The effect of increasing arterial partial pressure of CO2 (PaCO2) on respiratory mechanics was investigated in six anesthetized, paralyzed cats ventilated by constant-flow inflation. Respiratory mechanics were studied after end-inspiratory occlusions. Zero frequency resistance (Rmax), infinite frequency resistance (Rmin), and static elastance (Est) were calculated for the respiratory system, lung, and chest wall. Alveolar ventilation was manipulated by the addition of dead space to achieve a range of PaCO2 values of 29.3-87.3 mmHg. Cats did not become hypoxic during the experiment. Under control conditions marked frequency dependence in Rmax, Rmin, and Est of the respiratory system, lungs, and chest wall was demonstrated. The chest wall contributed 50% of the total resistance of the respiratory system. With increasing PaCO2 the only resistance observed to increase was Rmax of the lung (P less than 0.01). There were also no changes in the static elastic properties of either the lungs or the chest wall. These results suggest that hypercapnia increases resistance by changes in the lung periphery and not in the conducting airways.  相似文献

A new form of gamma-glutamyltransferase was purified from human seminal plasma. The purified enzyme was composed of two non-identical subunits with apparent molecular masses of 150 and 95 kDa on polyacrylamide gel electrophoresis (PAGE) in the presence of sodium dodecyl sulfate (SDS), and showed a molecular mass of 500 and 250 kDa on gel filtration in the absence and presence of 1% Triton X-100, respectively. This enzyme was different from human renal gamma-glutamyltransferase not only in apparent molecular masses, but also in amino acid compositions of both the subunits to each other. Experiments with the antisera raised against the purified enzyme revealed that the enzyme was different from the renal, hepatic and testicular enzymes in reactivity to the antibody though partially related to those enzymes. Ouchterlony double diffusion analysis indicated that both human seminal plasma and prostatic extract contained two types of gamma-glutamyltransferase, one is that we purified and the other the renal type. Hence, it is most likely that gamma-glutamyltransferase accounting for most of the enzyme activity in semen results from prostata followed by secretion to seminal plasma.  相似文献

The N-terminal sequences of the E1 α, E1β and E2 subunits of the human branched-chain α-keto acid dehydrogenase complex have been determined by microsequencing. The N-termini of human E1β and E2 subunits (Val and Gly, respectively) are indentical to those of the corresponding rat and bovine subunits. However, the N-terminus of the human E1 α subunit (Ser) is identical to bovine, but differs from the rat E1 α (Phe0 subunit. Comparison of the N-terminal sequences of human and rat E1 α subunits shows that the serine residue at the + 1 position in the human sequence is replaced by a proline residue in the rat sequence. The presence of the proline residue apparently causes a 5′-shift by one residue in the cleavage site by the mitochondrial processing peptidase in the rat sequence, when compared to the human sequence. The results provide evidence that the mitochondrial processing peptidase cannot cleave an X-pro bond, similar to trypsin, chymotrypsinand microsomal signal peptidases.  相似文献

Flow and volume dependence of pulmonary mechanics in anesthetized cats   总被引:2,自引:0,他引:2  
The effects of inspiratory flow rate and inflation volume on pulmonary mechanics were investigated in six anesthetized-paralyzed cats ventilated by constant-flow inflation. Pulmonary mechanics were assessed using the technique of rapid airway occlusion during constant-flow inflation which allows measurement of the intrinsic pulmonary resistance (RLmin) and of the overall "pulmonary flow resistance" (RLmax), which includes the additional pulmonary pressure losses due to time constant inequalities within the lung and/or stress adaptation. We observed that, at fixed inflation volume, 1) RLmin fitted Rohrer's equation, 2) RLmax was higher at low than intermediate flows, and 3) RLmax-RLmin decreased progressively with increasing flow. At fixed flow, RLmax increased, whereas RLmin decreased with increasing volume. We conclude that during eupneic breathing in cats, the pulmonary flow resistance as conventionally measured includes a significant component reflecting stress adaptation.  相似文献

The spontaneously diabetic torii (SDT) fatty rat is a new model of type 2 diabetes showing overt obesity, hyperglycemia and hyperlipidemia. With early onset of diabetes mellitus, diabetic microvascular complications, including nephropathy, peripheral neuropathy and retinopathy, are observed at young ages. In the present study, blood glucose levels of female SDT fatty rats were controlled with phlorizin, a non-selective SGLT inhibitor, to examine whether and how these complications are caused by hyperglycemia. Phlorizin treatment adequately controlled plasma glucose levels during the experiment. At 29 weeks of age, urinary albumin excretion considerably increased in SDT fatty rats. Glomerulosclerosis and tubular pathological findings also indicate diabetic nephropathy. These renal parameters tended to decrease with phlorizin; however, effects were partial. Sciatic nerve conduction velocities were significantly delayed in SDT fatty rats compared with Sprague-Dawley (SD) rats. Intraepidermal nerve fiber density, an indicator of subclinical small nerve fiber neuropathy, significantly decreased in SDT fatty rats. Retinal dysfunction (prolongation of peak latency for oscillatory potential in electroretinograms) and histopathological eye abnormalities, including retinal folding and mature cataracts were also observed. Both nerve and eye disorders were prevented with phlorizin. These findings indicate that severe hyperglycemia mainly causes diabetic complications in SDT fatty rats. However, other factors, such as hyperlipidemia and hypertension, may affect diabetic nephropathy. These characteristics of diabetic complications will become helpful in evaluating new drugs for diabetic complications using SDT fatty rats.  相似文献

This study quantified stomatal conductance in a CO2-fertilized warm-temperate forest. The study considered five items: (1) the characteristics of the diurnal and seasonal variation, (2) simultaneous measurements of canopy-scale fluxes of heat and CO2 and the normalized difference vegetation index (NDVI), (3) the stomatal conductance of sunlit and shaded leaves, (4) a stomatal conductance model, and (5) the effects of leaf age on stomatal conductance. Sampled plants included evergreen and deciduous species. Stomatal conductance, SPAD, and leaf nitrogen content were measured between March and December 2001. Sunlit leaves had the largest diurnal and seasonal variation in conductance in terms of both magnitude and variability. In contrast, shaded leaves had only low conductance and slight variation. Stomatal conductance increased sharply in new shooting leaves of Quercus serrata until reaching a maximum 2 months after full leaf expansion. The seasonal changes in the canopy-scale heat and CO2 fluxes were similar to the change in the canopy-scale NDVI of the upper-canopy plants. These seasonal changes were correlated with the leaf-level H2O/CO2 exchanges of upper-canopy plants, although these did not represent the stomatal conductance in fall completely. Seasonal variations in the leaf nitrogen content and SPAD were similar, except leaf foliation, until day 130 of the year, when the behaviors were completely the opposite. A Jarvis-type model was used to estimate the stomatal conductance. We modified it to include SPAD as a measure of leaf age. The seasonal variation in stomatal conductance was not as sensitive to SPAD, although estimates for evergreen species showed improvements.  相似文献

We recently proposed an eight-parameter model of the respiratory system to account for its mechanical behavior when flow is interrupted during passive expiration. The model consists of two four-parameter submodels representing the lungs and the chest wall, respectively. The lung submodel consists of an airways resistance together with elements embodying the viscoelastic properties of the lung tissues. The chest wall submodel has similar structure. We estimated the parameters of the model from data obtained in four normal, anesthetized, paralyzed, tracheostomized mongrel dogs. This model explains why lung tissue and chest wall resistances should be markedly frequency dependent at low frequencies and also permits a physiological interpretation of resistance measurements provided by the flow interruption method.  相似文献
