides a unique control for phenotypes induced by genes loaded into the HAC. The alphoidtetO-HAC elimination is highly efficient when a high level of chromatin modifiers as tetR fusion proteins is achieved following transfection of cells by a retrovirus vector. However, such vectors are potentially mutagenic and might want to be avoided under some circumstances. Here, we describe a novel system that allows verification of phenotypic changes attributed to expression of genes from the HAC without a transfection step. We demonstrated that a single copy of tTAVP64 carrying four tandem repeats of the VP16 domain constitutively expressed from the HAC is capable to generate chromatin changes in the HAC kinetochore that are not compatible with its function. To adopt the alphoidtetO-HAC for routine gene function studies, we constructed a new TAR-BRV- tTAVP64 cloning vector that allows a selective isolation of a gene of interest from genomic DNA in yeast followed by its direct transfer to bacterial cells and subsequent loading into the loxP site of the alphoidtetO-HAC in hamster CHO cells from where the HAC may be MMCT-transferred to the recipient human cells. 相似文献

Effect of theophylline on electrical activity of bursting type in an identified Helix pomatia neuron

The effect of theophylline, an inhibitor of cyclic nucleotide phosphodiesterase, on electrical activity of bursting neuron RPa1 ofHelix pomatia was investigated. In a concentration of 1 mM theophylline, when added to the external solution, increases the frequency and number of action potentials in the burst and also the duration of the inter-burst interval and the amplitude of membrane potential waves. In concentrations of 2.5 and 5.0 mM theophylline leads to reversible inhibition of bursting activity. During rinsing this activity rises to a higher level and then returns to the original value. The action of theophylline develops and disappears (as a result of rinsing) in the course of 1–5 min, depending on concentration of the inhibitor. It is suggested that electrical activity of the molluscan bursting neuron is controlled through the cyclic nucleotide system.A. A.
