A unique lineage of bacteria belonging to the order Bacteroidales was identified as an intracellular endosymbiont of the protist Pseudotrichonympha grassii (Parabasalia, Hypermastigea) in the gut of the termite Coptotermes formosanus. We identified the 16S rRNA, gyrB, elongation factor Tu, and groEL gene sequences in the endosymbiont and detected a very low level of sequence divergence (<0.9% of the nucleotides) in the endosymbiont population within and among protist cells. The Bacteroidales endosymbiont sequence was affiliated with a cluster comprising only sequences from termite gut bacteria and was not closely related to sequences identified for members of the Bacteroidales attached to the cell surfaces of other gut protists. Transmission electron microscopy showed that there were numerous rod-shaped bacteria in the cytoplasm of the host protist, and we detected the endosymbiont by fluorescence in situ hybridization (FISH) with an oligonucleotide probe specific for the 16S rRNA gene identified. Quantification of the abundance of the Bacteroidales endosymbiont by sequence-specific cleavage of rRNA with RNase H and FISH cell counting revealed, surprisingly, that the endosymbiont accounted for 82% of the total bacterial rRNA and 71% of the total bacterial cells in the gut community. The genetically nearly homogeneous endosymbionts of Pseudotrichonympha were very abundant in the gut symbiotic community of the termite.  相似文献

The ratio of oleic acid in the plasma FFA fraction was higher in the Japanese than in the Ainu. When glucose was administered, the proportion of oleic acid was reduced, while that of stearic acid increased in both groups. In the Japanese, moreover, a decrease in linoleic acid and increases in palmitic and myristic acids were also observed. As the plasma FFA level rose after injection of norepinephrine, the proportion of oleic acid increased and that of palmitic, stearic, myristic and lauric acids decreased, and the composition approached the pattern of depot fat. It was inferred that the difference observed in the plasma FFA composition between the Ainu and Japanese was attributable to the difference in the plasma levels of FFA which was significantly lower in the Ainu.
Zusammenfassung Das Verhältnis von Oelsäure in der freien Fettsäurenfraktion (FFA) im Plasma war bei Japanern (n=9) höher als bei Ainus (n=5). Nach Glukose war das Oelsäure-Verhältnis vermindert, während in beiden Gruppen Stearinsäure anstieg. Bei den Japanern ergaben sich ausserdem ein Abfall der Linolensäure und Austiege der Palmitin- und Myristinsäure. Wenn der Plasma FFA-Spiegel nach Noradrenalin-Injektion anstieg, stieg der Oelsäureanteil und fielen die Palmitin-, Myristin- und Laurinsäureanteile und die Zusammensetzung war wie die von Depotfett. Es wird angenommen, dass die Unterschiede in der Zusammensetzung der Plasma FFA bei Japanern und Ainus Folge der unterschiedlichen FFA-Spiegel waren, die bei den Ainu signifikant tiefer lagen.

Resume La proportion d'acide oleique dans l'ensemble des acides gras libres (FFA) du plasma sanguin est plus marquée chez les Japonais (n=9) que chez les Aïnous (n=5). La proportion d'acide oleique a diminuê après l'administration de glucose, car, dans les deux groupes, la teneur en acide stéarique s'est renforcée. On constante en outre une diminution de l'acide linoleique et une augmentation des acides palmitique et myristique. Lorsque, après injection de noradrénaline, le taux de FFA du plasma se relève, on constate une augmentation des quantités relatives de l'acide oleique au détriment de la part prise par les acides palmitique, myristique et laurique et leur composition est semblable aux dépots adipeux. On peut ainsi admettre que les différences de composition du FFA plasmatique découlent du taux dissemblable du FFA entre Aïnous et Japonais. Ce taux est significativement inférieur chez les premiers.

Notch1 mutations are found in more than 50% of human T cell acute lymphoblastic leukemia (T-ALL) cells. However, the functions of Notch1 for human T cell development and leukemogenesis are not well understood. To examine the role of Notch1, human hematopoietic stem cells (HSCs), which had been transduced with a constitutively active form of Notch1 (ICN1), were transplanted into severely immunodeficient NOD/Shi-scid-IL2rγnull (NOG) mice. We found that the great majority of the ICN1-expressing hematopoietic cells in the bone marrow expressed surface markers for T cells, such as CD3, CD4, and CD8, and that this T cell development was independent of the thymus. Accordingly, phenotypically mature CD8+ single positive (SP) T cells were observed in the spleen. Furthermore, T-ALL developed in one NOG recipient mouse out of 26 that had been secondary transferred with the T cells developed in the first NOG mice. These results indicate that Notch1 signaling in HSCs promotes CD8+ SP T cell development, and that T cell leukemogenesis may require additional oncogenic factors other than Notch1 activation.  相似文献

Recently, the conversion of plantations into natural forests has attracted increasing attention, but little is known about how to convert a windthrown plantation into a natural forest. We examined the initial 3-year effects of salvage logging, site preparation, planting, and weeding in comparison with the effects of legacy retention after a typhoon blowdown on the changes in terrestrial environments and plant compositions and the similarity of the species composition to that of the intact natural forest. Our major findings are as follows: (1) A homogeneous, sunny, and flat environment continued for 3 years in the planted and weeded plot, whereas a heterogeneous environment was observed in the legacy retention plot; however, the light environment changed slightly in the planted plot. (2) The species composition in the planted plots changed greatly due to invasion by annual, biennial, and perennial herbs and early successional trees. Alien species also invaded the planted row. Conversely, the species composition changed little in the legacy retention, which was likely due to the presence of residual plants that prevented shade-intolerant species from being established. (3) The legacy retention, followed by the residual row, was the closest to the natural forest with respect to species composition, whereas the planted row was the most dissimilar from the natural forest. Legacy retention appears to be the most appropriate way to restore a natural forest immediately after a windthrow, yet supplemental planting may be necessary to restore the desired broadleaved species.  相似文献

