Genotypic composition of the erythrocyte population of peripheral blood in 16 aggregation chimeras: BALB/c (H-2dd)----C57BL/10(H-2bb) and BALB/c(H-2dd)----B10.D2(H-2dd) was studied during 10 months. The proportion of cells of parental components was defined visually in the coat and by electrophoresis of allozyme variants at the Gpi-1 locus in blood. The similar increase of blood cells percentage was observed in blood of both types of chimeras with age. In chimeras the skin grafts of both parental types survive. Chimeric drift is not caused by H-2 haplotypes differences between cells of two strains or disturbance of immunological tolerance in the chimeric mice. We propose that chimeric drift results from interaction of hemopoietic cells of different strains in early stages of hemopoiesis.  相似文献

An analysis of aphakia (ak) gene expression in 16 day ak/ak C/C----+/+ c/c chimaeric embryos has shown, that ak gene, acting in developing lens, blocks lens cell differentiation and disturbs the formation by these cells of the extracellular matrix composing the lens capsule material. The dependence of capsule structure in chimaeras on the genotype of underlying cells indicates that lens cells are responsible for the formation of lens capsule.  相似文献

The eye development has been studied in the 12-day-old, 14-day-old embryos and in neonates of Cm/+ ak/ak genotype. The gene coloboma (Cm) in heterozygous state causes a typical coloboma of the iris and the gene aphakia (ak) blocks the lens development in the homozygotes. It has been shown that in Cm/+ ak/ak mice the eyes go through mainly the same abnormal development as that in +/+ ak/ak animals. In mice of both genotypes the lens morphogenesis blocking at the vesicle stage and the retinal fold in the dorsal half of the eye develops. However, the ventral retinal fold which is characteristic for the +/+ ak/ak mice does not form in the Cm/+ ak/ak animals that is the result of the interaction of Cm and ak genes in the eye morphogenesis. The Cm gene suppressing the growth of the retina ventral half inhibits the formation of its fold in Cm/+ ak/ak embryos. As a result of the gene interaction a certain normalization of the eye development compared to the +/+ ak/ak mice is observed in the Cm/+ ak/ak animals. The obtained data show that the Cm gene expresses in the cell clones of the retina ventral half.  相似文献

The exposure of F. tularensis vaccine strain 15/10 in the form of aerosol for more than 10 minutes results in the decrease of its virulence, immunogenicity and the content of species-specific antigen and in the increase of the dissociation level. Proceeding from this fact, during aerosol immunization with this vaccine strain exposure must not exceed 5 minutes for unstabilized aerosol and 10 minutes for aerosol stabilized with 5% glycerin. Under these conditions the properties of F. tularensis strain 15/10 are retained practically on the initial level.  相似文献

An electron microscope study of lenses in 11--15 and 17 days old embryos of mice homozygous by dominant cataract-Fr (CatFr) gene has shown that ultrastructural changes in the nuclear envelope are the earliest expression of the mutant gene. The primary lens fibers of 12 days old embryos CatFr/CatFr, unlike those of the normal ones, are characterized by the decrease in the number of nuclear pores, evagination of the outer nuclear membrane, marked and unequal extension of perinuclear space which connects itself with the endoplasmic reticulum cisternae. In 14 days old embryos breaks in the outer nuclear membrane and evaginations in the inner one, fusion of nuclear membranes and breaks of the nuclear envelope are observed and resulted in the release of the nuclear contents with the nucleolus in the cytoplasm. Similar ultrastructural changes are characteristic also of the nuclei of secondary lens fibers at a comparable stage of differentiation. The destructive changes of nuclei are accompanied by the degeneration and autophagocytosis of cellular organelles and matrix.  相似文献
