A lactobacilli dominated microbiota in most pre and post-menopausal women is an indicator of vaginal health. The objective of this double blinded, placebo-controlled crossover study was to evaluate in 14 post-menopausal women with an intermediate Nugent score, the effect of 3 days of vaginal administration of probiotic L. rhamnosus GR-1 and L. reuteri RC-14 (2.5×109 CFU each) on the microbiota and host response. The probiotic treatment did not result in an improved Nugent score when compared to when placebo. Analysis using 16S rRNA sequencing and metabolomics profiling revealed that the relative abundance of Lactobacillus was increased following probiotic administration as compared to placebo, which was weakly associated with an increase in lactate levels. A decrease in Atopobium was also observed. Analysis of host responses by microarray showed the probiotics had an immune-modulatory response including effects on pattern recognition receptors such as TLR2 while also affecting epithelial barrier function. This is the first study to use an interactomic approach for the study of vaginal probiotic administration in post-menopausal women. It shows that in some cases multifaceted approaches are required to detect the subtle molecular changes induced by the host to instillation of probiotic strains.
Alzheimer''s disease (AD) is the most frequently diagnosed form of dementia resulting in cognitive impairment. Many AD mouse studies, using the methyl donor S-adenosylmethionine (SAM), report improved cognitive ability, but conflicting results between and within studies currently exist. To address this, we conducted a meta-analysis to evaluate the effect of SAM on cognitive ability as measured by Y maze performance. As supporting evidence, we include further discussion of improvements in cognitive ability, by SAM, as measured by the Morris water maze (MWM).
Methods
We conducted a comprehensive literature review up to April 2014 based on searches querying MEDLINE, EMBASE, Web of Science, the Cochrane Library and Proquest Theses and Dissertation databases. We identified three studies containing a total of 12 experiments that met our inclusion criteria and one study for qualitative review. The data from these studies were used to evaluate the effect of SAM on cognitive performance according to two scenarios: 1. SAM supplemented folate deficient (SFD) diet compared to a folate deficient (FD) diet and 2. SFD diet compared to a nutrient complete (NC) diet. Hedge''s g was used to calculate effect sizes and mixed effects model meta-regression was used to evaluate moderating factors.
Results
Our findings showed that the SFD diet was associated with improvements in cognitive performance. SFD diet mice also had superior cognitive performance compared to mice on an NC diet. Further to this, meta-regression analyses indicated a significant positive effect of study quality score and treatment duration on the effect size estimate for both the FD vs SFD analysis and the SFD vs NC analysis.
Conclusion
The findings of this meta-analysis demonstrate efficacy of SAM in acting as a cognitive performance-enhancing agent. As a corollary, SAM may be useful in improving spatial memory in patients suffering from many dementia forms including AD. 相似文献
Life-history traits interact in important ways. Relatively few studies, however, have explored the relationships between life-history traits in long-lived taxa such as trees. We examined patterns of energy allocation to components of reproduction and growth in three species of California oaks (Quercus spp.) using a combination of annual acorn censuses, dendrometer bands to measure radial increment, and litterfall traps. Our results are generally consistent with the hypothesis that energy invested in reproduction detracts from the amount of energy available for growth in these long-lived taxa; i.e., there are trade-offs between these traits. The relationships between reproduction and growth varied substantially among specific trait combinations and tree species, however, and in some cases were in the direction opposite that expected based on the assumption of trade-offs between them. This latter finding appears to be a consequence of the pattern of resource use across years in these long-lived trees contrasting with the expected partitioning of resource use within years in short-lived taxa. Thus, the existence and magnitude of putative trade-offs varied depending on whether the time scale considered was within or across years. Collectively, our results indicate that negative relationships between fundamental life-history traits can be important at multiple levels of modular organization and that energy invested in reproduction can have measurable consequences in terms of the amount of energy available for future reproduction and both current and future growth. 相似文献
N-myristoylation is a crucial covalent modification of numerous eukaryotic and viral proteins that is catalyzed by N-myristoyltransferase (NMT). Prokaryotes are lacking endogeneous NMT activity. Recombinant production of N-myristoylated proteins in E. coli cells can be achieved by coexpression of heterologous NMT with the target protein. In the past, dual plasmid systems were used for this purpose.
