The great challenges for researchers working in the field of vaccinology are optimizing DNA vaccines for use in humans or large animals and creating effective single-dose vaccines using appropriated controlled delivery systems. Plasmid DNA encoding the heat-shock protein 65 (hsp65) (DNAhsp65) has been shown to induce protective and therapeutic immune responses in a murine model of tuberculosis (TB). Despite the success of naked DNAhsp65-based vaccine to protect mice against TB, it requires multiple doses of high amounts of DNA for effective immunization. In order to optimize this DNA vaccine and simplify the vaccination schedule, we coencapsulated DNAhsp65 and the adjuvant trehalose dimycolate (TDM) into biodegradable poly (DL-lactide-co-glycolide) (PLGA) microspheres for a single dose administration. Moreover, a single-shot prime-boost vaccine formulation based on a mixture of two different PLGA microspheres, presenting faster and slower release of, respectively, DNAhsp65 and the recombinant hsp65 protein was also developed. These formulations were tested in mice as well as in guinea pigs by comparison with the efficacy and toxicity induced by the naked DNA preparation or BCG. The single-shot prime-boost formulation clearly presented good efficacy and diminished lung pathology in both mice and guinea pigs.  相似文献

Purpose

Built in 1941, the Progreso Pier was the first concrete structure in the world built with nickel-containing stainless steel reinforcement. The Pier has been in service for over 70 years without any significant repair or maintenance activities. The aim of this study was to understand the environmental and economic implications of selecting nickel-containing stainless steel reinforcement using the Progreso Pier as the case study.

Methods

A combined environmental life cycle assessment (LCA) and life cycle costing (LCC) study was conducted. The analysis considered the potential environmental impacts and the net present cost of the stainless steel reinforced structure from cradle to grave and compared it to the same structure using conventional carbon steel.

Results and discussion

The results indicated that while using stainless steel reinforcement resulted in a marginally higher environmental impact after initial construction, this is offset by the increased service life and, hence, less frequent maintenance and reconstruction activities. Relative to the as-built stainless steel reinforcement design, the environmental impacts of the carbon steel reinforced design are between 69 and 79 % higher over the analysis period. Similar observations were made for the other investigated impact categories. The cost implications of using stainless steel reinforcement show economic benefits that are complementary to the environmental benefits. Similar to the LCA, the service life benefits outweigh the higher unit costs for stainless steel, assuming a discount rate of 0.01 % as the baseline scenario. The carbon steel reinforced design has a net present cost that is 44 % higher than the as-built stainless steel reinforcement design. The crossover point for the two designs occurs at year 50, which corresponds to the reconstruction activity. A sensitivity analysis shows that the results and conclusions are sensitive to the choice in discount rate: Rates 3 % and lower produce net present costs that are lower for the as-built design; rates 4 % and higher produce net present costs that are lower for the alternative design.

Conclusions

The study demonstrates how LCA and LCC are complementary tools that can be used in decision-making for sustainable construction. The Progreso Pier exemplifies the importance of considering the entire life cycle with service life and recycling as well as long-term life cycle impacts of infrastructure projects from an environmental and economic perspective.

The Lower Cambrian Chengjiang fauna is reviewed and shown to be closely comparable with the younger Burgess Shale fauna. but with various differences in detail. A diverse group of more or less annulated lobopod animals including 'armoured lobopods' are regarded as representatives of the phylum Onychophora. 'Trilobitomorphs' include several new types. Probable protaspides of the trilobitomorph Naraoia are described. No molluses or deuterostomes have been identified. The preservational orientations of the various taxa are reviewed and compared with orientations of the Burgess Shale taxa. Orientation in the sediment is found to be closely correlated to the original shape of individuals. Several new genera and species are described: the segmented. worm-shaped Yunnanozoon lividum gen. et sp.n., the 'armoured lobopods' Onychodictyon ferox gen. et sp.n. and Cardiodictyon catenulum gen. et sp.n. and the arthropods Saperion glumaceum gen. et sp.n., Sinoburius Iunaris gen. et sp.n., and Xandarella spectaculum gen. et sp.n.  相似文献

In order to study the mechanisms behind the infection process of the necrotrophic fungus Botrytis cinerea, the subcellular distribution of hydrogen peroxide (H2O2) was monitored over a time frame of 96 h post inoculation (hpi) in Arabidopsis thaliana Col-0 leaves at the inoculation site (IS) and the area around the IS which was defined as area adjacent to the inoculation site (AIS). H2O2 accumulation was correlated with changes in the compartment-specific distribution of ascorbate and glutathione and chloroplast fine structure. This study revealed that the severe breakdown of the antioxidative system, indicated by a drop in ascorbate and glutathione contents at the IS at later stages of infection correlated with an accumulation of H2O2 in chloroplasts, mitochondria, cell walls, nuclei and the cytosol which resulted in the development of chlorosis and cell death, eventually visible as tissue necrosis. A steady increase of glutathione contents in most cell compartments within infected tissues (up to 600% in chloroplasts at 96 hpi) correlated with an accumulation of H2O2 in chloroplasts, mitochondria and cell walls at the AIS indicating that high glutathione levels could not prevent the accumulation of reactive oxygen species (ROS) which resulted in chlorosis. Summing up, this study reveals the intracellular sequence of events during Botrytis cinerea infection and shows that the breakdown of the antioxidative system correlated with the accumulation of H2O2 in the host cells. This resulted in the degeneration of the leaf indicated by severe changes in the number and ultrastructure of chloroplasts (e.g. decrease of chloroplast number, decrease of starch and thylakoid contents, increase of plastoglobuli size), chlorosis and necrosis of the leaves.  相似文献

Purified and crude flagellar isolates from cells of Bacillus pumilus NRS 236 were treated with acid, alcohol, acid-alcohol, or heat, and were examined electron microscopically in negatively stained and shadow-cast preparations. Under certain conditions, each of these agents causes the flagella to break between the proximal hooks and the spiral filaments. In such preparations, filaments are seen in various stages of disintegration, whereas hooks of fairly constant length retain their integrity and morphological identity. When crude isolates of flagella are treated under these conditions, the hooks remain attached to membrane fragments or bear basal material. These findings substantiate previous structural observations that led to the view that the proximal hook is a distinct part of the bacterial flagellum and further confirm that the hook is tightly associated with basal material and the cytoplasmic membrane. It appears that the hook is a polarly oriented structure, and that the interactions between the hook and the basal material or the cytoplasmic membrane are different from those between the hook and the filamentous portion of the organelle. Moreover, both types of interaction apparently differ still from those by which the flagellin subunits are held together in the flagellar filament. Hooks were isolated by exploiting the differences in relative stability shown by the various morphological regions of the bacterial flagellum.  相似文献