Eleven strains of acetic acid bacteria were isolated from stems of sugarcane, fruits, and a flower in Japan. The isolates were separated into three groups, Groups I, II, and III, in the genus Acetobacter according to phylogenetic analysis based on 16S rRNA sequences. The isolates had sequence similarities of 99.8-100% within the Group, 99.3-99.6% to those of the type strains of each related Acetobacter species, and less than 98.4% to those of the type strains of other Acetobacter species. Genomic DNA G+C contents of Groups I, II, and III were 59.2-59.4, 60.5-60.7, and 58.7-58.9 mol%, respectively. The isolates in the Group showed high values of DNA-DNA relatedness to each other, but low values less than 46% to the type strains of related Acetobacter species. A good correlation was found between the three Groups and groups based on DNA G+C contents and DNA-DNA relatedness. All the strains had Q-9 as the main component, and Q-8 and Q-10 as minor components. The isolates in the three Groups did not completely match with any Acetobacter species on catalase reaction, the production of ketogluconic acids from D-glucose, growth on ammoniac nitrogen with ethanol (Hoyer-Frateur medium and Frateur modified Hoyer medium), growth on 30% (w/v) D-glucose, growth in 10% (v/v) ethanol, or DNA G+C contents. On the basis of phylogenetic relationships in the genus Acetobacter and chemosystematic and phenotypic characteristics, the three Groups were regarded as novel species in the genus Acetobacter. Acetobacter okinawensis sp. nov. is proposed for Group I, Acetobacter papayae sp. nov. for Group II, and Acetobacter persicus sp. nov. for Group III.  相似文献

A new species of benthic marine dinoflagellate, Pyramidodinium spinulosum Horiguchi, Moriya, Pinto & Terada is described from the deep (36 m) seafloor off Mageshima Island, Kagoshima Prefecture, Japan in the subtropical region of the northwest Pacific. The life cycle of the dinoflagellate consists of a dominant, attached, dome‐shaped, vegetative form and short‐lasting, motile cell. Asexual reproduction takes place by the formation of two motile cells within each non‐motile cell. The released motile cells swim only for a short period and transform directly into the dome‐shaped vegetative form. The duration of the cell cycle varies and can be extremely long, ranging 5–38 days under culture conditions. The non‐motile cell is enclosed by a cell wall and its surface is covered with many (80 – 130) spines of various length. The dinoflagellate is photosynthetic and contains many (more than 50) discoidal chloroplasts. Phylogenetic analysis reveals that the dinoflagellate is closely related to the type species of the genus Pyramidodinium, P. atrofuscum which also possesses a dominant, attached, non‐motile form. However, P. spinulosum can be clearly distinguished from P. atrofuscum by the cell shape (dome‐shaped vs. pyramid‐shaped) and surface ornamentation (spines vs. wart‐like processes) of the non‐motile form. Based on these morphological differences together with molecular evidence, it was concluded that this organism from a deep water sand sample should be described as a second species of the genus Pyramidodinium, P. spinulosum.  相似文献

Introduction

Recent advances in microbiome research have revealed the diverse participation of gut microbiota in a number of diseases. Bacteria-specific endogenous small molecules are produced in the gut, are transported throughout the whole body by circulation, and play key roles in disease establishment. However, the factors and mechanisms underlying these microbial influences largely remain unknown.

Objectives

The purpose of this study was to use metabolomics to better understand the influence of microbiota on host physiology.

Methods

Germ-free mice (GF) were orally administered with the feces of specific pathogen-free (SPF) mice and were maintained in a vinyl isolator for 4 weeks for establishing the so-called ExGF mice. Comparative metabolomics was performed on luminal contents, feces, urine, plasma, and tissues of GF and ExGF mice.

Results

The metabolomics profile of 1716 compounds showed marked difference between GF and ExGF for each matrix. Intestinal differences clearly showed the contribution of microbiota to host digestive activities. In addition, colonic metabolomics revealed the efficient conversion of primary to secondary metabolites by microbiota. Furthermore, metabolomics of tissues and excrements demonstrated the effect of microbiota on the accumulation of metabolites in tissues and during excretion. These effects included known bacterial effects (such as bile acids and amino acids) as well as novel ones, including a drastic decrease of sphingolipids in the host.

Conclusion

The diverse effects of microbiota on different sites of the host metabolome were revealed and novel influences on host physiology were demonstrated. These findings should contribute to a deeper understanding of the influence of gut microbiota on disease states and aid in the development of effective intervention strategies.

We present 3 adult cases of visceral toxocariasis from the same family, who each consumed thin slices of raw bovine liver weekly, and developed eosinophilia and multiple small lesions in their livers and lungs. Serological examinations using the larval excretory–secretory product of Toxocara canis strongly indicated infection with Toxocara species larvae. The patients responded well to treatment with albendazole. Ingestion of raw liver from paratenic animals is considered to be a common transmission route of human toxocariasis, especially in adults.  相似文献

Design, syntheses, and structure-activity relationships of a novel class of 2-{3-oxospiro[isobenzofuran-1(3H),4'-piperidin]-1'-yl}benzimidazole NPY Y5 receptor antagonists are described. The benzimidazole structures were newly designed based on the urea linkage of our prototype Y5 receptor antagonists (2 and 3). By optimizing substituents on the benzimidazole core part of the lead compound 5a, we were able to develop a potent, orally available, and brain-penetrable Y5 selective antagonist (5k).  相似文献
