Gel filtration of human serum shows that there are two separate peaks of activity that will cause fibroblasts in culture to adopt their characteristic bipolar morphology (i.e., cell spreading). One of these activities has a molecular weight just below 250 000 D while the other is in the range 65 000–85 000 D. The lower molecular weight activity is unlikely to be related to cold-insoluble globulin, yet is quantitatively the more significant.  相似文献

Concentrations of estradiol-17β (E2), progesterone (Po), and luteinizing hormone (LH) in serum and estrone glucuronide (E1G) and pregnanediol glucuronide (PdG) in urine were determined by direct radioimmunoassay in five yearling and three adult female white-tailed deer (Odocoileus virginianus) from 14 October to 20 December 1985. Elevated levels of LH were recorded before the first estrus of the breeding season and immediately preceding or during behavioral estrus. Urinary E1G and PdG concentrations were poorly correlated with serum levels of E2 and Po, respectively. Serum Po levels prior to the first estrus of the breeding season indicate that pubertal yearlings experience silent ovulations and suggest that silent ovulations may be important in the transition from adolescence to cycling adult in white-tailed deer. © 1992 Wiley-Liss, Inc.  相似文献

One hundred and twenty-six cases of pityriasis rosea seen over two years in north Staffordshire were analysed for clustering in time and space. A statistically significant degree of clustering was found; this was evident among female patients considered separately but not among male patients. The incidence of the condition was higher in patients working in, or attending, educational establishments. These findings support the hypothesis that pityriasis rosea is caused by an infective agent. A search for an infective organism and a transmission mechanism now seems justifiable.  相似文献

The Rose tissue culture chamber has been divided into 2 compartments by using half-thickness gaskets with an interposed Nuclepore (General Electric Co.) filter. This permits study of the interaction of 2 different cell populations on each side of the membrane. Radioiodinated serum albumin concentration studies demonstrate rapid distribution of labelled protein across the membrane while its pore size of 0.5 μ or 0.8 μ prevents migration of the cells themselves. Equilibrium between plain medium and radioactive medium was established in 7-9 hr.  相似文献

Rhizosheaths function in plant?soil interactions, and are proposed to form due to a mix of soil particle entanglement in root hairs and the action of adhesive root exudates. The soil‐binding factors released into rhizospheres to form rhizosheaths have not been characterised. Analysis of the high‐molecular‐weight (HMW) root exudates of both wheat and maize plants indicate the presence of complex, highly branched polysaccharide components with a wide range of galactosyl, glucosyl and mannosyl linkages that do not directly reflect cereal root cell wall polysaccharide structures. Periodate oxidation indicates that it is the carbohydrate components of the HMW exudates that have soil‐binding properties. The root exudates contain xyloglucan (LM25), heteroxylan (LM11/LM27) and arabinogalactan‐protein (LM2) epitopes, and sandwich‐ELISA evidence indicates that, in wheat particularly, these can be interlinked in multi‐polysaccharide complexes. Using wheat as a model, exudate‐binding monoclonal antibodies have enabled the tracking of polysaccharide release along root axes of young seedlings, and their presence at root hair surfaces and in rhizosheaths. The observations indicate that specific root exudate polysaccharides, distinct from cell wall polysaccharides, are adhesive factors secreted by root axes, and that they contribute to the formation and stabilisation of cereal rhizosheaths.  相似文献

Pollen allergens have been shown to occur in fine particles much smaller than pollen grains. As such allergenic particles have a high probability of entering the lower human airways, it is important to further investigate their origin and their biological and immunological properties. Accordingly, we have modified the particle collection surface of an Andersen Impactor using adhesive coated microscope slides to enable identification by high resolution microscopic analysis and immunodetection of allergenic particulates.  相似文献

In the corticotroph-like murine pituitary tumor cell line, AtT-20, adrenocorticotropic hormone release is triggered by corticotropin-releasing hormone and is attenuated by the synthetic adrenal steroid dexamethasone. The precise mechanisms by which dexamethasone inhibits secretion are under investigation. We examined whether dexamethasone can modulate release via regulation of calcium homeostasis. More specifically, we have evaluated the effects of dexamethasone on calcium current, intracellular calcium concentration, and adrenocorticotropic hormone release. Using perforated patch-clamp and calcium imaging with fura PE3/AM, we found that dexamethasone decreases calcium current and intracellular calcium levels. The inhibition of current by dexamethasone is not, however, altered by the calcium channel antagonists nifedipine (L-type) or omega-agatoxin IVA (P/Q-type), despite the presence of these calcium channel subtypes in AtT-20 cells and the exclusive coupling of adrenocorticotropic hormone release to the L-type channel in these cells. We also evaluated the temporal relationship between dexamethasone-mediated inhibition of secretion and calcium influx. Whereas a prolonged (2 h) incubation with dexamethasone inhibits corticotropin-induced release by approximately 40%, a rapid (10 min) incubation (a time interval sufficient for dexamethasone-mediated inhibition of calcium transients) does not inhibit release. These data suggest, therefore, that dexamethasone does, indeed, modulate calcium homeostasis in AtT-20 cells, but that this effect is not responsible for its inhibition of secretion.  相似文献

An examination of the distribution of superoxide dismutase (SOD) activities in whole worm homogenates from six gastro-intestinal nematodes, at different life-cycle stages, demonstrated that the third larval stages of each species contained notably more activity than the corresponding adults. Significant SOD activities were detected in culture fluids following in vitro maintenance of third and fourth larval stage and adult Trichostrongylus vitrinus and Teladorsagia circumcincta (formerly Ostertagia circumcincta). Marked interspecies variation was observed when SOD isoenzyme profiles of adult and third larval stage Nippostrongylus brasiliensis, Trichostrongylus vitrinus, Teladorsagia circumcincta and Haemonchus contortus were compared. In addition, SOD activity and isoenzyme polymorphism were greater in Nippostrongylus brasiliensis harvested from primed rats compared to parasites recovered from naive rats.  相似文献
