A large portion of the usual eukaryotic genome is comprised of repetitive sequences. A common situation, when several related but different repeat families share the same conserved motif, complicates repeat classification and repeat boundary definition. If the repeats are aligned by the motif position, then the sequence profile (pattern) resulting from the alignment will represent overlapping of the profiles (patterns) corresponding to the individual families. A novel algorithm for the decomposition of overlapping patterns is proposed. It can be used with both continuous and gapped patterns. The technique is based on accumulation of simultaneously occurring pattern features found by cross-correlation procedure with limited lag length; thus, the name is Cumulative Local Cross-Correlation (referred further as CLCC). Its sensitivity is tested on human genomic sequences. Software implementation of the algorithm is available on request from the author. 相似文献

Positional distributions of various dinucleotides in experimentally derived human nucleosome DNA sequences are analyzed. Nucleosome positioning in this species is found to depend largely on GG and CC dinucleotides periodically distributed along the nucleosome DNA sequence, with the period of 10.4 bases. The GG and CC dinucleotides oscillate counterphase, i.e., their respective preferred positions are shifted about a half-period from one another, as it was observed earlier for AA and TT dinucleotides. Other purine-purine and pyrimidine-pyrimidine dinucleotides (RR and YY) display the same periodical and counterphase pattern. The dominance of oscillating GG and CC dinucleotides in human nucleosomes and the contribution of AG(CT), GA(TC), and AA(TT) suggest a general nucleosome DNA sequence pattern - counterphase oscillation of RR and YY dinucleotides. AA and TT dinucleotides, commonly accepted as major players, are only weak contributors in the case of human nucleosomes. 相似文献

We studied 42 species of saprophagous, Neotropical Copestylum (Diptera, Syrphidae) reared from decaying Cactaceae and Agavaceae. Thirty‐three species were reared during fieldwork in Bolivia, Costa Rica, Ecuador, Mexico, Peru, and Trinidad from 1998–2007. Nine species came from museum and private collections. Seven were new species. We describe these new species and the third stage larva and/or puparium and breeding sites of 40 species. Not described are two apparent species related to Copestylum apicale (Loew, 1866) reared from Cactaceae. Resolution of their status was beyond the scope of this paper but reference is made to their distinctive larval morphology. Based on early stage characters all reared species can be placed in ten species groups, all but three of which have been recognized previously on the basis of adult characters. A high level of congruence was found between adult and larval characters in terms of these species groups. Eight of the groups appear to be related closely and may represent a monophyletic lineage within Copestylum that has diversified in xeric habitats. Early stage morphology varied within and amongst groups but two trends in functional morphology are recognizable. One trend is towards feeding in watery decay and the other towards feeding in firmer decay. The latter trend is characterized by species that scoop food and use grinding mills in their head skeletons to break it up. They also have armoured thoraces with varying arrangements of sclerotized spicules or stiffened setae for gripping and protection during tunnelling, a short anal segment, and a short posterior breathing tube for protecting the openings. The former trend is characterized by species with opposite and contrasting features. They filter food and have well‐developed pre‐oral setal filters but they lack grinding mills or only have poorly developed grinding mills. They have reduced thoracic armature, elongate anal segments, and posterior breathing tubes which facilitates simultaneous feeding and respiration. Comparison with 23 Copestylum species reared from bromeliads (Bromeliaceae) suggests a common pattern of diversification in that species groups with the largest body sizes are more specialized. 相似文献

The novel Cu(II), Ni(II), Zn(II), Co(II) coordination compounds with Schiff base ligand - N,N′-bis(2-tosylaminobenzylidene)-1,3-diaminopropanol have been synthesized and studied. The structures of bis-azomethine as well as Co(II) and Zn(II) mononuclear metallochelates have been determined by X-ray analysis. The magnetic properties of all complexes were studied and interpreted in terms of HDVV theory. Kinetics, biodistribution, and histological studies were performed to evaluate the particle‐size effects on the distribution of 15 nm and 50 nm PEG‐coated colloidal gold (CG) particles and 160 nm silica/gold nanoshells (NSs) in rats and rabbits. The above nanoparticles (NPs) were used as a model because of their importance for current biomedical applications such as photothermal therapy, optical coherence tomography, and resonance‐scattering imaging. The dynamics of NPs circulation in vivo was evaluated after intravenous administration of 15 nm CG NPs to rabbit, and the maximal concentrations of gold were observed 15–30 min after injection. Rats were injected in the tail vein with PEG‐coated NPs (about 0.3 mg Au/kg rats). 24 h after injection, the accumulation of gold in different organs and blood was determined by atomic absorption spectroscopy. In accordance with the published reports, we observed 15 nm particles in all organs with rather smooth distribution over liver, spleen and blood. By contrast, the larger NSs were accumulated mainly in the liver and spleen. For rabbits, the biodistribution was similar (72 h after intravenous injection). We report also preliminary data on the light microscopy and TEM histological examination that allows evaluation of the changes in biotissues after gold NPs treatment. 目的探讨应用BR-TRG-I型体腔热灌注治疗系统进行持续循环腹腔热灌注治疗(continuous circulatory hyperthermic intraperitoneal perfusion treatment,CCHIP)对实验动物的生命体征、肝肾功能及内脏器官病理形态的影响。方法选用家猪作为实验动物,应用BR-TRG-I型体腔热灌注治疗系统建立CCHIP动物模型,分别用44℃、45℃的设定温度进行CCHIP。在CCHIP期间记录实验动物的生命体征变化;CCHIP前及治疗后1d、3d、7d和14d抽取外周血液保存备检;CCHIP结束后即刻及2周后处死动物,观察内脏器官的大体形态学改变,切取肝、肾、小肠等器官进行病理检查。结果44℃CCHIP持续1.5h对猪的生命体征、肝肾功能无明显影响,肝、肾、小肠等脏器损伤较轻,2周后恢复正常;45℃CCHIP持续1.5h对猪的生命体征影响严重,肝肾功能损害持续2周尚不能恢复正常,肝、肾、小肠等脏器病理损伤明显。结论44℃温度CCHIP1.5h安全可行,可以作为CCHIP的安全温度;45℃温度CCHIP1.5h可对实验动物肝肾功能造成严重损害,不适合作为CCHIP的治疗温度。 相似文献

Structural Conservation of Components in the Amino Acid Sensing Branch of the TOR Pathway in Yeast and Mammals 总被引:1,自引:0,他引:1
The highly conserved Rag family GTPases have a role in reporting amino acid availability to the TOR (target of rapamycin) signaling complex, which regulates cell growth and metabolism in response to environmental cues. The yeast Rag proteins Gtr1p and Gtr2p were shown in multiple independent studies to interact with the membrane-associated proteins Gse1p (Ego3p) and Gse2p (Ego1p). However, mammalian orthologs of Gse1p and Gse2p could not be identified. We determined the crystal structure of Gse1p and found it to match the fold of two mammalian proteins, MP1 (mitogen-activated protein kinase scaffold protein 1) and p14, which form a heterodimeric complex that had been assigned a scaffolding function in mitogen-activated protein kinase pathways. The significance of this structural similarity is validated by the recent identification of a physical and functional association between mammalian Rag proteins and MP1/p14. Together, these findings reveal that key components of the TOR signaling pathway are structurally conserved between yeast and mammals, despite divergence of sequence to a degree that thwarts detection through simple homology searches. 相似文献
