We examined the dynamics of cryptogamic soil crusts in a derived (disclimax) grassland near Orange in south‐eastern Australia. Changes in the cover of cryptogamic crusts and floristics and abundance of the constituent species were measured on four treatments with two levels each of grazing and cultivation. Twenty‐two lichens, mosses and liverworts were found at the study site and, of these, 13 were collected in the quadrats. Three moss species (Barbula calycina, Eccremidium arcuatum and Bryum pachytheca) and one lichen species (Cladonia tessalata) accounted for 67% of total cover‐abundance scores. Generally, cover‐abundance was significantly higher in the unvegetated microsites than in the vegetated microsites. Species richness was not significantly different between the four grazing‐cultivation treatments but, on average, there were significantly more species in the unvegetated microsites (mean = 3.2 species) than in the vegetated microsites (0.54 species). Grazing and cultivation resulted in significantly greater cover of bare ground and consequently significantly greater crust cover. Averaged across all treatments, approximately half of the area of unvegetated soil was occupied by cryptogams. Overall, the results indicate that lichens and bryophytes are important components of derived temperate grasslands, surviving in even densely vegetated swards. This study suggests that strategies which disturb the soil surface (e.g. grazing and cultivation) will stimulate the abundance and cover of soil crust organisms by increasing the availability of unvegetated microsites. 相似文献

Diseases caused by parasitic protozoa remain a major health problem, mainly due to old toxic drugs and rising drug resistance. Nucleoside hydrolases are key enzymes of the purine salvage pathway of parasites from the Trypanosomatidae family and are considered as possible drug targets. N-Arylmethyl substituted iminoribitols have been developed as selective nanomolar affinity inhibitors against the purine-specific nucleoside hydrolase of Trypanosoma vivax. The current paper describes the crystal structures of the T. vivax nucleoside hydrolase in complex with two of these inhibitors, to 1.3 and 1.85 Å resolution. These high resolution structures provide an accurate picture of the mode of binding of these inhibitors and their mechanism of transition-state mimicry, and are valuable tools to guide further inhibitor design. Comparison of the current structures with previously solved structures of the enzyme in complex with ground-state and transition-state-analogue inhibitors also allows for the elucidation of a detailed molecular mechanism of active-site loop opening/closing. These loop movements can be coupled to the complex kinetic mechanism of the T. vivax nucleoside hydrolase. 相似文献

A total of 67 taxa, belonging to 15 genera, were recorded from aquatic samples collected in Zackenberg (Northeast Greenland).
Most taxa belong to the genera Difflugia, Centropyxis, Nebela and Euglypha. Trinema lineare showed the highest relative abundance. Seven taxa were recorded for the first time in the Arctic. A TWINSPAN classification
reveals two groups, reflecting a local geographical distinction. In the ponds of the old Zackenberg delta area (group 1),
two assemblages were found: the Paraquadrula globulosa-Cyphoderia perlucidus assemblage and the Difflugiella crenulata-Cryptodifflugia compressa assemblage. The lakes, the pools and ponds east of the Zackenberg river (group 2) also harbour two assemblages: the Trinema enchelys-Euglypha sp.1 assemblage and the Centropyxis aerophila-Difflugia globulosa assemblage. Although the lakes are chemically unique, having extremely low values for turbidity, colour, SiO2−
2, Cl− and hardness, this is not reflected in the characteristic testacean communities.

Some inter- and intraspecific crosses may result in reduced viability or sterility in the offspring, often due to genetic incompatibilities resulting from interactions between two or more loci. Hybrid necrosis is a postzygotic genetic incompatibility that is phenotypically manifested as necrotic lesions on the plant. We observed hybrid necrosis in interspecific lettuce (Lactuca sativa and Lactuca saligna) hybrids that correlated with resistance to downy mildew. Segregation analysis revealed a specific allelic combination at two interacting loci to be responsible. The allelic interaction had two consequences: (1) a quantitative temperature-dependent autoimmunity reaction leading to necrotic lesions, lethality, and quantitative resistance to an otherwise virulent race of Bremia lactucae; and (2) a qualitative temperature-independent race-specific resistance to an avirulent race of B. lactucae. We demonstrated by transient expression and silencing experiments that one of the two interacting genes was Rin4. In Arabidopsis thaliana, RIN4 is known to interact with multiple R gene products, and their interactions result in hypersensitive resistance to Pseudomonas syringae. Site-directed mutation studies on the necrosis-eliciting allele of Rin4 in lettuce showed that three residues were critical for hybrid necrosis. 相似文献

Background: The Mongolian gerbil model is often used to investigate the interactions between different gastric Helicobacter species and the gastric tissue. A preliminary screening of a gerbil population intended for use in Helicobacter suis infection studies revealed a natural yeast infection in the stomach of these animals. After identification, we have investigated the effect of the gastric yeast infection on the outcome of an experimental H. suis infection in Mongolian gerbils.
Materials and methods: Yeast cells were isolated from the stomachs of Mongolian gerbils. Identification was done by Internally Transcribed rRNA Spacer 2 Region PCR fragment length analysis. To investigate a possible pathologic role of this yeast, Mongolian gerbils were infected experimentally with this yeast. Co-infection with the newly isolated H. suis was performed to investigate possible interactions between both micro-organisms.
Results: Kazachstania heterogenica was found colonizing the stomach of Mongolian gerbils, mainly in the antrum. Few pathologic changes were seen in the stomachs of infected animals. Experimental co-infection of gerbils with this yeast and the newly isolated H. suis showed a significant increase in inflammation in animals infected with both micro-organisms compared to animals infected only with H. suis .
Conclusions: K. heterogenica colonizes the stomach of Mongolian gerbils in exactly the same regions as gastric Helicobacter species. The uncontrolled presence of this yeast in the gerbil stomach can lead to an overestimation of the inflammation caused by Helicobacter in this animal model. 相似文献

Effects of heparin, spermidine, and Be2+ ions on the ATPase and beta-glycerophosphatase and RNA-ase activities of the rat liver cell nuclei were studied. Be2+ was shown to inhibit the ATPase activity and, to a lesser extent, beta-glycerophosphatase activities. Physiological concentrations of heparin and spermidine also lowered the mentioned two activities, as well as the RNAase activity of the nuclei. Evidence is presented for the inhibitory effect of heparin and spermidine on endonucleases. 相似文献
