The bcl-2 protein plays a role in the regulation of programmed cell death (PCD), overriding apoptosis. Its expression has been reported in breast ductal cells, where it is believed to be involved in the hormonal regulation of hyperplasia and involution. to date, bcl-2 gene product has not been investigated on breast cancer FNA. the expression of bcl-2 protein was evaluated using an immunoalkaline phosphatase technique in 54 pre-operative breast cancer aspirates and in paraffin-embedded sections from 20 matched surgical specimens. A high rate of bcl-2 protein expression was found on FNA samples (65%) and on the corresponding tissue sections (60%); there was a nearly absolute concordance in the two specimens, with 19/20 (95%) cases showing a concordant staining. These findings concur with the view that bcl-2 gene is frequently expressed in breast cancer, possibly through a hormonal-dependent pathway.  相似文献

Several axenic strains of pathogenic and nonpathogenic Entamoeba histolytica were tested for their capacity to digest native radioactive type I collagen gels and to produce liver abscesses when injected into the liver of newborn hamsters. The results demonstrate that the pathogenic strains of amebas (HM1:IMSS, HM3:IMSS, HM38:IMSS, and HK9) have a collagenolytic activity that closely correlates with their in vivo capacity to produce liver lesions. The nonpathogenic isolate (Laredo) did not show collagenolytic activity and failed to produce lesions in the liver of newborn hamsters. The results also demonstrate that type I collagen obtained from rodents and cats is degraded less by amebic collagenase than is bovine collagen, which is similar to human collagen. These findings suggest that species susceptibility to invasive infection may depend, among other factors, on the characteristics of the extracellular components of host tissues.  相似文献

A mathematical analysis for fast changes of ethylene concentration in an open flow system (non-steady-state conditions) is presented and experimentally tested. In this way it becomes possible to determine true values of ethylene production in the minute range following physiological and environmental changes which influence ethylene evolution. By this procedure ethylene kinetics can also be compared in absolute values independent of flow rate and plant chamber volume.  相似文献

Treatment of Cucumis leaf discs with light and low temperature (1°C) resulted in degradation of the total lipids. In addition, a decrease of the linolenic acid content of the glycolipids of leaves and leaf discs took place while at the same time an increase in the content of unidentified degradation products from the glycolipids was observed. The decrease of the linolenic acid content was not due to galactolipase activity, since no monogalactosyl diglyceride acyl hydrolase activity was found. Dark and low temperature did not alter the fatty acid composition. Blue light and low temperature resulted in a decrease of the linolenic acid content, while yellow-, red- and far red light in combination with low temperature were ineffective.  相似文献

X-ray crystallographic studies of gramicidin A crystallized from methanol (P21) and ethanol (P212121), and of a Cs+ gramicidin A complex crystallized from methanol (P2221, P21212 or P212121) are reported here. The asymmetric unit consists of two molecules of gramicidin A in the native crystals and four molecules in the cesium complex crystal. Patterson analyses show that gramicidin A in these crystals forms a cylindrical helical channel. In the two types of native gramicidin crystals, the diameter of this channel is about 5 å and its length is about 32 å. Cesium ions are bound inside this channel in crystals of the cesium-gramicidin A complex. The channel in this complex is considerably shorter (26 Å) and wider (6·8Å) than in the native forms. The Patterson maps of these three crystal forms are compatible with either the single-stranded β-helix (Urry, 1971) or the double-stranded parallel or anti-parallel, β-helix (Veatch et al., 1974).  相似文献

The synthesis and testing of several diazocarbonyl JH analogs (diazo JHA) which act as photoaffinity labels for insect juvenile hormone binding proteins are described. The best competitor, 10,11-epoxyfarnesyl diazoacetate, has been shown to irreversibly reduce [3H]-JH III binding to both ovarian and hemolymph JHBP from Leucophaeamaderae after irradiation at 254 nm for 20 seconds. No loss of activity was observed after incubation of JHBP and diazo JHA without irradiation. Protection from photoinactivation by diazo JHA II was achieved by the presence of an equimolar amount of JH III during the photolysis. Photoaffinity labeled proteins show loss of binding capacity without alteration of the binding affinity. This is the first example of the use of a photoaffinity label in the study of JH action on a molecular level, and may become a valuable tool in the elucidation of JH-receptor-chromatin interactions.  相似文献

Behavior can be regarded as a result of various processes ofdecision based on the information provided by the sensory organs.In this review the role of the so-called additional heat sense,next to vision, smell and mechanoreception is discussed withrespect to the feeding behavior of snakes. The hierarchy ofthe sensory information in various phases of the feeding behaviordiffers between snakes possessing heat receptors (e.g., speciesof the Crotalinae and Pythoninae) and those without (e.g., speciesof the Viperinae and Colubrinae). Probably depending on theinfluence of ecological demands, visual or chemical cues arethe main information in the behavioral phases before the strikeHowever, in situations with little visual input, e.g., in darkness,rodents' burrows, etc., hunting behavior is guided in the firstplace by radiation of warm objects in Crotalus, Python and Trimeresurusflavoviridis, and by substrate vibrations in Vipera aspis, Pituophismelanoleucus and Boa constrictor. I suggest that in the sensoryhierarchy, heat information functionally replaces the mechanicalinformation which is utilized by snakes without pit organs.Poststrike behavior on the other hand is mainly guided by chemicalcues in all snakes.  相似文献

Heating locally the hypocotyl of Bidens pilosa L. elicits awave of depolarization. The mechanism of the wave has been investigatedby means of microelectrophysiological techniques. The amplitudeof the transmembrane potential variation induced by an extracellularion concentration change (K+, Na+, Ca2+, Cl) was thesame in the resting conditions as during the slow wave. At pH4.0, the amplitude of the slow wave was reduced by 56% comparedwith the control performed at pH 7.0. In the presence of theuncoupler CCCP, the slow wave was not observed. The Ca2+ -chelatorEGTA and the Ca22+ -channel blocker La3+ reduced, respectively,the amplitude of the slow wave by 78% and 68%. These resultsindicate the involvement of Ca2+ in triggering the slow wave.A transient modification of the electrogenic H+ pump activity(inactivation-activation) and of the transmembrane H+ flux inthe slow wave are discussed.
