The presence of Marek's disease tumor-associated surface antigen (MATSA) was demonstrated by the direct and indirect membrane immunofluorescent tests, in chicks inoculated 7-10 days earlier with herpesvirus of turkeys (HVT), O1 strain. In in vitro cultures of spleen lymphocytes and ovaries obtained from these chicks, MATSA-positive cells were also detected after 1-7 days cultivation. A possible mechanism of protection by HVT vaccine against Marek's disease is proposed.  相似文献

Protein phosphorylation by activation of protein kinase C was examined using quiescent cultures of the mouse epidermal keratinocyte line BALB/MK-2. Treatment with phorbol ester caused rapid phosphorylation of five proteins with molecular weights of 80,000, 70,000, 40,000, 34,000, 28,000. Of these proteins, the 70,000 molecular weight one (p70) was studied further. Its position on two-dimensional gel suggested that p70 is nuclear envelope lamin B. This possibility was confirmed by the co-migration of p70 with the lamin fraction of mouse liver and its immunoprecipitation with antinuclear lamina antibody. The lamin B fraction consists of lamin B1 and lamin B2. Evidence that p70 is lamin B2 was obtained by peptide mapping and amino acid sequencing. Lamin B2 is the only lamin that shows a substantial increase in phosphorylation on treatment of BALB/MK-2 cells with phorbol ester.  相似文献

The entire and partial gag regions of human immunodeficiency virus type 1 (HIV-1) were overproduced in Escherichia coli and used for epitope mapping of antibodies against p17. We found that a mouse monoclonal antibody to p17, V17 recognizes the mature p17 but not the unprocessed Gag proteins containing the entire p17 moiety. Further analysis revealed that V17 recognizes the C-terminal 12-amino-acid region of p17 having free C-terminus. This monoclonal antibody may be useful for monitoring the maturation of virus particles.  相似文献

Rubella hemagglutination inhibition (HI) antibodies in 266 children with rubella syndrome born in 1965 in the Ryukyu Islands and their mothers were followed for seven years. Titers of rubella HI antibody in the mothers declined slowly, while those in the children declined rapidly up to 40 months of age. Thereafter decline of titers became extremely slow and only seven cases (three per cent) became seronegative for rubella HI antibody. Rubella HI antibody titers seemed to have no particular correlation to the severity of clinical manifestations.  相似文献

A concise method was designed for preparation of C-reactive protein (CRP) from pleural effusion. By addition of L-alpha-lecithin to the pleural effusion in the presence of calcium ions, a flocculence of the CRP-lecithin complex formed. Subsequent treatment of the CRP-lecithin complex with chloroform and sodium citrate buffer enabled extraction of the CRP in the buffer layer. This extracted CRP was further purified by sequential treatment on column chromatography of DEAE cellulose (DE52) and gel filtration using Sephacryl S-300. The isolated protein was proved to be native CRP with a high degree of purity, as determined by electrophoretical and immunological analysis. The yield was 41.8% recovery from the starting material. E1%(280) of the CRP preparation was estimated to be 18.75.  相似文献

Reactive oxygen species (ROS) are known participants in several cellular processes. Superoxide anion radical, one example of ROS, forms as a result of normal cellular respiration and is usually cleared successfully by superoxide dismutase (SOD) and other radical scavengers. However, when superoxide exceeds the clearance capacity of SOD and other ROS scavengers, superoxide initiates a number of pathologic processes. This review examines pathologies involving superoxide, including: cancer, neurodegenerative diseases, ischemia/reperfusion injury, and inflammation. We will also explore the basic science principles of superoxide and SOD, including: SOD evolution, SOD mutations, biochemistry, physiology, and pathophysiology. In reviewing the basic science, clinical pathology, and therapeutic research, we hope to clearly demonstrate plausible pharmacologic targets of action. We have revised data about basic science, clinical pathology and therapeutic research in an effort to propose plausible pharmacological targets of action. The understanding of these aspects is critical in the accomplishment of a successful clinical intervention.  相似文献
