Abstract: Among various tissues of mouse, β1,4- N -acetylgalactosaminyltransferase (GM2/GD2 synthase) gene is expressed predominantly in the brain. Further analysis of the gene expression in the mouse CNS was performed by northern blotting and by enzyme assays using extracts from various parts of the CNS. In situ hybridization was also done to investigate the distribution of cells generating GM2/GD2 synthase. In northern blots, diverse levels of the gene expression were observed, depending on the regions examined. By in situ hybridization, pyramidal cells in the hippocampus, granular cells in dentate gyrus and cerebral cortex, Purkinje cells in cerebellum, and mitral cells in the olfactory bulb expressed high levels of the mRNA; these results corresponded to the results obtained by northern blot. Enzyme levels in these sites were accordingly high. However, enzyme levels in certain areas with low mRNA intensities, such as thalamus and pons medulla, were higher than expected from the results of northern blotting. The significance of the high gene expression in certain areas for brain function and the reason for the discrepancy between mRNA level and enzyme activity in some regions are discussed. 相似文献

Oxidative stress mediated clustering of membrane protein band 3 plays an essential role in the clearance of damaged and aged red blood cells (RBCs) from the circulation. While a number of previous experimental studies have observed changes in band 3 distribution after oxidative treatment, the details of how these clusters are formed and how their properties change under different conditions have remained poorly understood. To address these issues, a framework that enables the simultaneous monitoring of the temporal and spatial changes following oxidation is needed. In this study, we established a novel simulation strategy that incorporates deterministic and stochastic reactions with particle reaction-diffusion processes, to model band 3 cluster formation at single molecule resolution. By integrating a kinetic model of RBC antioxidant metabolism with a model of band 3 diffusion, we developed a model that reproduces the time-dependent changes of glutathione and clustered band 3 levels, as well as band 3 distribution during oxidative treatment, observed in prior studies. We predicted that cluster formation is largely dependent on fast reverse reaction rates, strong affinity between clustering molecules, and irreversible hemichrome binding. We further predicted that under repeated oxidative perturbations, clusters tended to progressively grow and shift towards an irreversible state. Application of our model to simulate oxidation in RBCs with cytoskeletal deficiency also suggested that oxidation leads to more enhanced clustering compared to healthy RBCs. Taken together, our model enables the prediction of band 3 spatio-temporal profiles under various situations, thus providing valuable insights to potentially aid understanding mechanisms for removing senescent and premature RBCs. 相似文献

Stand dynamics was studied over 13 years in a cool-temperate conifer-hardwood forest, northern Japan. A total 30 hardwood
species and one conifer, Abies sachalinensis, larger than 1.5 cm DBH were recorded. The total stand density was 1677 trees ha−1 at the beginning, decreasing to 1184 trees ha−1 (30% reduction) over the study period, but the total stand basal area was almost unchanged (about 49 m2 ha−1). This large reduction in total density was mainly due to the death of saplings and infrequent recruitment. Number of recruits
gradually decreased with time, while that of dead trees was constant. Cause of death of small trees was mainly due to suppression
by tall trees. Skewness of the DBH frequency distribution varied among the species. A less skewed frequency distribution (i.e.,
few number of saplings) was shown by shade-intolerant species such as Populus maximowiczii and Betula maximowicziana, and a more skewed frequency distribution (i.e., large number of saplings) by shade-tolerant species such as Acer mono and Tilia japonica. DBH frequency distribution changed to less skewed patterns with reduction of density in most species during the census period.
Rank of shade tolerance positively correlated with tree density and skewness, and negatively correlated with mean DBH. Skewness
also positively correlated with recruitment rates. Furthermore, rank of shade tolerance positively correlated with seed size.
These results suggest that shade-intolerant species regenerated immediately after disturbances by wide dispersal of small
seeds, but their recruitment was interrupted after that. By contrast, shade-tolerant species were able to recruit even after
the ceasation of recruitment of shade-intolerant species, but suffered severe mortality due to the increasing shading with
the progress of stand development. This study suggests that the stand is still developing, with changes in species composition
and size structure, and that species differences in shade tolerance and seed size are important for the stand structural changes. 相似文献

Background
Horse fibrinogen has been identified as a plasma specific ferritin-binding protein. There are two ways in the binding of ferritin-binding protein with ferritin: one is direct binding and the other is indirect binding which is heme-mediated. The aim of this study was to analyze the binding between horse fibrinogen and ferritin.Findings
Although fibrinogen in horse plasma did not show the binding to ferritin coated on the plate wells, after following heat-treatment (60°C, 30 min) of horse plasma, plasma fibrinogen as well as purified horse fibrinogen bound to plates coated with horse spleen ferritin, but not with its apoferritin which lost heme as well as iron after the treatment of reducing reagent. Binding of purified or plasma fibrinogen to ferritin was inhibited by hemin and Sn-protoporphyrin IX (Sn-PPIX), but not by PPIX or Zn-PPIX.Conclusions
Heat-treatment of horse plasma enabled plasma fibrinogen to bind to plate well coated with holo-ferritin. From the binding analysis of fibrinogen and ferritin, it is suggested that horse fibrinogen recognized iron or tin in complexed with the heme- or the hemin-ring, and also suggest that some fibrinogens circulate in the form of a complex with ferritin and/or heat-labile factors which inhibit the binding of fibrinogen with ferritin.287.