Methodology/Principal Findings
Here we describe a single vector system for efficient coexpression of substrate and enzyme suitable for production of co- or posttranslationally modified proteins. The approach was validated using the HIV-1 Nef protein as an example. A simple and efficient protocol for production of highly pure and completely N-myristoylated Nef is presented. The yield is about 20 mg myristoylated Nef per liter growth medium.
Conclusions/Significance
The single vector strategy allows diverse modifications of target proteins recombinantly coexpressed in E. coli with heterologous enzymes. The method is generally applicable and provides large amounts of quantitatively processed target protein that are sufficient for comprehensive biophysical and structural studies. 相似文献
Conversion of wetlands in cultivated agricultural landscapes is one of the primary drivers of wetland loss in Alberta, Canada, despite a provincial wetland policy that prioritizes wetland avoidance. While other sectors of the agricultural industry have established initiatives to maintain wetlands, a common narrative within the conventional cropping sector is that wetland retention leads to lost acreage and overlap of crop inputs, and that there are financial benefits associated with wetland drainage. The objective of this research was to explicitly quantify crop productivity within drained wetland basins, in an effort to better understand the extent to which producers financially benefit from drainage practices. Working collaboratively with canola producers in central Alberta over the 2019 growing season, wetland basins within four quarter sections were mapped using an Unmanned Aerial Vehicle, and wetland basins with clear evidence of surface drainage were identified. Agricultural input and yield data provided by producers was then used to quantify profitability within each drained basin. Average profit for drained basins for each producer ranged between ? $145/acre and $76/acre, with an average of $55/acre across all operations. This is compared to an average profit of $203/acre for non-wetland areas across all operations. The results suggest that the financial benefits of drainage are highly variable, and for many drained basins, producers may experience financial losses that may be overlooked when profits are examined only at the field- or operation-level. While this study included a small number of operations, and was limited to one type of crop over a single growing season, the results still provide important insight into the extent to which producers benefit financially from the practice of wetland drainage.
Purpose: Elevated carbohydrate antigen 125 (CA125) predicts adverse outcome after transcatheter aortic valve implantation (TAVI). While known underlying pathophysiological mechanisms of elevated CA125 include serosal effusions and inflammatory stimuli, clinical determinants associated with elevated CA125 in patients referred for TAVI remain unknown. Therefore, we investigated clinical, laboratory and echocardiographic determinants of elevated CA125 in patients with severe aortic valve stenosis referred for TAVI.Methods: This study includes 650 patients with severe aortic stenosis referred for TAVI. Baseline CA125 was determined by an immunoassay and dichotomized (elevated versus normal) based on the manufacturer cutoff value (>35?U/mL).Results: CA125 elevation was present in 28% (181/650). Patients with elevated CA125 had an overall worse clinical profile and were more symptomatic with a higher rate of NYHA class III/IV (80% versus 58%; p?0.001). In a multivariate analysis, independent predictors of elevated CA125 were New York Heart Association (NYHA) class, baseline hemoglobin, C-reactive protein (CRP), left ventricular ejection fraction and severe tricuspid regurgitation.Conclusion: Elevated CA125 levels in patients referred for TAVI summarize a subset of patients with an overall worse clinical profile who are more symptomatic. 相似文献
Cooperative breeding is generally associated with increased philopatry and sedentariness, presumably because short-distance dispersal facilitates the maintenance of kin groups. There are, however, few data on long-distance dispersal in cooperative breeders—the variable likely to be important for genetic diversification and speciation. We tested the hypothesis that cooperative breeders are less likely to engage in long-distance dispersal events by comparing records of vagrants outside their normal geographic range for matched pairs (cooperatively vs. non-cooperatively breeding) of North American species of birds. Results failed to support the hypothesis of reduced long-distance dispersal among cooperative breeders. Thus, our results counter the conclusion that the lower rate of speciation among cooperative breeding taxa found in recent analyses is a consequence of reduced vagility. 相似文献
The Raman spectra of ovalbumin, ovomucoid, and conalbumin are reported. Spectral shifts in the conformationally sensitive amide I and amide III lines as a result of thermal denaturation indicate the formation of intermolecular β- sheets. A medium intensity line at 1260 cm?1 in the spectra of ovomucoid and ribonuclease is demonstrated to contain a substantial contribution from tyrosine residues. 相似文献
